BACKGROUND Autoimmunity has emerged as a probable disease modifier in patients with clinically diagnosed type 2 diabetes mellitus(T2DM),that is,patients who have insulin resistance,obesity,and other cardiovascular risk factors,suggesting that the presence of glutamic acid decarboxylase(anti-GAD65),islet antigen 2(anti-IA2),and zinc transporter 8(anti-Zn8T)antibodies could have deleterious effects on beta cell function,causing failure and earlier requirement for insulin *** To evaluate anti-GAD65,anti-IA2 and anti-Zn8T as predictors of early insulin requirement in adolescents with a clinical diagnosis of *** This was a case–control study in patients with clinically diagnosed with T2DM(68 cases and 64 controls with and without early insulin dependence respectively),male and female,aged 12–18 ***,blood pressure,glucose,insulin,C-peptide,glycated hemoglobin A1c,and lipid profiles were *** was used to measure anti-GAD65,anti-IA2,and anti-Zn8T *** statistics,Pearson'sχ2 test,Student's t test,and logistic regression was performed.Plly *** There were 132 patients(53.8%female),with a mean age was 15.9±1.3 years,and there was a disease evolution time of 4.49±0.88 *** presence of anti-GAD65,anti-IA2,and anti-Zn8T positivity was found in 29.5%,18.2%,and 15.9%,*** the groups by early or no insulin dependence showed that the group with insulin had a higher frequency of antibody positivity:anti-GAD65 odds ratio(OR):2.42(1.112–5.303,P=0.026);anti-IA2:OR:1.55(0.859–2.818,P=0.105);and anti-Zn8T:OR:7.32(2.039–26.279,P=0.002).CONClUSION Anti-GAD65 positivity was high in our ***-GAD65 and anti-Zn8T positivity showed a significantly depleted beta cell reserve phenotype,leading to an increased risk of early insulin dependence.

Background:Skeletal muscle energetics decline with age,and physical activity(PA)has been shown to offset these declines in older ***,many studies reporting these effects were based on self-reported PA or structured exercise ***,we examined the associations of accelerometry-measured and self-reported PA and sedentary behavior(SB)with skeletal muscle energetics and explored the extent to which PA and sedentary behavior would attenuate the associations of age with muscle ***:As part of the Study of Muscle,Mobility and Aging,enrolled older adults(n=879),810(age=76.4±5.0 years old,mean±SD;58%women)had maximal muscle oxidative capacity measured ex vivo via high-resolution re spirometry of permeabilized myofibers(maximal oxidative phosphorylation(maxOXPHOS))and in vivo by ^(31)phosphorus magnetic resonance spectroscopy(maximal adenosine triphosphate(ATP_(max))).Accelerometry-measured sedentary behavior,light activity,and moderate-to-vigorous PA(MVPA)were assessed using a wrist-worn ActiGraph GT9X over 7 ***-reported sedentary behavior,MVPA,and all PA were assessed with the Community Healthy Activities Model Program for Seniors(CHAMPS)*** regression models with progressive covariate adjustments evaluated the associations of sedentary behavior and PA with muscle energetics,as well as the attenuation of the age/muscle energetics association by MVP A and sedentary *** a sensitivity analysis,we also examined activPAl-measured daily step count and time spent in sedentary behavior and their associations with muscle ***:Every 30 min/day more of ActiGraph-measured MVPA was associated with 0.65 pmol/(s×mg)higher maxOXPHOS and 0.012 mM/s higher ATP_(max)after adjusting for age,site/technician,and sex(plight activity was not associated with maxOXPHOS or ATP_(max).Meanwhile,every 30 min/day spent in ActiGraph-measured sedentary behavior was associated with 0.39 pmol/s×mg lower maxOXPHOS and0.

The effect of water deficit was determined on both in vitro and soil seedling as well as in cells in suspension of Agave americana *** order to do the establishment of cells,the formation of callus was induced;for it two auxins were evaluated:2,4-dichlorophenoxyacetic acid(2,4-D)and 4-mino-3,5,6-trichloropicolinic acid(picloram)at three concentrations(0.25,0.5 and 0.75 mg l−1)in three explants(leaf,root and meristems)cultured in MS semisolid *** callogenesis response was related to the type and section of the explant,as well as the regulator used,and a cell suspension was established using 0.5 mg l−1 naphthaleneacetic acid(NAA)+0.5 mg l−1 Benzylaminopurine(BAP).Seedlings were exposed to polyethyleneglycol(15%and 30%w/v)with a water potential of−0.87 and−2.67 MPa,respectively,under soil *** stress was applied through restricted *** weight,root system growth,and chlorophyll concentration were some of the parameters that were affected by the effect of water deficit on *** *** concentration values were significantly decreased by 15 at 30%PEG(19.6 SPAD units)compared to the control *** in vitro plants,the highest concentration of proline was found in the roots,being the treatment with 30%polyethylene glycol where the highest concentration of this osmoregulator was obtained(62.5 mg g−1 DW).Under restricted irrigation conditions,an increase in proline concentration was observed both in the aerial part(2.2μg 100 g−1 DW)and in the root system(1.8μg 100 g−1 DW).However,the concentrations found were approximately ten times greater,less than those found under in vitro ***,the accumulation of proline can be considered an indicator of stress in Agave Americana *** in vitro.

