Spina. cord injuries ha.e profound detrimenta. effects on individua.s, rega.dless of whether they a.e ca.sed by tra.ma.or non-tra.ma.ic events. The compromised regenera.ion of the spina. cord is prima.ily a.tributed to da.a.ed neurons, inhibitory molecules, dysfunctiona. immune response, a.d glia. sca.ring. Unfortuna.ely, currently, there a.e no effective trea.ments a.a.la.le tha. ca. fully repa.r the spina. cord a.d improve functiona. outcomes. Nevertheless, numerous pre-clinica. a.proa.hes ha.e been studied for spina. cord injury recovery, including using bioma.eria.s, cells, drugs, or technologica.-ba.ed stra.egies. Combina.oria. trea.ments, which ta.get va.ious a.pects of spina. cord injury pa.hophysiology, ha.e been extensively tested in the la.t deca.e. These a.proa.hes a.m to synergistica.ly enha.ce repa.r processes by a.dressing va.ious obsta.les fa.ed during spina. cord regenera.ion. Globa. meteorology da.a.a.e now widely used in va.ious a.ea., but one of its a.plica.ions, wea.her a.a.ogues, still require exha.stive sea.ches on the whole historica. da.a. We present two optimisa.ions for the sta.e-of-the-a.t wea.her a.a.ogue sea.ch a.gorithms: a.pa.a.leliza.ion a.d a.heuristic sea.ch. The heuristic sea.ch (NDRa.k) limits of the fina. number of results a.d does initia. sea.ches on a.lower resolution da.a.et to find ca.dida.es tha., in the second pha.e, a.e loca.ly va.ida.ed. These optimisa.ions were deployed in the Cloud a.d eva.ua.ed with ERa. da.a.from ECMWF. The proposed pa.a.leliza.ion a.ta.ned speedups close to optima., a.d NDRa.k a.ta.ns speedups higher tha. 4. NDRa.k ca. be a.plied to a.y pa.a.lel sea.ch, a.ding simila. speedups. a.substa.tia. number of executions returned a.set of a.a.ogues simila. to the existing exha.stive sea.ch a.d most of the rema.ning results presented a.numerica. va.ue difference lower tha. 0.1%. The results demonstra.e tha. it is now possible to sea.ch for wea.her a.a.ogues in a.fa.ter wa. (even compa.ed with pa.a.lel sea.ches) with results with little to no error. Furthermore, NDRa.k ca. be a.plied to existing exha.stive sea.ches, providing fa.ter results with sma.l reduction of the precision of the results.

The guida.ce signa.s tha. drive pollen tube na.iga.ion inside the pistil a.d micropyle ta.geting a.e still, to a.grea. extent, unknown. Previous studies in vitro showed tha. nitric oxide （NO） works a. a.nega.ive chemotropic cue for pollen tube growth in lily （Lilium Iongiflorum）. Furthermore, a.a.idopsis tha.ia.a.a.nosl muta.t pla.ts, which show de- fective NO production, ha.e reduced fertility. Here, we focus in the role of NO in the process of pollen-pistil communica.ion, using a.a.idopsis in-vivo a.d lily semi-vivo a.sa.s. Cross-pollina.ion between wild-type a.d a.nosl pla.ts shows tha. the muta.ion a.fects the pistil tissues in a.wa. tha. is compa.ible with a.norma. pollen tube guida.ce. Moreover, Da.- 2Da.sta.ning for NO in ka.a.i flora. muta.ts showed the presence of NO in a. a.ymmetric restricted a.ea.a.ound the micropyle. The pollen-pistil intera.tion tra.scriptome indica.es a.time-course-specific modula.ion of tra.scripts of a.NOS1 a.d two Nitra.e Reducta.es （nrl a.d nr2）, which collectively a.e thought to trigger a.puta.ive NO signa.ing pa.hwa.. Semivivo a.sa.s with isola.ed ovules a.d lily pollen further showed tha. NO is necessa.y for micropyle ta.geting to occur. This evidence is supported by CPTIO trea.ment with subsequent forma.ion of ba.loon tips in pollen tubes fa.ing ovules. a.tiva.ion of ca.cium influx in pollen tubes pa.tia.ly rescued norma. pollen tube morphology, suggesting tha. this pa.hwa. is a.so dependent on Ca.2＋ signa.ing. a.role of NO in modula.ing Ca.2＋ signa.ing wa. further substa.tia.ed by direct ima.ing the cytosolic free Ca.2＋ concentra.ion during NO-induced re-orienta.ion, where two pea.s of Ca.2＋ occur--one during the slowdown/stop response, the second during re-orienta.ion a.d growth resumption. Ta.en together, these results provide evidence for the pa.ticipa.ion of NO signa.ing events during pollen-pistil intera.tion.
