Polyphenols as potential enhancers of stem cell therapy against neurodegeneration

The potential of polyphenols for treating chronic-degenerative diseases(particularly neurodegenerative diseases)is ***,the selection of the best polyphenol for each treatment,the mechanisms by which they act,and their efficacy are frequently *** this review,the basics and the advances in the field,as well as suggestions for using natural and synthetic polyphenols alone or in a combinatorial strategy with stem cell assays,are compiled and ***,stem cells exhibit several responses when polyphenols are added to their environment,which could provide us with knowledge for advancing the elucidation of the origin of *** also,polyphenols are being included in the innovative strategies of novel therapies for treating neurodegenerative diseases as well as metabolic diseases related to *** this regard,flavonoid compounds are suggested as the best natural polyphenols due to their several mechanisms for acting in ameliorative effects;but increasing reports are involving other *** if some facts limiting bioactivity prevent them from conventional use,some natural polyphenols and derivatives hold the promise for being improved compounds,judged by their induced *** current results suggest polyphenols as enhancers of stem cell therapy against the targeted diseases.

Coronavirus disease 2019(COVID-19),which is caused by severe acute respiratory syndrome-coronavirus-2(SARS-CoV-2),has affected millions of people *** was declared a pandemic by the World Health Organization in March *** hyperinflammatory response to the entry of SARS-CoV-2 into the host through angiotensin-converting enzyme 2 is the result of a“cytokine storm”and the high oxidative stress responsible for the associated *** only respiratory symptoms are reported,but gastrointestinal symptoms(diarrhea,vomiting,and nausea)and liver abnormalities(high levels of aspartate aminotransferase,alanine aminotransferase transaminases,and bilirubin)are observed in at least 30%of *** food intake and a delay in medical services may lead to malnutrition,which increases mortality and poor *** review provides some strategies to identify malnutrition and establishes nutritional approaches for the management of COVID-19 and liver injury,taking energy and nutrient requirements and their impact on the immune response into *** roles of certain phytochemicals in the prevention of the disease or as promising target drugs in the treatment of this disease are also considered.

AIM To examine the association between weekend alcohol consumption and the biochemical and histological alterations at two different concentrations of alcohol in both genders in *** Wistar rats weighing 170-200 g were divided into groups as follows:(1) Control groups; and(2) weekend alcohol-consumption group: 2 d/weekly per 12 wk, at two different concentrations:(1) Group of males or females with a consumption of a solution of alcohol at 40%; and(2) group of males or females with a consumption of a solution of alcohol at 5%. At the end of the experiment, serum and liver samples were obtained. The following enzymes and metabolites were determined in serum: Alanine Aminotransferase(ALT), Aspartate Aminotransferase(AST), Lactate Dehydrogenase, and Gamma-Glutamyltransferase, and glucose, triglycerides, cholesterol, bilirubin, and albumin. Liver samples from each group were employed to analyze morphological abnormalities by light *** In all of the weekend alcohol-consumption groups, AST activity presented a significant, 10-fold rise. Regarding ALT activity, the groups with weekend alcohol consumption presented a significant increase that was six times greater. Bilirubin levels increased significantly in both groups of females. We observed a significant increase in the parameters of fatty change and inflammation due to weekend alcohol consumption. Only the group of females that consumed alcohol at 40% presented slight hepatocel ular disorganization CONCLUSION The results obtained herein provide solid evidence that weekend alcohol consumption gives rise to liver damage, demonstrated by biochemical and histological alterations, first manifested acutely, and prolonged weekend alcohol consumption can cause greater, irreversible damage.

Methylation of DAPK and THBS1 genes in esophageal gastric-type columnar metaplasia

AIM: To explore methylation of DAPK, THBS1, CDH-1, and p14 genes, and Helicobacter pylori(H. pylori) status in individuals harboring esophageal columnar ***: Distal esophageal mucosal samples obtained by endoscopy and histologically diagnosed as gastric-type(non-specialized) columnar metaplasia, were studied thoroughly. DNA was extracted from paraffin blocks, and methylation status of deathassociated protein kinase(DAPK), thrombospondin-1(THBS1), cadherin-1(CDH1), and p14 genes, was examined using a methyl-sensitive polymerase chain reaction(MS-PCR) and sodium bisulfite modification protocol. H. pylori cag A status was determined by ***: In total, 68 subjects(33 females and 35 males), with a mean age of 52 years, were included. H. pylori cag A positive was present in the esophageal gastric-type metaplastic mucosa of 18 individuals. DAPK, THSB1, CDH1, and p14 gene promoters were methylated by MS-PCR in 40(58.8%), 33(48.5%), 46(67.6%), and 23(33.8%) cases of the 68 esophageal samples. H. pyloristatus was associated with methylation of DAPK(P = 0.003) and THBS1(P = 0.019).CONCLUSION: DNA methylation occurs in cases of gastric-type(non-specialized) columnar metaplasia of the esophagus, and this modification is associated with H. pylori cag A positive infection.

