Post-cholecystectomy iatrogenic bile duct injuries(IBDIs),are not uncommon and although the frequency of IBDIs vary across the literature,the rates following the procedure of laparoscopic cholecystectomy are much higher than open *** injuries caries a great burden on the patients,physicians and the health care systems and sometime are *** are associated with different manifestations that are not limited to abdominal pain,bile leaks from the surgical drains,peritonitis with fever and sometimes *** injuries if not witnessed during the surgery,can be diagnosed by combining clinical manifestations,biochemical tests and imaging *** such techniques abdominal US is usually the first choice while Magnetic Resonance Cholangio-Pancreatography seems the most *** approach was the ideal approach for such cases,however the introduction of Endoscopic Retrograde Cholangio-Pancreatography(ERCP)was a paradigm shift in the management of such injuries due to accepted success rates,lower cost and lower rates of associated morbidity and ***,the literature lacks consensus for the optimal timing of ERCP intervention in the management of *** management of IBDIs can be tailored according to the nature of the underlying *** the subgroup of patients with complete bile duct ligation and lost ductal continuity,transfer to surgery is indicated without *** patients will not benefit from endoscopy and hence should not do unnecessary *** low–flow leaks *** bed leaks,conservative management for 1-2 wk prior to ERCP is advised,in contrary to high-flow leaks *** duct leaks and stricture lesions in whom early ERCP is *** plus stenting is the ideal management line for cases of *** radiologic techniques are promising options especially for cases of failed endoscopic repair and also for cases with altered *** studies will

Background Changing the distance between a sample and an exposed solid-state nuclear track detector affects the number of alpha tracks recorded by the *** concept had been used to distinguish high-energy alpha particles by computing the alpha emission rate at two distances(0 and 2 cm)from animal bone *** and methods Two Cr-39 detectors were placed in a container with bone ash inside to measure the alpha emission rate when the detectors were in contact with the ash and hung at a distance of 2 cm from the *** The alpha emission rate was found to be 62.7×10^(−4) Bq cm^(−2) when the detector was in contact with the sample(a small exposure area)and 324.4×10^(−4) Bq cm^(−2) when the sample was placed 2 cm away(a larger exposure area).A mathematical equalization of the exposure areas was conducted(the area of the detector exposed to the alpha emitter sample at a distance of 2 cm was equalized to the area exposed when in contact with the sample).After equalization,a reduction in the average value of the alpha emission rate from 324.4×10^(−4) to 17.4×10^(−4) Bq cm^(−2) was *** The increase in distance between the sample and the detector allowed only high-energy alpha particles with a range greater than the traveled distance to reach the ***,this system can distinguish the type and number of nuclides in the sample by changing the distance between the detector and the sample according to each nucleus ***,the results show that the alpha emission rates in these bone samples are higher than the local values.

Preemptive living donor kidney transplantation:Access,fate,and review of the status in

BACKGROUND Preemptive living donor kidney transplantation(PLDKT)is recommended as the optimal treatment for end-stage renal *** To assess the rate of PLDKT among patients who accessed KT in our center and review the status of PLDKT in *** We performed a retrospective review of the patients who accessed KT in our center from November 2015 to November *** addition,the PLDKT status in Egypt was reviewed relative to the *** Of the 304 patients who accessed KT,32 patients(10.5%)had preemptive access to KT(PAKT).The means of age and estimated glomerular filtration rate were 31.7±13 years and 12.8±3.5 mL/min/1.73 m2,***-nine patients had KT,including 3 PLDKTs only(5.1%of total KTs and 9.4%of PAKT).Twenty-nine patients(90.6%)failed to receive PLDKT due to donor unavailability(25%),exclusion(28.6%),regression from donation(3.6%),and patient regression on starting dialysis(39.3%).In multivariate analysis,known primary kidney disease(P=0.002),patient age(P=0.031)and sex(P=0.001)were independent predictors of achievement of KT in our ***,PAKT was not significantly(P=0.065)associated with the achievement of *** of the literature revealed lower rates of PLDKT in Egypt than those in the *** Patient age,sex,and primary kidney disease are independent predictors of achieving living donor *** its non-significant effect,PAKT may enhance the low rates of *** main causes of non-achievement of PLDKT were patient regression on starting regular dialysis and donor unavailability or exclusion.

Study on Binding Affinity of a Glutathione <i>S</i>-Transferase (GST) Fusion Protein to DNA Probe

The aims of this work were to: i) purify GST-fusion protein from bacterial cell extracts of Escherichia coli;ii) quantify the protein by SDS PAGE and Bradford assay;iii) determine protein-DNA interaction of the purified protein by Electrophoretic Mobility Shift Assay. Bacterial culture prepared by inoculation of a single E. coli colony that had a GST fusion protein (gst: six-X10 hd) constructed by ligation of the six-7-hd (X10) sequence into the BamHI and EcoRI sites of the vector pGEX-2T, grown overnight, was sonicated using Cole-Palmer Ultrasonic Homogenizer. Fusion protein was eluted from the beads with Tris-glutathione buffer (50 mM Tris [pH 8.1], 20 mM glutathione), which contained reduced Glutathione. SDS-PAGE was used to calculate the extracted bound protein. Total protein quantification was then estimated by the Bradford assay. Bovine Serum Albumin (BSA) absorbance values were used to plot the standard curve used to calculate the concentrations of the sample proteins. Nylon membrane was used for the electrophoretic transfer;membrane was cross linked and detected by Pierce’s Chemiluminescent Nucleic Acid Detection module. Results showed that X10 gave a strong band shift observed in Lanes 6 and 7 for both 200 ng and 400 ng elute 1 samples;however, there was no shift in the bands for the wild-type, positive control. The concentration of the elute 1 was obtained by the Bradford assay as 242.52 ng/μl and that of elute 2 was 106.30 ng/μl. Similarly, the result obtained by gel analysis was 300 ng/μl (0.3 μg/μl) and 150 ng/μl (0.15 μg/μl) for elutes 1 and 2 respectively.
