Antitumor Effect of Apcin on Endometrial Carcinoma via p21-Mediated Cell Cycle Arrest and Apoptosis

Objective Endometrial carcinoma(EC)is a prevalent gynecological malignancy characterized by increasing incidence and mortality *** underscores the critical need for novel therapeutic *** such potential target is cell division cycle 20(CDC20),which has been implicated in *** study investigated the effect of the CDC20 inhibitor Apcin on EC and elucidated the underlying mechanism *** The effects of Apcin on EC cell proliferation,apoptosis,and the cell cycle were evaluated using CCK8 assays and flow *** sequencing(Rna-seq)was subsequently conducted to explore the underlying molecular mechanism,and Western blotting and coimmunoprecipitation were subsequently performed to validate the *** studies were performed to evaluate the antitumor effects in *** analysis was also conducted to identify CDC20 as a potential therapeutic target in *** Treatment with Apcin inhibited proliferation and induced apoptosis in EC cells,resulting in cell cycle *** associated with apoptosis and the cell cycle were activated following treatment with ***,Apcin treatment led to the upregulation of the cell cycle regulator p21,which was verified to interact with CDC20 and consequently decrease the expression of downstream cyclins in EC *** vivo experiments confirmed that Apcin treatment significantly impeded tumor *** CDC20 expression was observed in EC tissue than in nonmalignant tissue,and increased CDC20 expression in EC patients was associated with shorter overall survival and progress free *** CDC20 is a novel molecular target in EC,and Apcin could be developed as a candidate antitumor drug for EC treatment.

Three-year field study on grass growth and soil hydrological properties in biochar-amended soil

Field monitoring was conducted to investigate and quantify the long-term effects of peanut shell biochar on soil-grass interaction over three *** 10 m5 m grassed plots were constructed in completely decomposed granitic *** of them were amended,respectively,with 5%and 10%biochar contents(m3/m3)for grass growth,while the third was without biochar *** the threeyear monitoring,plant characteristics,saturated water permeability(ks)of grassed soil and soil suction were *** monitored results show that the grass leaf area index(LAI)and root length density(RLD)with biochar amendment were improved by 38%and 200%,*** the grassed plot without biochar,a threshold RLD existed with a value of 1.7 cm/cm3,beyond which ks raised *** threshold RLD increased by 52%when biochar content increased from 0%to 10%.This implies that biochar may restrict the increase in ks of grassed soil due to the rise in the threshold *** presence of biochar and grass can retain over 100%higher suction after heavy rainfalls,while 54%lower peak suction under evapotranspiration(ET)compared with the non-amended *** can alleviate the negative effects on hydraulic properties caused by plant growth and reduce ET-induced excessive water loss.A 5%peanut shell biochar content is recommended for the long-term management of vegetated earthen infrastructures.

Cellular senescence is the results of aging and age-related diseases,and the development of anti-aging methods may improve health and extend *** natural flavonol fisetin has been shown to antagonize senescence in vitro and increases longevity in vivo,but has poor water solubility and limited *** this study,a food-grade and senescent cell-targeted delivery system for fisetin was developed based on whey protein isolate-galactooligosaccharides(WPI-GOS)Maillard conjugate,which could recognize senescence associatedβ-galactosidase in senescent *** fisetin nanoparticles possessed a high encapsulation efficiency,excellent dispersibility in water,good storage stability and well ***,they could effectively accumulate and retain in senescent cells with excellent senescent cell-targeting efficacy,and inhibit the oxidative stress-induced cellular senescence in ***,this novel nanoparticle system based on WPI-GOS Maillard conjugate showed promise to deliver hydrophobic bioactive ingredients like fisetin to senescent cells to improve their bioavailability and anti-senescence effect.

Multi-Fidelity Simulation of Gas Turbine Overall Performance by Directly Coupling High-Fidelity Models of Multiple Rotating Components

Multi-fidelity simulations incorporate computational fluid dynamics(CFD) models into a thermodynamic model,enabling the simulation of the overall performance of an entire gas turbine with high-fidelity *** iterative coupled methods rely on characteristic maps,while fully coupled methods directly incorporate high-fidelity ***,fully coupled methods face challenges in simulating rotating components,including weak convergence and complex *** address these challenges,a fully coupled method with logarithmic transformations was developed to directly integrate high-fidelity CFD models of multiple rotating *** developed fully coupled method was then applied to evaluate the overall performance of a KJ66 micro gas turbine across various off-design *** developed fully coupled method was also compared with the traditional iterative coupled ***,experimental data from ground tests were conducted to verify its *** convergence history indicated that the proposed fully coupled method exhibited stable convergence,even under far-off-design *** experimental verification demonstrated that the multi-fidelity simulation with the fully coupled method achieved high accuracy in off-design *** analysis revealed inherent differences in the coupling methods of CFD models between the developed fully coupled and traditional iterative coupled *** inherent differences provide valuable insights for reducing errors between the component-level model and CFD models in different coupling *** developed fully coupled method,introducing logarithmic transformations,offers more realistic support for the detailed and optimal design of high-fidelity rotating components within the overall performance platform of gas turbines.

