Mineralogy and geochemistry of sands of the lower course of the Sanaga River, Cameroon: implications for weathering,provenance, and tectonic setting

This study focuses on the mineralogy and bulk chemical composition of 19 sediment samples,collected from the Sanaga River bed,between Nanga-Eboko and the Atlantic Ocean in Cameroon,to infer provenance,weathering,and tectonic setting.The textural analysis revealed that these sediments are mainly coarse-grained sands.X-ray diffraction and heavy mineral analyses show that these sands are made of quartz,microcline,plagioclase,muscovite,ilmenite,anatase,magnetite,opaques,and epidote.The Index of Chemical Variability values(1.07–2.68)indicates that the sands are immature.The Chemical Index of Alteration(53–66%),Plagioclase Index of Alteration(PIA;57–75%),and Mafic Index of Alteration(54–67%)values revealed a moderate intensity of weathering for these sediments.According to the compositional maturity diagram,the sands are mainly litharenites.The studied sands show enrichment in light rare earth elements relative to heavy rare earth elements and a negative anomaly in Eu(Eu/Eu*=0.32–0.83).These sands were derived from felsic metamorphic rocks dated Meso to Neoproterozoic from the Pan-African(700–1000 Ma)domain of south Cameroon.Th/U ratios(mean=5.40;n=19)reveal that these sands originating from felsic source rocks and are low to moderately recycled.On the tectonic discrimination diagrams,all the samples are plotted in the rift and passive margin domains,which is consistent with the tectonic history of Pan African in southern Cameroon.

Dear Editor,The most severe type of male infertility,nonobstructive azoospermia(noa),is characterized by clinical heterogeneity,implying the involvement of different acquired and genetic factors.However,the cause of noa is unidentified in a substantial number of patients after the exclusion of all-known acquired factors and routine genetic testing,such as karyotype and Y chromosome microdeletion analysis.1 With the recent rapid development of whole-exome sequencing(WES),an increasing number of novel monogenic causes have been identified.Recently,RAD51-associated protein 2(RAD51AP2)was reported to be a novel causative gene of noa in a monogenic recessive manner in two sporadic patients.2 Here,we studied the WES data for a family with two noa patients and found that both carried a novel homozygous loss-of-function(LoF)mutation in RAD51AP2.

目的研究常染色体显性遗传2型多囊肾病基因(PKD2)在精子尾部组装过程中的作用。方法回顾分析1例携带PKD2基因突变的非梗阻性无精症(noa)患者的临床资料,Sanger测序验证该基因突变,对睾丸组织进行HE和免疫荧光染色;通过对正常人精子涂片进行免疫荧光染色观察PKD2在精子的定位,利用人视网膜色素上皮(hRPE)细胞检测PKD2在纤毛发育过程中的定位。结果该noa患者PKD2基因存在c.1571T>A(P.I524N)杂合突变,其精子细胞尾部发育缺陷;PKD2主要定位于精子鞭毛起始部,与参与精子尾部组装及发育的重要蛋白Ac-Tubulin及γ-Tubulin存在共定位,在hRPE细胞纤毛发育过程中PKD2主要定位于纤毛的基体。结论本研究报道了1例全新的PKD2杂合突变的noa患者病例,结果提示PKD2可能参与精子尾部的起始组装,为进一步研究精子鞭毛组装的机制提供了基础,PKD2基因c.1571T>A(P.I524N)杂合突变可能是无精症诊疗的新靶点。

Being infected by SARS-CoV-2 may cause damage to multiple organs in patients, such as the lung, liver and heart. Angiotensin-converting enzyme 2(ACE2), reported as a SARS-CoV-2 receptor, is also expressed in human male testes. This suggests a potential risk in human male reproductive system. However, the characteristics of ACE2-positive cells and the expression of other SARS-CoV-2 process-related genes are still worthy of further investigation. Here, we performed singlecell RNA seq(scRNA-seq) analysis on 853 male embryo primordial germ cells(PGCs) and 2,854 normal testis cells to assess the effects of the SARS-CoV-2 virus on the male reproductive system from embryonic stage to adulthood. We also collected and constructed the scRNA-seq library on 228 Sertoli cells from three non-obstructive azoospermia(noa) patients to assess the effects at disease state. We found that ACE2 expressing cells existed in almost all testis cell types and Sertoli cells had highest expression level and positive cells ratio. Moreover, ACE2 was also expressed in human male PGCs. In adulthood, the level of ACE2 expression decreased with the increase of age. We also found that ACE2 positive cells had high expressions of stress response and immune activation-related genes. Interestingly, some potential SARS-CoV-2 process-related genes such as TMPRSS2, BSG, CTSL and CTSB had different expression patterns in the same cell type. Furthermore, ACE2 expression level in noa donors’ Sertoli cells was significantly decreased. Our work would help to assess the risk of SARS-CoV-2 infection in the male reproductive system.

