Effects of Purified Indian Cattle Tick Rhipicephalus microplus Saliva Toxins on Various Enzymes in Blood Serum, Liver and Muscles of Albino Mice

In the present investigation, in vivo effects of purified ticks’ saliva toxin were evaluated on the level of certain important cellular metabolic enzymes i.e. acid phosphatase (ACP), alkaline phosphatase (ALP), glutamate pyruvate transaminase, glutamate oxaloacetate transaminase and lactic dehydrogenase. For this purpose, sub-lethal doses, 40% and 80% of 24 h LD50 purified saliva toxins of Rhipicephalus microplus (Canestrini, 1888) were injected subcutaneously in the albino mice. In treated mice saliva toxins targeted membrane-bound enzymes i.e. serum acid phosphatase and alkaline phosphatase, its level was increased from 118.30% to 163.63% at the 6th hr in comparison to the control. Besides this, the levels of serum glutamate pyruvate transaminase (GPT) and glutamate oxaloacetate transaminase (GOT) and lactic dehydrogenase (LDH) also increased up to 161.11% (at 6th hr), 148.27 (at 8th hr) and 125.45% (at 6th hr) respectively in comparison to control. An increase in the level of LDH showed insufficient oxygen supply, massive disintegration of cells and leakage of the enzyme into the circulation. It clearly indicated the toxic effects of saliva toxins on the membrane of blood cells, hepatocytes and myocardial muscle cell functions in albino mice. On the other hand activity of acetyl cholinesterase was reduced by 65.51% at the 6th hr of the saliva toxin injection in comparison to the control. This inhibition of acetyl cholinesterase activity caused the accumulation of acetylcholine molecules at the synaptic junctions and led to prolonged activation of acetylcholine receptors. It caused permanent stimulation of nerves and muscle cells that may result in muscular paralysis and finally death of the animal.

This article explains ongoing changes in global climate and their effect on the resurgence of vector and pathogen populations in various parts of the world.Today,major prevailing changes are the elevation of global temperature and accidental torrent rains,floods,droughts,and loss of productivity and food commodities.Due to the increase in water surface area and the longer presence of flood water,the breeding of insect vectors becomes very high;it is responsible for the emergence and re-emergence of so many communicable diseases.Due to the development of resistance to chemicals in insect pests,and pathogens and lack of control measures,communicable zoonotic diseases are remerging with high infectivity and mortality.This condition is becoming more alarming as the climate is favoring pathogen-host interactions and vector populations.Rapid changes seen in meteorology are promoting an unmanageable array of vector-borne infectious diseases,such as malaria,Japanese encephalitis,filarial,dengue,and leishmaniasis.Similarly,due to unhygienic conditions,poor sanitation,and infected ground and surface water outbreak of enteric infections such as cholera,vibriosis,and rotavirus is seen on the rise.In addition,parasitic infection ascariasis,fasciolosis,schistosomiasis,and dysentery cases are increasing.Today climate change is a major issue and challenge that needs timely quick solutions.Climate change is imposing non-adaptive forced human migration territorial conflicts,decreasing ecosystem productivity,disease outbreaks,and impelling unequal resource utilization.Rapid climate changes,parasites,pathogens,and vector populations are on the rise,which is making great threats to global health and the environment.This article highlighted the necessity to develop new strategies and control measures to cut down rising vector and pathogen populations in endemic areas.For finding quick solutions educational awareness,technology up-gradation,new vaccines,and safety measures have to be adopted to break the cycle of dreadful communicable

Present review article explains tick-borne diseases,transmission,host immune responses,diagnosis and control in relation to climatic variations.Ticks are hematophagous ectoparasites which suck large volumes of blood from livestock and humans.They release large numbers of protozoans,bacteria,rickettsia and viral pathogens during blood feeding and transmit disease pathogens through saliva.Due to heavy blood sucking by ticks animals face significant blood and weight loss that affect their overall health.Due to more severe illness,high economic losses were noted in livestock.This article highlights medically important tick borne diseases in man and livestock,its pathogenesis,diagnosis and treatment methods.The present article emphasizes invasion of hosts,host-pathogen interactions,tick saliva toxin induced host immune responses and biological effects.This article highlighted various tick control methods i.***.physical killing,acaricidal,biological,hormonal,genetic and immunological methods such as administration of protective antibody and vaccines for disease control in human being and his livestock.The authors suggest non-chemical environmentally safe methods for successful control of tick borne diseases to kill cattle,bird and canine invading ticks.

Targeting colorectal cancer using dietary flavonols

Colorectal cancer is among the well-known forms of cancer and a prominent cause of cancer demises worldwide.In vitro experiments reinforced by animal studies,as well as epidemiological studies of human colorectal cancer propose that the growth of this disease can be moderated by eating aspects.Dietary intake including green vegetables and fruits may result in the reduction of colon cancer chances.The finding suggests that the combinations of dietary nutrients may deliver additive or synergistic effects and might be a powerful method to avoid or eradicate colon cancer beginning and/or development.Flavonols are one of the most widespread dietary nutrients of the polyphenols-flavonoids and major constituent of Allium and Brassicaceae vegetables.Flavonols present in vegetables of Allium and Brassicaceae family are kaempferol,myricetin,quercetin,and isorhamnetin.These flavonols are claimed to have antiproliferative activity in vivo and in vitro against colorectal cancer.The objective of this review is to summarize the role of flavonols obtained from dietary sources in the prevention and treatment of colorectal cancer.

