Differential expression of cell cycle regulators in HCV-infection and related hepatocellular carcinoma

AIM:To investigate cell cycle proteins in chronic hepatitis C virus infection in order to analyze their role in the process of hepatocyte transformation and to characterize their prognostic properties. MeTHODS:Subjects of the current study included 50 cases of chronic hepatitis C(CHC) without cirrhosis,30 cases of CHC with liver cirrhosis(LC) ,and 30 cases of hepatitis C-related hepatocellular carcinoma(HCC) admitted to the Department of Hepato-Gastroenterology,Theodor Bilharz Research Institute(TBRI) ,Giza,*** wedge liver biopsies,taken during laparoscopic cholecystectomy,were also included as normal *** investigations including urine and stool analysis,liver function tests and prothrombin concentration;serologic markers for viral hepatitis and ultrasonography were done for all cases of the study together with immunohistochemical analysis using primary antibodies against Cyclin D1,Cyclin e,p21,p27 and Rb/p105 proteins. ReSULTS:Normal wedge liver biopsies didn't express Cyclin e or Rb/p105 immunostaining but show positive staining for Cyclin D1,p21 and *** D1 expressed nuclear staining that was sequentially increased from CHC to LC(Pes;meanwhile,Cyclin e revealed nuclear positivity only in the case of HCCs patients that was directly correlated to Rb/p105 *** expression of p21 and p27 was significantly increased in CHC and LC cases compared to normal controls and HCCs with no significant difference between well-and poorlydifferentiated tumors.p21 showed only a nuclear pattern of staining,while,p27 presented with either cytoplasmic and/or nuclear reactivity in all studied *** analysis revealed a direct relation between Cyclin D1 and p21 in CHC cases(Petween Cyclin D1 and Cyclin e in HCCs(Pever,an inverserelationship was detected between Cyclin D1 and p21 or p27(Petween p21 and Rb/p105(Pegulation of Cyclin D1 in CHC p

Friction and wear performance of polyether ether ketone(PeeK)polymers in three lubrication regimes

This experimental study investigates the friction and wear of three coatings commonly used in industrial applications,particularly in hydrodynamic *** three materials under investigation were Babbitt,polyether ether ketone(PeeK)reinforced with 15%carbon fibers,and PeeK reinforced with 20%carbon *** first polymer material was extruded,while the other was produced by fused deposition modelling(FDM).The materials were subjected to sliding tests in a pin-on-disc configuration,with a steel ball serving as the counter *** tests were conducted at room temperature,with a load of 10 N and under three different lubrication conditions:dry,grease,and *** linear speed was set at 0.3 m/s for the dry and semi-solid lubrication tests,while for the oil tests,the speed was set at 0.25 m/*** greases used had consistency grades of NGLI 000 and NGLI *** ISO VG 68 circulation oil was used for the oil lubrication ***,thermodynamic analyses were performed under the most severe conditions(i.e.,dry)to investigate the steel-Babbitt and steel-PeeK contact.

At this current time,data stream classification plays a key role in big data analytics due to its enormous *** of the existing classification methods used ensemble learning,which is trustworthy but these methods are not effective to face the issues of learning from imbalanced big data,it also supposes that all data are *** weakness of current methods is that it takes a long evaluation time when the target data stream contains a high number of *** main objective of this research is to develop a new method for incremental learning based on the proposed ant lion fuzzy-generative adversarial network *** proposed model is implemented in spark *** each data stream,the class output is computed at slave nodes by training a generative adversarial network with the back propagation error based on fuzzy bound *** method overcomes the limitations of existing methods as it can classify data streams that are slightly or completely unlabeled data and providing high scalability and *** results show that the proposed model outperforms stateof-the-art performance in terms of accuracy(0.861)precision(0.9328)and minimal MSe(0.0416).

