Effects of stinging nettle (Urtica dioica) powder on laying performance,egg quality,and serum biochemical parameters of Japanese quails

The aim of this experiment was to evaluate the effects of dietary supplementation of stinging nettle powder(SNP) on laying performance,egg quality,and some selected serum biochemical parameters of quails.One hundred and forty-four 10-wk-old Japanese quails(initial body weight=199±18 g) were divided into 3 dietary treatment groups(basic diet without SNP [SNPO],SNPO with 3% SNP [SNP3],SNPO with 6% SNP [SNP6]) with 4 replicates of 12 quails for a rearing period of 12 wk.At 22 wk of age,the final body weights of the SNP3 and SNP6 groups were significantly(P=0.001) reduced compared to that of the SNPO group.Daily feed intake was not statistically different among the groups.The mean number of eggs laid ranged from 65 to 69 with laying rates from 76.8% to 82.1%.The percentage of cracked eggs was not significantly different among the groups and ranged from 1.6% to 1.9%.The egg weight was similar and the feed conversion ratio was closer among the groups.The egg yolk cholesterol,serum cholesterol and serum triglyceride levels in the SNP6 group were significantly reduced(P <0.001) compared to those of the SNPO group.Serum Ca,P and Mg were not significantly influenced by the supplementation.In conclusion,the results demonstrated that the supplementation of SNP to the quail diet at the level of 6%reduced quail egg yolk cholesterol,serum total cholesterol and serum triglyceride levels and did not negatively influence quail performance.

Heritability and Genetic Correlation of Niamey’s Local Chicken Growth (Niger)

The exploitation of industrial strains of chickens in the Sahelian climate of Niger is characterized by a decline in performance and significant costs associated with their maintenance. In contrast, local chickens are well adapted to these environmental conditions but with poor production performance. Genetic selection of these local chickens could improve their productivity. The first step is to determine if the genetic parameters of their growth are high enough to ensure a successful selection strategy. To do so, weekly weights of 69 parents and 119 offspring were followed for 20 weeks. The heritability and genetic correlations of these weights were estimated through the Bayesian approach using the MCMCglmm package on R software. At hatching, weights ranged from 23 to 25 g. At 20 weeks, these weights ranged from 1031 to 1052 g for females and 1308 to 1445 g for males. Heritabilities for hatch weights at 4, 8, 12, 16, and 20 weeks of age were estimated to be 0.56, 0.31, 0.52, 0.53, 0.52 and 0.48 respectively and all genetic correlations were positive. In particular, weight at 8 weeks of age showed both good heritability (h2 = 0.52) and strong, positive genetic correlations with weights at older ages. These results indicate that genetic selection to improve weight at 8 weeks of age would be a good strategy to improve the overall growth performance of these chickens.

Performances of local poultry breed fed black soldier fly larvae reared on horse manure

In poultry,feed based on maggots,like larvae of black soldier fly(Hermetia illucens) is an attractive option to substitute current ingredients which are expensive and often in direct or indirect competition with human food.Little information is currently available on the utility of these larvae in poultry feed,so goals of this study were to determine whether larvae could be reared on horse manure under traditional farming conditions and to evaluate the growth performances of a local poultry fed these larvae and the fatty acids profiles of their meat.After freezing and thawing,larvae were introduced in the feed of Ardennaise chickens between 30 and 80 days of age.Birds in the control group received a commercial standard feed,while those in the treatment group received the same commercial feed in which 8% was substituted with whole fresh larvae corresponding to 2% on a dry matter basis.Means ± standard errors of larval length and weight were 20.67 ± 2.21 mm and 0.14 ± 0.02 g,respectively.Mean larval percentages of dry matter and of substances extractable in diethyl ether were 24.6% and 23.1%,respectively.Larval fatty acids profiles were predominantly composed of lauric acid(28.1%) and palmitic acid(22.0%).Least squares means of weekly weights of chicken,adjusted for the effects of sex,replication and initial weights,were significantly higher(P < 0.05) by 77.03 ± 53.37 g in larvae-fed than in control chickens.All other measurements were not statistically different between larvae-fed and control chicken,including fatty acid profiles,protein content and w6/w3 ratio.