Methane from enteric fermentation is the gas with the greatest environmental impact emitted by ***(Lv)addition to feedstocks could be a strategy to mitigate rumen methane emissions via decreasing the population of methanogenic archaea(MA).Thus,this paper provides the first overview of the effects of Lv supplementation,focusing on the inhibition of methane production,rumen microbiota,and ruminal *** indicated that Lv treatment had a strong anti-methanogenic effect on pure strains of ***,there are uncertainties from in vitro rumen fermentation trials with complex substrates and rumen ***-state fermentation(SSF)has emerged as a cost-effective option to produce *** this way,SSF of agricultural residues as an Lv-carrier supplement in sheep and goats demonstrated a consistent decrease in ruminal methane *** experimental evidence for in vitro conditions showed that Lv did not affect the volatile fatty acids(VFA).However,in vivo experiments demonstrated that the production of VFA was *** did not negatively affect the digestibility of dry matter during in vitro and in vivo methods,and there is even evidence that it can induce an increase in *** the rumen microbiota,populations of MA were reduced,and no differences were detected in alpha and beta diversity associated with Lv ***,some changes in the relative abundance of the microbiota were *** studies are recommended on:(i)Lv biodegradation products and stability,as well as its adsorption onto the solid matter in the rumen,to gain more insight on the“available”or effective Lv concentration;and(ii)to determine whether the effect of Lv on ruminal fermentation also depends on the feed composition and different ruminants.

Structural Changes in Human Teeth after Heating up to 1200°C in Argon Atmosphere

The phase transformation of hydroxyapatite (HAP, Ca10(PO4)6(OH)2) to the beta tricalcium phosphate phase (β-TCP, β-Ca3(PO4)2) at 1100°C is well known. However, in the case of human tooth, the HAP phase transformation is still an open area. For example, the CaO phase has sometimes been reported in the set of phases that make up the teeth. In this study, physical changes of human teeth when subjected to heat treatment in inert atmosphere (argon) were studied. The results were compared with those obtained in air atmosphere, from room temperature (25°C) up to 1200°C. Morphological changes were analyzed by light and scanning electron microscopy (SEM). The HAP to β-TCP phase transformation was followed in powder samples by X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR). Heating of teeth results in the removal of organic material and structural water before the HAP to β-TCP phase transformation, the increment in hardness and the induced crystal growth. The percentage of the phases, crystal growth and lattice parameter variations as a function of temperature was quantified by Rietveld analysis. The black color was observed in dentin heated under argon atmosphere. Differences in expansivity produce fractures in dentin at 300°C in argon and at 400°C in air. In dentin, the coexistence of the HAP and β-TCP phases was observed after 800°C in argon and after 600°C in air;in enamel it was observed at 600°C in argon compared with 400°C in air. In general, the role played by the argon atmosphere during the thermal treatment of the teeth is to retard the processes observed in air.

Callus cultures of Annona muricata and Annona purpurea were induced in Murashige and Skoog(MS)medium supplemented with different concentrations of 1-naphthylacetic acid(NAA),6-benzyladenine(BA)and 2,4-dichlorophenoxyacetic acid(2,4-D)utilized hypocotyls with *** highest percentage of callus formation was the treatment supplemented with 3 mg L-1 NAA for ***(100%)while for *** in lower percentage(75%).BA stimulated the formation of shoots in all the evaluated concentrations,being the concentration of 2 mg L-1 the one that induced the greater formation of shoots for ***(23 shoots/explant)and ***(28 shoots/explant).The content of total phenols,flavonoids and antioxidant activity was measured in the callus obtained from both *** results showed that a higher content of total phenols was quantified in callus of ***(27.8 mg g-1 dw)compared to ***(23.2 mg g-1 dw).The highest content of total flavonoids was observed in the callus of ***(8.0μg g-1 dw).Antioxidant activity was determined by the 2,2-diphenyl-1-picrylhydracil radical *** concentration required for 50%inhibition(IC50)of the 2,2-diphenyl-1-picrylhydracil radicals were 4.22μg mL-1 in methanolic extracts of callus of ***,while in extracts of callus of *** was 2.86μg mL-1,in both cases was greater than that found for *** culture of the species studied in this work represents an alternative for the production of natural antioxidants.

Stressors in Nurse Students’ Formation in the New Normality Post COVID-19

Background: Mental health has been impaired and at risk due to the worldwide COVID-19 pandemic. The consequences due to confinement impacted every scenario, which directly influenced the daily routine of nurse students;in each setting students faced stressors that trigger fear, anxiety and others, since being in confinement learning of topics moved to the home, laboratory practices in hospitals were cancelled leaving the room that is uncertain up to their return to in-person activities. It is important to highlight the need for innovation and strengthening of theoretical-pedagogic aspects centered at the student’s context as a human being with their own needs and problems, who will interact with others in the continuous process of health-illness. Objective: the aim was to identify the stressors in the nurse students’ formation in the new normality post-COVID-19. Methods: Qualitative and phenomenological research with 27 participants aged 20 - 25 years, nurse students of a public university. The information collection was through four focal groups of 6-7 members each, data analysis was done according to Miles & Huberman after signed informed consent of each participant, and authorized by the chairperson of the Nurse’ career. Results: Category 1, Cumulative stressors with sub-categories 1.1 Uncertainty, 1.2 Isolation, 1.3 Invisibility, 1.4 Mockery, 1.5 Exclusion. Category 2, Expectancy states with sub-categories 2.1 Low self-esteem, 2.2 Insecurity, 2.3 Anxiety, 2.4 Depression, 2.5 Temporary leave, 2.6 Search for authenticity. Category 3, Internalization processes with sub-categories 3.1 Social rejection, 3.2 Self-censorship, 3.3 Discrediting, 3.4 Disempowerment. Category 4, Academic aspects affected with sub-categories 4.1 Deficient studying habits, 4.2 Deficient assimilations of knowledge, 4.3 Archived knowledge in the computer, 4.4 Absence of practice in previous semesters.