Using immune cell-based bioactivity assays to compare the inflammatory activities of oil sands process-affected waters from a pilot scale demonstration pit lake

In this study,we provide evidence that oil sands process-affected waters(OSPW)contain factors that activate the antimicrobial and proinflammatory responses of immune ***,using the murine macrophage RAW 264.7 cell line,we establish the bioactivity of two different OSPW samples and their isolated ***,we directly compared the bioactivity of two pilot scale demonstration pit lake(DPL)water samples,which included expressed water from treated tailings(termed the before water capping sample;BWC)as well as an after water capping(AWC)sample consisting of a mixture of expressed water,precipitation,upland runoff,coagulated OSPW and added *** inflammatory(*** activating)bioactivity was associated with the AWC sample and its organic fraction(OF),whereas the BWC sample had reduced bioactivity that was primarily associated with its inorganic fraction(IF).Overall,these results indicate that at non-toxic exposure doses,the RAW 264.7 cell line serves as an acute,sensitive and reliable biosensor for the screening of inflammatory constituents within and among discrete OSPW samples.

AIM： To analyze the epidemiological, clinical and laboratory findings of infectious ***： A retrospective study on cases of infective keratitis, attended our institution from Mar. 2013 to Feb. 2015, was done at Mansoura Ophthalmic Center, egypt. Corneal scrapings were performed and processed for direct microscopy and culture in appropriate media using standard laboratory ***： Out of 245 patients enrolled for study, 247 corneal scrapings were obtained. Ocular trauma was the most common predisposing factor （51.4%）, followed by diabetes mellitus （15.1%）. Cultures were positive in 110 scraping samples （44.5%）： 45.5% samples had pure fungal infection, 40% had pure bacterial infections and 10% had mixed fungal and bacterial growths. Acanthamoeba was detected in 5 （4.5%） samples. The most common fungal pathogen was Aspergillus spp. （41%）. The most common bacterial isolates were Staphylococcus aureus （38.2%） and Pseudomonas aeruginosa （21.8%）.CONCLUSION： Incidence of fungal keratitis is high in our region. Therapeutic approach can initially be based on clinical features and sensitivity/resistance patterns. Microbiological research should direct the antimicrobial treatment. Antibiotic resistance to fluoroquinolones and aminoglycosides is an important consideration.

Toxoplasma gondii infection among chronic hepatitis C patients:A casecontrol study

Objective:To determine the detection rate of anti-Toxoplasma gondii(***) IgG and IgM in chronic HCV patients attending the Department of Tropical Medicine Mansoura University hospital in ***:This study included 120 adult chronic HCV patients.81 decompensate cirrhosis(late-stage)and 39 chronic HCV non cirrhotic patients(early-stage) and40 healthy blood donors as *** samples uere examined for anti-Toxoplasma IgM and anti-Toxoplasma IgG antibodies by ***-time RT-polymerase chain reaction assay was done for quantitation of hepatitis C ***:*** IgG antibodies were detected in 75(92.6%) of 81 late-stage cirrhotic patients.30(76.9%) of the 39 chronic HCV non cirrhotic patients(early-Stage) and in 6(15ft) of 40 controls with statistically significant difference(Pes were found in 11(13.6%) in late stage patients,5(12.8%)in early stage and in 3(7.5%) of controls with no statistical significant difference(P=0.610).There was no correlation between stage of fibrosis and IgM or IgG antibodies positivity in our studied groups(P=0.526).High IgG levels significantly correlated with high viral load(P=0.026).Conclusions:Our findings suggest that the serious opportunistic *** infection represent a potential significant risk for chronic HCV *** should be considered in their investigations and follow-up.

Particulate matter emissions(PM10)from the combustion,in a residential stove,of two com-mercial brands of certified(eNplus A1)pellets,a non-certified brand and laboratory made pellets of acacia were tested for their ability to induce ecotoxic,cytotoxic,and mutagenic re-sponses in unicellular organisms and a human cell *** was evaluated through the Vibrio fischeri bioluminescence inhibition ***,cytotoxicity was assessed at two time points(24-and 48-hr)through two complementary techniques in order to evaluate the cellular metabolic activity and membrane integrity of human lung epithelial cells *** Ames test using two Salmonella typhimurium strains(TA100 and TA98)was employed to assess the mutagenic potential of the polycyclic aromatic hydrocarbon fraction extracted from the PM10 *** obtained with the bioluminescent bacteria indicated that only particles from the combustion of acacia pellets were *** samples induced im-pairment on the A549 cells metabolic activity,while no significant release of lactate dehy-drogenase was ***10 emissions from acacia pellets were the most cytotoxic,while samples from both certified pellets evoked significant cytotoxicity at lower ***-icity time-dependency was only observed for PM10 from the combustion of acacia pellets and one of the brands of certified *** activity was not detected in both *** murium *** study emphasises the role of the raw material for pellet manu-facturing on the toxicological profile of PM *** raw materials should be deeply investigated before their use in pelletisation and combustion in residential appli-ances.