Persistent gastritis induced by Helicobacter pylori(H.pylori)is the strongest known risk factor for gastric cancer(GC).H.pylori is prevalent in about 50%world's population,while it causes cancer in less than 2%of exposed individuals.1 Our studies found that H.pylori infection can induce oncogenic properties in AGS cells by deregulating multiple factors associated with the cell cycle,apoptosis,and other important events associated with cancer progression.

For decades,the gut has been thought to play an important role in sepsis pathogenesis.Sepsis is a serious life-threatening,chronic condition of an infection caused by dysregulated host immune response in most of the intensive care unit patients.Probiotics have dual roles in polymicrobial sepsis i.***.probiotics may induce sepsis in many cases and may prevent its prognosis in many cases.Experimental evidence from both pre-clinical and clinical studies have demonstrated that probiotic therapy ameliorates various inflammatory mediators such as tumor necrosis factor,interleukin-10(IL-10),IL-6,etc.,in septicemia.In addition,probiotic use was also found to reduce the severity of pathological conditions associated with irritable bowel disorder and prevent development of endocarditis in septicemia.On contrary,probiotic therapy in neonatal and athymic adult mice fail to provide any beneficial effects on mortality and sepsis-induced inflammation.Importantly,in few clinical trials probiotic use was found to aggravate sepsis by promoting inflammatory cascade rather than suppressing it.This review discusses various studies regarding the beneficial or harmful effects associated with probiotic therapy in sepsis.

Role of adjunctive nonpharmacological strategies for treatment of rapid-cycling bipolar disorder

Rapid-cycling bipolar disorder(RCBD)is a phase of bipolar disorder defined by the presence of≥4 mood episodes in a year.It is a common phenomenon characterized by greater severity,a predominance of depression,higher levels of disability,and poorer overall outcomes.It is resistant to treatment by conventional pharmacotherapy.The existing literature underlines the scarcity of evidence and the gaps in knowledge about the optimal treatment strategies for RCBD.However,most reviews have considered only pharmacological treatment options for RCBD.Given the treatment-refractory nature of RCBD,nonpharmacological interventions could augment medications but have not been adequately examined.This review carried out an updated and comprehensive search for evidence regarding the role of nonpharmacological therapies as adjuncts to medications in RCBD.We identified 83 reviews and meta-analyses concerning the treatment of RCBD.Additionally,we found 42 reports on adjunctive nonpharmacological treatments in RCBD.Most of the evidence favoured concomitant electroconvulsive therapy as an acute and maintenance treatment.There was preliminary evidence to suggest that chronotherapeutic treatments can provide better outcomes when combined with medications.The research on adjunctive psychotherapy was particularly scarce but suggested that psychoeducation,cognitive behavioural therapy,family interventions,and supportive psychotherapy may be helpful.The overall quality of evidence was poor and suffered from several methodological shortcomings.There is a need for more methodologically sound research in this area,although clinicians can use the existing evidence to select and individualize nonpharmacological treatment options for better management of RCBD.Patient summaries are included to highlight some of the issues concerning the implementation of adjunctive nonpharmacological treatments.

Background The uncanny valley hypothesis states that users may experience discomfort when inter-acting with almost human-like artificial characters.Advancements in artificial intelligence,robotics,and computer graphics have led to the development of life-like virtual humans and humanoid robots.Revisiting this hypothesis is necessary to check whether they positively or negatively affect the current population,who are highly accustomed to the latest technologies.Methods In this study,we present a unique evaluation of the uncanny valley hypothesis by allowing participants to interact live with four humanoid robots that have varying levels of human-likeness.Each participant completed a survey questionnaire to evaluate the affinity of each robot.Additionally,we used deep learning methods to quantify the participants’emotional states using multimodal cues,including visual,audio,and text cues,by recording the participant-robot interactions.Results Multi-modal analysis and surveys provided interesting results and insights into the uncanny valley hypothesis.

Lead free polycrystalline ceramics （K0.5Na0.5）Nb（1-x）MoxO3 （x = 0, 0.02, 0.04, 0.06 and 0.08） have been synthesized via solid state reaction method. The formation of single phase perovskite structure up to 6 mol% of Mo^6＋ has been confirmed by X-ray diffraction pattern. Impedance spectroscopy reveals that bulk resistance decreases with increasing temperature, which indicates negative temperature coefficient of resistance （NTCR） behaviour of the compounds. The diffuse reflectance spectroscopy results indicate a red shift of the band gap energy of K0.5Na0.5NbO3 （KNN, from 4.28 to 3.61 eV） with increasing Mo^6＋ concentration due to structural modification. The photoluminescence spectra of doped samples are composed of two emission bands at room temperature. One emission band is near band edge ultraviolet （UV） emission （354 nm） and other is visible emission band （-397 nm） which may explore the possibility of these ceramics to be used in optical device applications.