A statistical analysis of the Kappa-type energy spectrum distribution of radiation belt electrons observed by Van Allen Probes

The energy spectrum of energetic electrons is a key factor representing the dynamic variations of Earth’s Van Allen radiation *** measurements have indicated that the commonly used Maxwellian and Kappa distributions are inadequate for capturing the realistic spectral distributions of radiation belt *** we adopt the Kappa-type(KT)distribution as the fitting function and perform a statistical analysis to investigate the radiation belt electron flux spectra observed by the Van Allen *** calculating the optimal values of the key KT distribution parameters(i.e.,κandθ2)from the observed spectral shapes,we fit the radiation belt electron fluxes at different L-shells under different geomagnetic *** this manner,we obtain typical values of the KT distribution parameters,which are statistically feasible for modeling the radiation belt electron flux profiles during either geomagnetically quiet or active periods.A comparison of the KT distribution model results with those using the Maxwellian or Kappa distribution reveals the advantage of the KT distribution for studying the overall properties of the radiation belt electron spectral distribution,which has important implications for deepening the current understanding of the radiation belt electron dynamics under evolving geomagnetic conditions.

Cascaded ELM-Based Joint Frame Synchronization and Channel Estimation over Rician Fading Channel with Hardware Imperfections

Due to the interdependency of frame synchronization(FS)and channel estimation(CE),joint FS and CE(JFSCE)schemes are proposed to enhance their functionalities and therefore boost the overall performance of wireless communication *** traditional JFSCE schemes alleviate the influence between FS and CE,they show deficiencies in dealing with hardware imperfection(HI)and deterministic line-of-sight(LOS)*** tackle this challenge,we proposed a cascaded ELM-based JFSCE to alleviate the influence of HI in the scenario of the Rician fading ***,the conventional JFSCE method is first employed to extract the initial features,and thus forms the non-Neural Network(NN)solutions for FS and CE,***,the ELMbased networks,named FS-NET and CE-NET,are cascaded to capture the NN solutions of FS and *** and analysis results show that,compared with the conventional JFSCE methods,the proposed cascaded ELM-based JFSCE significantly reduces the error probability of FS and the normalized mean square error(NMSE)of CE,even against the impacts of parameter variations.

Unveiling the performance of a novel alkalizing bacterium Enterobacter ***-2 in immobilization of available Cd

A novel alkalizing strain Enterobacter ***-2 that could resist 400 mg/L Cd was isolated from Cd-contaminated soil,which immobilized 96.05%Cd^(2+)from *** distribution analysis demonstrated that more than half of the Cd^(2+)was converted into extracellular precipitated Cd through mobilization of the alkali-producing mechanism by the strain LYX-2,achieving the high immobilization efficiency of Cd^(2+).Biosorption experiments revealed that strain LYX-2 had superior biosorption capacity of 48.28 mg/g for *** experiments with Brassica rapa *** performed with and without strain *** to control,15.92%bioavailable Cd was converted to non-bioavailable Cd and Cd content in aboveground vegetables was decreased by 37.10%with addition of strain *** Cd was mainly immobilized through extracellular precipitation,cell-surface biosorption and intracellular accumulation of strain LYX-2,which was investigated through Cd distribution,Scanning Electron Microscope and Energy-Dispersive X-ray Spectroscopy(SEM-EDS),Fourier Transform Infrared Spectroscopy(FT-IR),X-ray Photoelectron Spectroscopy(XPS)and Transmission Electron Microscopy(TEM)*** addition,the application of strain LYX-2 significantly promoted the growth of vegetables about *** results indicated that highly Cd-resistant alkalizing strain LYX-2,as a novel microbial passivator,had excellent ability and reuse value to achieve the remediation of Cd-contaminated soil coupled with safe production of vegetables simultaneously.

Bilayer borophene:an efficient catalyst for hydrogen evolution reaction

The electrocatalytic hydrogen evolution reaction is a crucial technique for green hydrogen ***,finding affordable,stable,and efficient catalyst materials to replace noble metal catalysts remains a significant *** experimental breakthroughs in the synthesis of two-dimensional bilayer borophene provide a theoretical framework for exploring their physical and chemical *** this study,we systematically considered nine types of bilayer borophenes as potential electrocatalysts for the hydrogen evolution *** first-principles calculations revealed that bilayer borophenes exhibit high stability and excellent conductivity,possessing a relatively large specific surface area with abundant active *** surface boron atoms and the bridge sites between two boron atoms can serve as active sites,displaying high activity for the hydrogen evolution ***,the Gibbs free energy change associated with adsorption for these bilayer borophenes can reach as low as−0.002 eV,and the Tafel reaction energy barriers are lower(0.70 eV)than those on ***,the hydrogen evolution reaction activity of these two-dimensional bilayer borophenes can be described by engineering their work ***,we considered the effect of pH on hydrogen evolution reaction activity,with significant activity observed in an acidic *** theoretical results reveal the excellent catalytic performance of two-dimensional bilayer borophenes and provide crucial guidance for the experimental exploration of multilayer boron for various energy applications.
