Chemically and biologically activated biochars slow down urea hydrolysis and improve nitrogen use efficiency

Biochar is considered a potential technology to enhance chemical fertilizer use efficiency through intensification of the interactions between nutrients and the functional groups on biochar ***,little is known about how the application of activated biochars mixed with urea influences nitrogen(N)mineralization and crop performance in paddy ***,a sawdust-derived fresh biochar(FBC)(ca.400℃)was activated chemically with 15%hydrogen peroxide and biologically with a nutrient solution mixed with a soil inoculum to obtain a chemically activated biochar(CBC)and a biologically activated biochar(BBC),*** chemical and surface properties of FBC,CBC,and BBC were evaluated using spectroscopic methods,i.e.,Fourier transform infrared spectroscopy and 13C nuclear magnetic resonance,and potentiometric charge *** N retention capacity of biochars and their interaction with urea hydrolysis were examined in a laboratory incubation ***,a field experiment was carried out in a paddy field with the biochars unmixed or mixed with urea at a 1:1 *** results showed that negative surface functional groups and negative charges were increased on both activated biochars,especially *** activated biochars contributed to a significant reduction in urea-biochar suspension ph and increased N retention in the incubation *** the enhanced surface properties of the activated biochars,no similar increases in rice biomass and grain yield were observed for these biochars in the field ***,rice biomass,grain yield,apparent N use efficiency,and agronomic N use efficiency were significantly higher with the application of the three biochars compared to no-biochar ***,the results indicate that the application of urea mixed with biochar could enhance crop performance,especially in the case of activated biochar,which would enhance N retention in the soil,reducing N loss.

Bioreactor technology for clonal propagation of plants and metabolite production

Plant cell culture in bioreactors is an enabling tool for large scale production of clonal elite plants in agriculture, horticulture, forestry, pharmaceutical sectors, and for biofuel production. Advantages of bioreactors for plant cell culture have resulted in various types of bioreactors differing in design, operating technologies, instrumentations, and construction of culture vessels. In this review, different types of bioreactors for clonal propagation of plants and secondary metabolites production are discussed. Mechanical and biochemical parameters associated with bioreactor design, such as aeration, flow rate, mixing, dissolved oxygen, composition of built-up gas in the headspace, and ph of the medium, are pivotal for cell morphology, growth, and development of cells within tissues, embryos, and organs. The differences in such parameters for different bioreactor designs are described here, and correlated to the plant materials that have been successfully cultured in different types of bioreactors.

Systematic mutational analysis of epitope-grafted ED3 immunogenicity reveals a DENV3-DENV4 bi-serospecific ED3 mutant

Objective:To identify the residue determinants of the serospecificity and sero-cross-reactivity of dengue virus(DENV)envelope protein domain 3(ED3),which contains two major putative epitopes of ***:We constructed ED3 from DENV3(3ED3)and DENV4(4ED3),and six epitope-grafted variants,where we transferred epitope 1(L304I,K305D,V309M,and S310A)and/or epitope 2(D383N,K384S,K387T,and N389h)of 4ED3 onto 3ED3 and *** albino mice aged 3-4 weeks were immunized against wildtype and epitope-grafted ED3 variants and anti-ED3 IgG antibody responses were determined using ***:Mouse immunization using 3ED3 and 4ED3 generated serotype-specific antisera,as ***,most epitope-grafted ED3s produced antisera serospecific to the template ED3 with little or no cross-recognition of ED3 of the serotype from which the epitopes were *** indicated that a mere grafting of the epitope was not sufficient to transfer serospecificity,contrary to our ***,one epitope-grafted ED3 mutant,where epitope 1 of 3ED3 was grafted onto 4ED3(4ED3^(epi1)),generated antisera that was serospecific to both 4ED3 and ***:The 4ED3^(epi1)is a chimeric ED3 that produces antisera possessing serospecificity to both 3ED3 and 4ED3 onto a common 4ED3 *** 4ED3^(epi1),therefore,provides a unique tool for analyzing serospecificity and sero-cross-reactivity in *** believe that chimeric ED3 may provide a template for future recombinant ED3 possessing serospecificity of multiple DENVs onto a single scaffold and may pave a way developing tri-and/or tetravalent anti-DENV antisera.

