Screening and identification of mimotopes of the major shrimp allergen tropomyosin using one-bead-one- compound peptide libraries

The one-bead-one-compound （OBOC） combinatorial peptide library is a powerful tool to identify ligand and receptor interactions. Here, we applied the OBOC library technology to identify mimotopes specific to the immunoglobulin E （IgE） epitopes of the major shellfish allergen tropomyosin. OBOC peptide libraries with 8-12 amino acid residues were screened with serum samples from patients with shellfish allergy for IgE mimotopes of tropomyosin. Twenty-five mimotopes were identified from the screening and their binding reactivity to tropomyosin-specific IgE was confirmed by peptide ELISA. These mimotopes could be divided into seven clusters based on sequence homology, and epitope mapping by EpiSearch of the clustered mimotopes was performed to characterize and confirm the validity of mimotopes. Five out of six of the predicted epitopes were found to overlap with previously identified epitopes of tropomyosin. To further confirm the mimicry potential of mimotopes, BALB/c mice were immunized with mimotopes conjugated to keyhole limpet hemocyanin and assayed for their capacity to induce tropomyosin-specific antibodies. BALB/c mice that received mimotope immunization were found to have an elevated level of tropomyosin-specific immunoglobulin G, but not mice that received an irrelevant mimotope. This study pioneers the successful application of the OBOC libraries using whole sera to screen and identify multiple shrimp allergen mimotopes and validates their mimicry potential using in vitro, in vivo, and in silico methods.

Increasing dietary fiber intake in terms of kiwifruit improves constipation in Chinese patients

AIM： To investigate if increased dietary fiber, in terms of kiwifruit, is effective in Chinese constipated patients. METHODS： 33 constipated patients and 20 healthy volunteers were recruited for a 4-wk treatment of kiwi fruit twice daily. Response during wk 1-4 was defined as an increase in complete spontaneous bowl, motion （CSBM）≥ 1/wk. Secondary efficacy included response during wk 1-4, individual symptoms and scores of bowel habits and constipation. Responses were compared with the baseline run-in period. Colonic transit time and anorectal manometry were performed before and after treatment. RESULTS： Responder rate was 54.5% in the constipated group. The mean CSBM increased after treatment （2.2±2.6 vs 4.4± 4.6, P = 0.013）. There was also improvement in the scores for bothersomeness of constipation （P = 0.02）, and satisfaction of bowel habit （P = 0.001）, and decreased in days of laxative used （P = 0.003）. There was also improvement in transit time （P = 0.003） and rectal sensation （P 〈 0.05）. However, there was no change in the bowel symptoms or anorectal physiology in the healthy subjects. CONCLUSION： Increasing dietary fiber intake is effective in relieving chronic constipation in Chinese population.

Comparison between empirical prokinetics, Helicobacter test-and-treat and empirical endoscopy in primary-care patients presenting with dyspepsia: A one-year study

AIM： To investigate the optimal strategy to treat dyspeptic patients in primary care. METHODS： Dyspeptic patients presenting to primary care outpatient clinics were randomly assigned to- （1） empirical endoscopy, （2） H pylori test-and-treat, and （3） empirical prokinetic treatment with cisapride. Early endoscopy was arranged if patients remained symptomatic after 2 wk. Symptom severity, quality-of-life （SF-36） as well as patient preference and satisfaction were assessed. All patients underwent endoscopy by wk 6. Patients were followed up for one year. RESULTS： Two hundred and thirty four patients were recruited （163 female, mean age 49）. 46% were H pylori positive. 26% of H pylori tested and 25% of empirical prokinetic patients showed no improvement at wk 2 follow-up and needed early endoscopy. 15% of patients receiving empirical cisapride responded well to treatment but peptic ulcer was the final diagnosis. Symptom resolution and quality-of-life were similar among the groups. Costs for the three strategies were HK$4343, $1771 and $1750 per patient. 66% of the patients preferred to have early endoscopy. CONCLUSION： The three strategies are equally effective. Empirical prokinetic treatment was the least expensive but peptic ulcers may be missed with this treatment. The H pylori test-and-treat was the most cost-effective option .

＜正＞Objectives: Reactivation of varicella zoster virus （VZV）, clinically manifested as herpes zoster （HZ）, is an important complication after haematopoietic stem cell transplantation （HSCT）. At present, there is no effective prophylaxis for this condition. We have investigated the effect of vaccinating donors with a live-attenuated VZV vaccine before HSCT in terms of patients’ humoral and cellular immunity to VZV and the occurrence of HZ after HSCT.

OBJECTIVE Pain sensations in the periphery are carried by primary sensory neurons whose cell bodies lay in the dorsal root ganglia(DRG).The present study focuses on examining the interactions between DRG neurons and glial cells in vitro in relation to their roles as detectors of tissue *** Acutely dissociated DRG cells were isolated from adult SD rats,and plated on poly-DL-ornithine and laminin-coated tissue culture *** in 3 cell groups were compared over time in culture: mixed DRG cells(45%neurons on plating),neuron-enriched cells(70%neurons on plating) and glial cells(99%glial cells on plating).Cell proliferation was determined by counting cells,protein assay and MTT assay.[H]cAMP production in the presence of specific receptor agonists and antagonists was assayed to detect Gs-coupled *** acidic fibrillary protein(GFAP),glutamine synthase(GS) and Toll-like receptor 4(TLR4) mRNA and protein expression were determined by real time PCR and immunocyto-chemistry, *** The presence of Gs-coupledβAR,CGRP,EP and IP receptors were identified in glial cell preparations although only CGRP receptors have been reported on satellite glial cells in ***,IP receptor-stimulated adenylyl cyclase activity in glial cells was significantly inhibited by the presence of DRG *** neurons did not overtly affect glial cell proliferation or the expression of GFAP(classical markers of glial cell activation),but appeared to inhibit the expression of TLR4 and prevent the loss of GS in glial *** Time-dependent changes occur in DRG glial cells in vitro with regard to the expression of TLR4,Gs-coupled receptors and glial cell markers,and all these changes are influenced by the presence of DRG neurons. Whether similar neuron-glial interactions occur in vivo remains to be determined.

The cMyb transcription factor is a key regulator of hematopoietic cell proliferation and differentiation and the dysregulation of cMyb activity often causes hematological ***,its pathogenetic role remains largely ***,we introduced a cmyb overexpressing transgenic zabrafish Tg(cmyb:eGFP),in which the expression level of cmyb is elevated continuously under the control of the endogenous cmyb *** cmyb overexpressing zebrafish,granulocytic hyperplasia was exhibited in all fish fromembryonic stage to ***,as the fish age,a substantial number of them develop acute myeloid leukemia(AML)-and acute lymphoid leukemia(ALL)-like disorders with the characteristics of infiltration of leukemic blasts in blood circulation and other non-hematopoietic tissues including gill,muscle,liver,eye and central nervous *** phenotypes resemble human myelodysplastic syndromes(MDS),AML and ALL,and anti-leukemia chemotherapy treatment could reduce the hematopoietic abnormality of Tg(cmyb:eGFP)embryos with cmyb overexpression were efficiently responded *** study establishes an animal model for dissecting the molecular basis underlying cMyb-associated leukemogenesisas well as for drug discovery.