Modeling and measuring glucose diffusion and consumption by colorectal cancer spheroids in hanging drops using integrated biosensors

As 3D in vitro tissue models become more pervasive,their built-in nutrient,metabolite,compound,and waste gradients increase biological relevance at the cost of analysis simplicity.Investigating these gradients and the resulting metabolic heterogeneity requires invasive and time-consuming methods.An alternative is using electrochemical biosensors and measuring concentrations around the tissue model to obtain size-dependent metabolism data.With our hanging-dropintegrated enzymatic glucose biosensors,we conducted current measurements within hanging-drop compartments hosting spheroids formed from the human colorectal carcinoma cell line HCT116.We developed a physics-based mathematical model of analyte consumption and transport,considering(1)diffusion and enzymatic conversion of glucose to form hydrogen peroxide(H_(2)O_(2))by the glucose-oxidase-based hydrogel functionalization of our biosensors at the microscale;(2)H_(2)O_(2)oxidation at the electrode surface,leading to amperometric H_(2)O_(2)readout;(3)glucose diffusion and glucose consumption by cancer cells in a spherical tissue model at the microscale;(4)glucose and H_(2)O_(2)transport in our hangingdrop compartments at the macroscale;and(5)solvent evaporation,leading to glucose and H_(2)O_(2)upconcentration.Our model relates the measured currents to the glucose concentrations generating the currents.The low limit of detection of our biosensors(0.4±0.1μM),combined with our current-fitting method,enabled us to reveal glucose dynamics within our system.By measuring glucose dynamics in hanging-drop compartments populated by cancer spheroids of various sizes,we could infer glucose distributions within the spheroid,which will help translate in vitro 3D tissue model results to in vivo.

Effect of biofilm formation by clinical isolates of Helicobacter pylori on the efflux-mediated resistance to commonly used antibiotics

AIM To evaluate the role of biofilm formation on the resistance of Helicobacter pylori(H. pylori) to commonly prescribed antibiotics, the expression rates of resistance genes in biofilm-forming and planktonic cells were compared.METHODS A collection of 33 H. pylori isolates from children and adult patients with chronic infection were taken for the present study. The isolates were screened for biofilm formation ability, as well as for polymerase chain reaction(PCR) reaction with HP1165 and hp1165 efflux pump genes. Susceptibilities of the selected strains to antibiotic and differences between susceptibilities of planktonic and biofilm-forming cell populations were determined. Quantitative real-time PCR(qP CR) analysis was performed using 16 S r RNA gene as a H. pylori-specific primer, and two efflux pumps-specific primers, hp1165 and hefA.RESULTS The strains were resistant to amoxicillin, metronidazole, and erythromycin, except for one strain, but they were all susceptible to tetracycline. Minimum bactericidalconcentrations of antibiotics in the biofilm-forming cells were significantly higher than those of planktonic cells. qP CR demonstrated that the expression of efflux pump genes was significantly higher in the biofilm-forming cells as compared to the planktonic ones.CONCLUSION The present work demonstrated an association between H. pylori biofilm formation and decreased susceptibility to all the antibiotics tested. This decreased susceptibility to antibiotics was associated with enhanced functional activity of two efflux pumps: hp1165 and hefA.

Combination of Model-based Observer and Support Vector Machines for Fault Detection of Wind Turbines

Support vector machines and a Kalman-like observer are used for fault detection and isolation in a variable speed horizontalaxis wind turbine composed of three blades and a full converter. The support vector approach is data-based and is therefore robust to process knowledge. It is based on structural risk minimization which enhances generalization even with small training data set and it allows for process nonlinearity by using flexible kernels. In this work, a radial basis function is used as the kernel. Different parts of the process are investigated including actuators and sensors faults. With duplicated sensors, sensor faults in blade pitch positions,generator and rotor speeds can be detected. Faults of type stuck measurements can be detected in 2 sampling periods. The detection time of offset/scaled measurements depends on the severity of the fault and on the process dynamics when the fault occurs. The converter torque actuator fault can be detected within 2 sampling periods. Faults in the actuators of the pitch systems represents a higher difficulty for fault detection which is due to the fact that such faults only affect the transitory state(which is very fast) but not the final stationary state. Therefore, two methods are considered and compared for fault detection and isolation of this fault: support vector machines and a Kalman-like observer. Advantages and disadvantages of each method are discussed. On one hand, support vector machines training of transitory states would require a big amount of data in different situations, but the fault detection and isolation results are robust to variations in the input/operating point. On the other hand, the observer is model-based, and therefore does not require training, and it allows identification of the fault level, which is interesting for fault reconfiguration. But the observability of the system is ensured under specific conditions, related to the dynamics of the inputs and outputs. Adaptive Dual Wavelet Threshold Denoising Function Combined with Allan Variance for Tuning FOG-SINS Filter