Estimation and comparison of night-time Oh levels in the UK urban atmosphere using two different analysis methods

Night-time Oh levels have been determined for UK urban surface environments using two methods, the decay and steady state approximation methods. Measurement data from the UK National Environmental Technology Centre archive for four urban sites （Bristol, harwell, London Eltham and Edinburgh） over the time period of 1996 to 2000 have been used in this study. Three reactive alkenes, namely isoprene, 1,3-butadiene and trans-2-pentene were chosen for the calculation of Oh levels by the decay method. hourly measurements of NO, NO〉 03, CO and 20 VOCs were used to determine night-time Oh level using the steady state approximation method. Our results showed that the night-time Oh levels were in the range of 1 × 10^3-1× 10^6 molecules/cm3 at these four urbap sites in the UK. The application of a t-test of these analyses indicated that except Bristol, there was no significant difference between the Oh levels found from the decay and steady state approximation methods. Night-time levels of the Oh radical appeared to peak in summer and spring time tracking the night-time 03 levels which also passed through a maximum at this time.

In vitro α-amylase inhibitory activity and in vivo hypoglycemic effect of methanol extract of Citrus macroptera ***

Objective:To investigate the therapeutic effects of methanol extract of Citrus macroptera Montr,fruit inα-amylase inhibitory activity(in vitro)and hypoglycemic activity in normal and glucose induced hyperglycemic rats(in vivo).Methods:Fruits of Citrus macroptera without rind was extracted with pure methanol following cold extraction and tested for presence of phytochemical constituents,α-amylase inhibitory activity,and hypoglycemic effect in normal rats and glucose induced hyperglycemic ***:Presence of saponin,steroid and terpenoid were identified in the *** results showed that fruit extract had moderateα-amylase inhibitory activity[IC_(50)value=(3.638±0.190)mg/mL]as compared to *** at 500 mg/kg and 1000 mg/kg doses fruit extract significantly(Ph but 1000 mg/kg dose significantly reduced blood glucose level at 2 h and 3 h(Phereas glibenclamide(5 mg/kg)significantly reduced glucose level at every hour after *** time effect is also considered extremely significant with F value=23.83 and P value=0.0001 in oral glucose tolerance ***:These findings suggest that the plant may be a potential source for the development of new oral hypoglycemic agent.

Knowledge and Skills for Management of Sexually Transmitted Infections by Rural Medical Practitioners in Bangladesh

Sexually transmitted infection (STI) management is considered rudimentary among rural medical practitioners (RMPs) in Bangladesh. We sought to understand the level of knowledge and skills in STI management and to assess the impact of a two-day training orientation among RMPs in Tangail district. Data were collected through a baseline survey of 225 practicing RMPs in the study area and a three-month follow-up survey of 99 RMPs who participated in a two-day STI/hIV orientation training. The level of formal training among RMPs ranged from none (22.7%), to paramedical training (14.7%) and local medical assistant training (62.6%). The baseline survey revealed a low level of STI/hIV knowledge and misconceptions about the transmission of STI/hIV among RMPs. RMPs mostly prescribed first line antibiotics for treatment of common reproductive tract infections (RTIs) including STIs, but they rarely prescribed the correct dosages according to the national RTI/STI management guidelines. Only 3% of RMPs were able to correctly answer all four hIV transmission (unprotected sexual intercourse, blood transfusion, needle sharing and mother to child transmission) questions at baseline, while 94.9% of RMPs answered all four correctly at three months following the training (p = 0.001). Only 10% of RMPs reported suggesting the recommended drug (azithromycin) and only 2% mentioned about the recommended dosage (2 gm single dose) for the treatment of urethritis/cervicitis;compared to 49.5% suggested azithromycin at follow-up with 39.4% mentioned the recommended 2 gm single dose (p = 0.001). Our study found low level of knowledge and poor practices related RTI/STI management among RMPs. Short orientation training and education intervention shown promise to increase knowledge and management skills for RTIs/STIs.

