Microbial community structure across grazing treatments and environmental gradients in the Serengeti

As one of the last remaining naturally grazed ecosystems on Earth,the Serengeti National Park is an ideal location to study the influence of migratory mammals on the structure of microbial communities and the factors that generate biogeography of soil ***,volcanic inputs generate environmental gradients that may also structure microbial *** studied 16S rRNA amplicons in a 13-year herbivore removal experiment to examine the influence of grazing and environmental gradients on the natural distribution of soil *** of mammalian herbivores shifted microbial community structure,with 31 taxa that were significant indicator taxa of the ungrazed treatment and three taxa that were indicators of the grazed *** abundance of many taxa were correlated with soil texture,phosphorus,iron,calcium and rainfall,and the evenness of taxa within samples was also correlated with these *** general linear mixed effects models with single predictors of multiple,highly correlated variables of beta diversity were consistent with a significant,but weak(2%),effect of grazing,and stronger effects of phosphorus(14%).Beta diversity of microbial communities was greater in grazed than in ungrazed plots;suggesting that the impacts of grazing on community assembly of microbes results from deterministic environmental filtering caused by the influence of herbivores on plant communities and soil properties rather than stochastic dispersal via herds of large *** herbivore effects are superimposed on deterministic environmental filtering by natural soil and precipitation gradients across the Serengeti.

Decoupling of DNA damage response signaling from DNA damages underlies temozolomide resistance in glioblastoma cells

Glioblastoma multiforme （GBM） is the most aggressive primary brain tumor in *** therapy includes surgery,radiation and chemotherapy with temozolomide （TMZ）.Major determinants of clinical response to TMZ include methylation status of the O6-methylguanine-DNA methyltransferase （MGMT） promoter and mismatch repair （MMR） *** the MGMT promoter is methylated in 45% of cases,for the first nine months of follow-up,TMZ does not change survival ***,MMR deficiency makes little contribution to clinical resistance,suggesting that there exist unrecognized mechanisms of *** generated paired GBM cell lines whose resistance was attributed to neither MGMT nor *** show that,responding to TMZ,these cells exhibit a decoupling of DNA damage response （DDR） from ongoing DNA *** display methylation-resistant synthesis in which ongoing DNA synthesis is not *** are also defective in the activation of the S and G2 phase *** proteins ATM,Chk2,MDC1,NBS1 and gammaH2AX also fail to form discrete *** results demonstrate that failure of DDR may play an active role in chemoresistance to *** damages by TMZ are repaired by MMR proteins in a futile,reiterative process,which activates DDR signaling network that ultimately leads to the onset of cell *** cells may survive genetic insults in the absence of *** anticipate that our findings will lead to more studies that seek to further define the role of DDR in ultimately determining the fate of a tumor cell in response to TMZ and other DNA methylators.

Influence of Tea Plantations, Forest and Mixed Farming on Stream Flow and Sediment Loads, Case of Sondu Miriu River Basin, Kenya

The changing patterns of land cover and land use in the tropical river basin over time are critical. The hydrological phenomena at basin and sub basin scale are affected positively or negatively by dynamics of the land cover and land use patterns. Hence identifying causes and driving factors aid in taking appropriate measures to avert the impacts. This study determined the influences of sub basins dominated by tea plantations, forests and agricultural land uses in terms of streamflow and sediment flux variability in Sondu Miriu River Basin in Kenya, East Africa. Field-based investigations were conducted through sampling of flow velocities, turbidity and TSSC obtained from existing River Gauging Stations established within the three sub basins. The sub basin dominated by mixed farming land cover exhibits high turbidity approximately 620 NTU and high levels of total suspended sediment concentration (TSSC) of the order of 630 mg/l in wet seasons. The turbidity levels and TSSC were low in sub basins dominated by forest and tea plantations with approximately mean value of 17 - 29 NTU and 0.019 g/l. The sediment loads in sub basin dominated by mixed farming in the pre planting season in January to February were about 900 tonnes/day higher than that in crop growing season. In sub basins dominated by forest cover and tea plantations, sediment loads were low ranging between 2 - 7 tonnes/day. The relationship between stream flows and area under tea plantations, forests and mixed farming ranged between R2 of 0.025 and 0.16. Tea plantations and forests influence the stream flows and sediment yields in long term duration while in mixed farming variations were observed seasonally. The strong relationships between rainfall and stream flows at the sub basins ranging between R2 of 0.84 and 0.97 revealed the significance of rainfall in hydrologic response of the Sondu Miriu River Basin.