Altered microRNA profiles of testicular biopsies from patients with nonobstructive azoospermia

Many studies have shown that microRNAs(miRNAs)play vital roles during the spermatogenesis.However,little is known about the altered miRNA profiles of testicular tissues in nonobstructive azoospermia(noa).Using microarray technology,the miRNA expression profiles of testicular biopsies from patients with noa and of normal testicular tissues were determined.Bioinformatics analyses were conducted to predict the enriched biological processes and functions of identified miRNAs.The microarray data were validated by quantitative reverse transcriptase polymerase chain reaction(qRT-PCR),the results of which were then validated with a larger sample size.Correlations between the miRNA expression levels and clinical characteristics were analyzed.Receiver operating characteristic(ROC)curve analysis was used to evaluate the diagnostic ability of miRNAs for azoospermia.Hierarchical clustering showed that 129 miRNAs were significantly differentially expressed between the noa and control groups.Bioinformatics analysis indicated that the differentially expressed miRNAs were involved in spermatogenesis,cell cycle,and mitotic prometaphase.In the subsequent qRT-PCR assays,the selected miRNA expression levels were consistent with the microarray results,and similar validated results were obtained with a larger sample size.Some clinical characteristics were significantly associated with the expression of certain miRNAs.In particular,we identified a combination of two miRNAs(miR-10b-3p and miR-34b-5p)that could serve as a predictive biomarker of azoospermia.This study provides altered miRNA profiles of testicular biopsies from noa patients and examines the roles of miRNAs in spermatogenesis.These profiles may be useful for predicting and diagnosing the presence of testicular sperm in individuals with azoospermia.

We performed this study to evaluate the clinical outcomes of microdissection testicular sperm extraction-intracytoplasmic sperm injection(micro-TESE-ICSI)treatment that used fresh or cryopreserved sperm in patients with nonobstructive azoospermia(noa).A total of 338 noa patients with 344 consecutive cycles received treatment in the reproductive medicine center of Peking University Third Hospital in Beijing,China,from January 2014 to December 2017.Fresh oocytes and fresh sperm were used in 222 patients with 234 cycles(Group A).Fresh oocytes and cryopreserved sperm were used in 116 patients with 110 cycles(Group B).We compared patient characteristics,embryonic development,and pregnancy outcomes between Groups A and B.There was no statistical difference in the patient characteristics,and no differences were observed with fertilization or quality embryo rates between Groups A and B.The rates of clinical pregnancy and live birth were both higher for Group A than those for Group B(both Pnoa.

精索静脉曲张或无精子症男性的睾酮水平

目的:调查不育症男性雄激素缺乏的发生率及其可能影响因素。方法:选择2011年1月至2012年12月因精索静脉曲张(varicocele,VC)、梗阻性无精子症(obstructive azoospermia,OA)和非梗阻性无精子症(nonobstructive azoospermia,noa)入北京大学第一医院泌尿外科手术治疗患者的病例资料进行回顾性分析,所有患者在治疗前检查血清总睾酮水平,OA、noa和VC患者数量分别为141,97和169例,所有患者均在上午检测总睾酮浓度。所有样本在男科实验室采用放射免疫方法检测总睾酮水平,低于300 ng/dL定义为雄激素缺乏。结果:共收集到病例407例,平均年龄(30.4±5.8)岁,平均睾酮水平(4.18±1.64)ng/dL(0.30~11.32 ng/dL)。总体雄激素缺乏的发生率为26.5%(108/407),在noa、OA和VC组的雄激素缺乏的发生率分别为40.2%(39/97)、19.7%(27/141)和24.9%(42/169)。noa和VC组雄激素缺乏的发生率高于OA组(Pnoa和VC可能是雄激素缺乏的危险因素,noa的年轻男性出现雄激素水平低下的可能性更大,noa和VC治疗后应该随访睾酮水平。在临床中对不育症男性应该评估是否有雄激素缺乏存在。

目的描述超声引导下生精小管定位新方法在显微镜取精术中的应用，评价其效果。方法选取非梗阻性无精子症(noa)患者44例，平均年龄(31±3.3)岁，均经过病史采集、临床触诊、精液分析、经超声检查及显微镜取精术。超声定位生精小管方法步骤：睾丸短轴切面寻找睾丸结构，睾丸长轴切面局部放大寻找增粗、饱满的生精小管，对其进行术前定位并描绘定位示意图，取精术前在睾丸实体进行定位。计算术前超声定位的生精小管与术中观察到的生精小管的符合率，并统计术中及术后并发症的发生率。结果 44例noa患者共84个睾丸均顺利完成手术，其中79个睾丸术前定位生精小管位置与术中所见一致，符合率达94.0%。术中平均出血量(6.6±4.1)mL,无一例输血，术后3 d内发生血肿0例，术后3 d内发生感染0例，均无严重术中、术后并发症；19例成功获得精子，精子获取率(SRR)为43.1%。结论超声引导生精小管定位新方法在睾丸显微镜取精术中可行，使手术步骤相对简便、高效，具有术中对睾丸损伤小，出血少，术后并发症少、恢复快等优势，可作为辅助显微镜下睾丸切开取精术(Micro-TESE)的一种新方法。