Traditional Use, Phytochemistry, Pharmacological and Toxicological Properties of Acanthus ilicifolius: A Review

The practice of plants for the deterrence and management of innumerable physiological states and diseases dates back to the history of humankind. Although many of today’s medicinal yields are achieved synthetically, they are often modelled on the chemical composition of medicinal plants. In this accordance, Acanthus genus (Family Acanthaceae) has acknowledged considerable attention due to its wide range of secondary metabolites and its customary usage in Indian and Chinese system of medicine. It is worth to mention that A. ilicifolius is enormously medicinal among all the species of Acanthus genus. This plant is considered to be rich sources of steroids, triterpenoids, saponins, flavonoids, alkaloids, and tannins. Traditionally, the plant has been used for asthma, diabetes, dyspepsia, hepatitis, leprosy, neuralgia, paralysis, ringworm, skin disease, snake bite, leukemia. Toxicological investigation in mice model has showed that, the ethanolic extract of the leaves of this plant have no toxic effect up to dose of 1000 mg/kg BW, 2000 mg/kg BW dose have mild toxicity and 5000 mg/kg BW dose have caused toxic effect in some organ system like liver, heart, pancreas, lung and kidney. Different extract of this plant also exhibited Anti-inflammatory, anti-leishmanial, osteoblastic, hepatoprotective, antioxidant and anticancer activities. This review article is an endeavor to cover current expansion in phytochemical and pharmacological potential of this plant species which will be a good reference tool for investigators who wish to work on natural products.

The present study investigated the potential probiotic effect of Enterococcus faecalis against a pathogenic microorganism,Vibrio harveyi occurring in the giant freshwater prawn(Macrobrachium rosenbergii).*** was isolated from the intestine of *** through biochemical tests and PCR-based molecular *** in-vitro challenge was conducted by mixing isolated probiotic(5.12 log CFU/g)and ***(3.09 log CFU/g).Feeds incorporated with ***(9.02 log CFU/g)were applied to the treatment group during in vivo test in the aquarium and earthen pond(70 m^(2),1 m depth).Pathogenic bacteria were found to be reduced after 8 h(from 5.02 to 3.62 log CFU/g)and 60 days(from 5.44 to 2.15 log CFU/g)of probiotic administration in the in vitro and in vivo test,*** *** mixed feed also showed higher weight gain(WG%,481.22%±17.71%;SGR%,2.93%±0.18%)and digestive enzymes activities(amylase,1.26±0.08 unit/mg;protease,2.78±0.02 unit/mg)compared to control group(WG%,371.31%±13.35%;SGR%,2.30%±0.32%;amylase,0.7±0.03 unit/mg;protease,1.82±0.02 unit/mg).Furthermore,the probiotic also improved the immune response by augmenting NGh(from 73.33%±0.23%to 74.63%±0.11%)and SGh(from 22.24%±0.06%to 23.13%±0.08%)in the treatment ***,*** could be recommended to use against bacterial infections of the ***.

Development of Generic Drug Products by Pharmaceutical Industries Considering Regulatory Aspects: A Review

Development of generic drug product by pharmaceutical industry is a scientific and technical approach which is totally different from developing a reference or innovator product. Most of the developing countries focus on developing the generic drug products because huge amount of investment is required for innovation and to develop reference product. The generic medicine has to be bioequivalent to the innovator drug and ensure the same biological effect with proper safety and efficacy. Nowadays, the pharmaceutical industries focus on the development of generic product as this does not require that much time and cost compared to the innovator company. But development of generic product is also difficult as it contains the same therapeutic efficacy as innovator. The development approach is based on the target market, i.e. US market, EU market. If a manufacturer targets the US market, then all excipients should be USP grade, analysis should be conducted by USP method or in-house method and stability studies as well. Prior and during the development of generic drug product API selection, dosage form selection, reference product selection and characterization, formulation development, analytical method development, tech transfer or submission batch are prime concern. Then again, bioequivalence study, drug registration procedure and commercialization of the generic product considering regulatory guidance of respective regulatory agencies and the approaches taken by the regulatory agencies for the development of registration of generic medicines are also crucial as well for the development of generic drug product. The aim of this study was to review the entire stage of a generic drug development by a generic pharmaceutical company.