Remediation effect of compost on soluble mercury transfer in a crop of Phaseolus vulgaris

We studied the dynamics of mercury（Hg） transfer in Phaseolus vulgaris plants grown in soil with Hg-doped compost at the maximum levels permitted by Colombian law on organic amendments. Quantitative evaluation of transfer was made in different plant organs： roots,stem, leaves, pods and seeds. Matrix effect was determined in doped soil assays, using soil with and without addition of compost. Results showed that the use of organic matter reduced Hg transfer to the plant and the amount transferred was differentially distributed to the organs. We observed an inverse relationship between concentration and distance from the body to the root. It was evident that transfer was mediated by quantitative factors;the greater the presence of mercury in soil, the larger the amount that will be *** also indicate the remedial effect of compost and the presence of a barrier, at the root level, against mercury translocation to the plant aerial parts.

Dietary analysis of Homonota darwini (Squamata:Gekkonidae) in Northern Patagonia

Our study investigated the diet of the southernmost gecko in the world, Homonota darwini. Fifty-three specimenswere captured during spring and summer in four locations in Patagonia, Argentina. The stomach contents of the specimens wereidentified, and we found that prey consisted of six main groups: Coleoptera, Hymenoptera, Homoptera and Araneae, and theadults and larvae of moth Lepidoptera. Lepidoptera was the major dietary component. The presence of ants and moths as commonprey suggests an ambush feeding strategy. In spring, females consumed more ants than males although no other dietary differencesbetween males and females were evident. We found nine geckos with empty stomachs and six parasitized by *** of empty stomachs corroborates previous observations of other nocturnal geckos and non-gecko lizards. Reduced foragingsuccess of nocturnal lizards could be due to difficulty in prey detection due to dim light, reduced or erratic activity of insectprey at night, or shorter activity times of geckos relative to diurnal success. Sex and season were not associated with the incidenceof empty stomachs. Principal component analysis showed that four food alternatives correlated with season. The constraint ofnocturnality, coupled with low night-time temperatures restricting feeding to only a few hours after sunset, appear to have causeda generality of diet which may limit energy acquisition. We conclude that H. darwini is an arthropod generalist and likely an ambushforager, as are many other nocturnal

Assessment of the Visual Analogue Score in the evaluation of the Pruritus of Cholestasis

Background and Aims:A visual analogue score (VAS),based on application of a visual analogue scale,has been widely used to assess pruritus in clinical studies of patients with cholestatic liver disease.A VAS is a numerical score of the severity of the perception of pruritus,and,hence,is inherently *** objective of this study was to assessthe reliability of a VAS as an index of pruritus in cholestatic ***:In 8 patients with chronic pruritus due to primary biliary cholangitis,values for a VAS of pruritus were compared with corresponding measurements of scratching activity,which were generated by a monitoring system specifically designed to quantitate this *** relationship between individual values for the VAS and corresponding values for scratching activity during a specific interval immediately preceding the recording of the VAS was examined by determining the Spearman's rank correlation ***:The mean Spearman's rank correlation coefficient between individual values for the VAS and corresponding mean values for scratching activity was 0.072;the range of these coefficients was-0.04 to 0.26.A VAS of pruritus is an unreliable index of scratching activity,and,hence,of the pathophysiological process responsible for the pruritus of ***:It is concluded that the use of a VAS as a primary quantitative endpoint in trials of the efficacy of potential therapies for the pruritus of cholestasis may be inappropriate.