A Machine Learning Approach to Predict HIV Viral Load Hotspots in Kenya Using Real-World Data

Feasibility of pancreatectomy following high-dose proton therapy for unresectable pancreatic cancer

AIM To review surgical outcomes for patients undergoing pancreatectomy after proton therapy with concomitant capecitabine for initially unresectable pancreatic *** From April 2010 to September 2013,15 patients with initially unresectable pancreatic cancer were treated withproton therapy with concomitant capecitabine at 1000 mg orally twice daily. All patients received 59.40 Gy(RBE) to the gross disease and 1 patient received 50.40 Gy(RBE) to high-risk nodal targets. There were no treatment interruptions and no chemotherapy dose reductions. Six patients achieved a radiographic response sufficient to justify surgical exploration,of whom 1 was identified as having intraperitoneal dissemination at the time of surgery and the planned pancreatectomy was aborted. Five patients underwent resection. Procedures included:Laparoscopic standard pancreaticoduodenectomy(n = 3),open pyloris-sparing pancreaticoduodenectomy(n = 1),and open distal pancreatectomy with irreversible electroporation(IRE) of a pancreatic head mass(n = 1). RESULTS The median patient age was 60 years(range,51-67). The median duration of surgery was 419 min(range,290-484),with a median estimated blood loss of 850 cm^3(range,300-2000),median ICU stay of 1 d(range,0-2),and median hospital stay of 10 d(range,5-14). Three patients were re-admitted to a hospital within 30 d after discharge for wound infection(n = 1),delayed gastric emptying(n = 1),and ischemic gastritis(n = 1). Two patients underwent R0 resections and demonstrated minimal residual disease in the final pathology specimen. One patient,after negative pancreatic head biopsies,underwent IRE followed by distal pancreatectomy with no tumor seen in the specimen. Two patients underwent R2 resections. Only 1 patient demonstrated ultimate local progression at the primary site. Median survival for the 5 resected patients was 24 mo(range,10-30).CONCLUSION Pancreatic resection for patients with initially unresectable cancers is feasible after

Helicobacter pylori (H. pylori) infection is well known to be associated with the development of precancerous lesions such as chronic atrophic gastritis (AG), or gastric intestinal metaplasia (GIM), and cancer. Various molecular alterations are identified not only in gastric cancer (GC) but also in precancerous lesions. H. pylori treatment seems to improve AG and GIM, but still remains controversial. In contrast, many studies, including meta-analysis, show that H. pylori eradication reduces GC. Molecular markers detected by genetic and epigenetic alterations related to carcinogenesis reverse following H. pylori eradication. This indicates that these changes may be an important factor in the identification of high risk patients for cancer development. Patients who underwent endoscopic treatment of GC are at high risk for development of metachronous GC. A randomized controlled trial from Japan concluded that prophylactic eradication of H. pylori after endoscopic resection should be used to prevent the development of metachronous GC, but recent retrospective studies did not show the tendency. Patients with precancerous lesions (molecular alterations) that do not reverse after H. pylori treatment, represent the “point of no return” and may be at high risk for the development of GC. Therefore, earlier H. pylori eradication should be considered for preventing GC development prior to the appearance of precancerous lesions.

目的探讨基于美国国家护理联盟(National League for Nursing)杰弗里斯(NLN Jeffries)仿真框架的心力衰竭护理高保真仿真教育对职前护理教育的影响。方法在美国萨克拉门托卡林顿学院进行心力衰竭高仿真模拟教学试点项目。23名学生参加了这项研究。本研究采用类实验设计,在教学前后测量学生的自我效能感、满意度和心力衰竭临床知识。结果结果显示,在高仿真模拟教学实施后,学生的自我效能感平均得分(45.39±7.88比38.91±8.35)、满意度(18.70±3.38比15.57±3.38)及心力衰竭临床知识得分(64.09±10.86比54.48±11.19)均明显提高(P<0.001)。该模拟教学设计中包含护士质量安全教育内容、以患者为中心的护理以及急性心力衰竭患者照护的团队合作,对教学效果产生了积极影响。结论基于NLN Jeffries理论框架的心力衰竭高仿真模拟教学提高了学生的知识、满意度、自我效能感及以患者为中心的护理、安全和团队合作意识。护理教育工作者在进行模拟教学计划时应考虑包含5个特性,即目标、解决问题、为学生提供支持、保真度和总结报告,同时结合安全、以患者为中心和团队合作教育,以保障教学效果。