Mineralogical and geochemical studies have been undertaken on the Triassic to lower Jurassic Adigrat Sandstone of the Blue Nile Basin of central Ethiopia to infer its source rock type,paleoweathering,and paleoclimatic *** Adigrat Sandstone occurs at the basal section of the Mesozoic sedimentary formation and unconformably overlays the Neoproterozoic–Paleozoic crystalline rocks,or locally,the Karroo sediments in the northern Blue Nile Basin.A mineralogical study reveals that quartz(Q),feldspars(F),and lithic fragments(l)are the framework grains of the *** the QFl diagram,the plot of the modal composition of the sandstone mainly falls within the feldspathic arenite and quartzose arenite *** geochemical data of the lower section of the sandstone mainly falls within the arkose and subarkose fields,whereas the upper section data falls within the quartzose and sublithic arenite *** and geochemical weathering indices indicate that the provenances of the Adigrat Sandstone were exposed to pronounced weathering intensity,where the lower part of the sandstone was controlled by arid to semi-arid conditions,whereas the upper section was linked to humid to semi-humid(tropical to subtropical)climatic *** and geochemical data also indicate that mafic to intermediate basement rocks were the primary source rocks of the *** the sediment was assumed to have been reworked by multi-cyclic sedimentary *** discriminant function diagram,the REE pattern,la/Th ***/Yb,and the Th–Hf–Co plot are consistent.A comparison of provenance studies for the Adigrat Sandstone in the Blue Nile Basin and the Mekele outlier of northern Ethiopia indicates that the sediment of the former is highly sorted,experienced higher weathering intensity,and compositionally derived from mafic to intermediate crystalline *** the other hand,the sediment of the latter is essentially a weathering product of felsic rocks.

Advances in the management of infertility in men with spinal cord injury

Couples with a spinal cord injured male partner require assisted ejaculation techniques to collect semen that can then be further used in various assisted reproductive technology methods to achieve a pregnancy. The majority of men sustaining a spinal cord injury regardless of the cause or the level of injury cannot ejaculate during sexual intercourse. Only a small minority can ejaculate by masturbation. Penile vibratory stimulation and electroejaculation are the two most common methods used to retrieve sperm. Other techniques such as prostatic massage and the adjunct application of other medications can be used, but the results are inconsistent. Surgical sperm retrieval should be considered as a last resort if all other methods fail. Special attention must be paid to patients with T6 and rostral levels of injury due to the risk of autonomic dysreflexia resulting from stimulation below the level of injury. Bladder preparation should be performed before stimulation if retrograde ejaculation is anticipated. Erectile dysfunction is ubiquitous in the spinal cord injured population but is usually easily managed and does not pose a barrier to semen retrieval in these men. Semen analysis parameters of men with spinal cord injury are unique for this population regardless of the method of retrieval, generally presenting as normal sperm concentration but abnormally low sperm motility and viability. When sperm retrieval is desired in this population, emphasis should be placed on initially trying the simple methods of penile vibratory stimulation or electroejaculation before resorting to more advanced and invasive surgical procedures.

Selective deletion of the soluble Colony-Stimulating Factor 1 isoform in vivo prevents estrogen-deficiency bone loss in mice

Neutralizing CSF1 in vivo completely prevents ovariectomy （OVX）-induced bone loss in mice. There are two isoforms of CSF1, soluble （sCSF1）, and membrane-bound （mCSF1）, but their individual biological functions are unclear. It had been previously reported that mCSF1 knockout （K/O） and wild type （Wt） female mice experience the same degree of bone loss following OVX. In Wt mice the expression of sCSF1 was elevated fourfold in skeletal tissue following OVX while expression of mCSF1 was unchanged. To examine the role of sCSF1 in OVX-induced bone loss, mice were engineered in which sCSF1 was not expressed but expression of mCSF1 was unaffected （sCSF1 K/O）. Isoform-specific reverse transcription PCR confirmed the absence of transcripts for sCSF1 in bone tissue isolated from these animals and no circulating CSF1 was detected by ElISA. Surprisingly, there were no significant differences in bone mineral density （BMD） between sCSF1 K/O mice and Wt controls as assessed by dual-energy X-ray absorptiometry and micro-CT. However, one month after OVX, femoral, spinal and total BMD had declined by 11.2%, 8.9%, and 8.7% respectively in OVX-Wt animals as compared to Sham-OVX. In contrast OVX sCSF1 K/O mice showed changes of ＋0.1%, - 2.4%, and ＋2.3% at the same 3 sites compared to Sham-OVX sCSF1 K/O mice. These data indicate important non-redundant functions for the two isoforms of CSF1 and suggest that sCSF1, but not mCSF1, plays a key role in estrogen-deficiency bone loss.

The objective of this work was to evaluate the effect of Beauveria bassiana(Bb 1205)on controlling Fusarium oxysporum ***(Fol 17108)in tomato plants in greenhouse *** of Bb 1205 was the most promising among the agronomic variables and expression of the activity of the enzymesβ-1,3-glucanases and *** of Bb 1205 occurred at a concentration of 1×108 conidia·ml−1,which was administered onto the leaves,directly into the soil and via *** with Fol 17108 occurred with 1×106 spores·ml−1,which were added directly to the *** was used for measuring agronomic parameters,namely activity of chitinases andβ-1,3-glucanases in foliage and *** Bb 1205 was added to the soil,the chlorophyll index and aerial part length showed significant *** addition,it was determined that root length,fresh weight of foliage,flower,and fruit count increased 82 days after inoculation(dai).Chitinase activity induced by Bb 1205 in leaves and roots of tomato plants infected with Fol 17108 was observed when injected into the stem at 32 dai(41.8 and 11.6-fold,respectively).Inoculation on the foliage showed a 10-fold increase ofβ-1,3-glucanases in the roots after 82 *** for leaves,a 3.8-fold increase was found when the stem was *** the different in vivo applications,Bb 1205 activated its defenses by expressing the chitinase enzymes andβ-1,3-glucanase,thus reducing the damage caused by Fol 17108,demonstrating increase plant growth thereafter.