Allan variance(AV)stochastic process identification method for inertial sensors has successfully combined the wavelet transform denoising scheme.However,the latter usually employs a traditional hard threshold or soft threshold that presents some mathematical problems.An adaptive dual threshold for discrete wavelet transform(DWT)denoising function overcomes the disadvantages of traditional approaches.Assume that two thresholds for noise and signal and special fuzzy evaluation function for the signal with range between the two thresholds assure continuity and overcome previous difficulties.On the basis of AV,an application for strap-down inertial navigation system(SINS)stochastic model extraction assures more efficient tuning of the augmented 21-state improved exact modeling Kalman filter(IEMKF)states.The experimental results show that the proposed algorithm is superior in denoising performance.Furthermore,the improved filter estimation of navigation solution is better than that of conventional Kalman filter(CKF).

Lens thickness assessment : anterior segment optical coherence tomography versus A-scan ultrasonography

AIMTo assess lens thickness measurements with anterior segment-optical coherence tomography (AS-OCT) in comparison with A-scan ultrasonography (A-scan US).METHODSThere were 218 adult subjects (218 eyes) aged 59.2±9.2y enrolled in this prospective cross-sectional study. Forty-three eyes had open angles and 175 eyes had narrow angles. Routine ophthalmic exam was performed and nuclear opacity was graded using the Lens Opacities Classification System III (LOCS III). Lens thickness was measured by AS-OCT (Visante OCT, Carl Zeiss Meditec, Dublin, CA, USA). The highest quality image was selected for each eye and lens thickness was calculated using ImageJ software. Lens thickness was also measured by A-scan US.RESULTSInterclass correlations showed a value of 99.7% for intra-visit measurements and 95.3% for inter-visit measurements. The mean lens thickness measured by AS-OCT was not significantly different from that of A-scan US (4.861±0.404 vs 4.866±0.351 mm, P=0.74). Lens thickness values obtained from the two instruments were highly correlated overall (Pearson correlation coefficient=0.81, P<0.001), and in all LOCS III specific subgroups except in grade 5 of nuclear opacity. Bland-Altman analysis revealed a 95% limit of agreement from -0.45 to 0.46 mm. Lens thickness difference between the two instruments became smaller as the lens thickness increased and AS-OCT yielded smaller values than A-scan US in thicker lens (β=-0.29, P<0.001)CONCLUSIONAS-OCT-derived lens thickness measurement is valid and comparable to the results obtained by A-scan US. It can be used as a reliable noncontact method for measuring lens thickness in adults with or without significant cataract.

Silica‐bonded N‐propylpiperazine sodium n‐propionate(SBPPSP) was found to act as an efficient solid base for the preparation of a series of 4H‐benzo[b]pyran derivatives. SBPPSP was used as a recyclable heterogeneous solid base catalyst for the synthesis of 3,4‐dihydropyrano[c]chromenes, 2‐amino‐4H‐pyrans, 1,4‐dihydropyrano[2,3‐c]pyrazoles, and 2‐amino‐4H‐benzo[e]‐chromenes via the condensation reaction of dimedone, ethyl acetoacetate, 3‐methyl‐1‐phenyl‐1H‐pyrazol‐5(4H)‐ one, and α‐naphthol, respectively, with aromatic aldehydes and malononitrile in refluxing aqueous ethanol. The heterogeneous solid base showed similar efficiency when reused in consecutive reactions.
