Background and Aims: Accurate endoscopic detection of premalignant lesions and earlycancers in the colon is essential for cure, since prognosis is closely related to lesion size andstage. Although it has great clinical potential, autofluorescence endoscopy has limited tumorto-normal tissue image contrast for detecting small preneoplastic lesions. We have developed amolecularly specific, near-infrared fluorescent monoclonal antibody (CC49) bioconjugate whichtargets tumor-associated glycoprotein 72 (TAG72), as a contrast agent to improve fluorescencebased endoscopy of colon cancer. Methods: The fluorescent anti-TAG72 conjugate was evaluated in vitro and in vivo in athymic nude mice bearing human colon adenocarcinoma (LS174T)subcutaneous tumors. Autofluorescence, a fluorescent but irrelevant antibody and the free fluorescent dye served as controls. Fluorescent agents were injected intravenously, and in vivowhole body fluorescence imaging was performed at various time points to determine pharmacokinetics, followed by ex vivo tissue analysis by confocal fluorescence microscopy and histology Results: Fluorescence microscopy and histology confirmed specific LS174T cell membrane targeting of labeled CC49 in vitro and ex vivo. In vivo fluorescence imaging demonstrated significant tumor-to-normal tissue contrast enhancement with labeled-CC49 at three hours postinjection, with maximum contrast after 48 h. Accumulation of tumor fluorescence demonstratedthat modification of CC49 antibodies did not alter their specific tumor-localizing properties, andwas antibody-dependent since controls did not produce detectable tumor fluorescence. Conclusions: These results show proof-of-principle that our near-infrared fluorescent-antibody probetargeting a tumor-associated mucin detects colonic tumors at the molecular level in real time,and offer a basis for future improvement of image contrast during clinical fluorescence endoscopy.

Brassinosteroids(BRs)are important regulators that control myriad aspects of plant growth and development,including biotic and abiotic stress responses,such that modulating BR homeostasis and signaling presents abundant opportunities for plant breeding and crop *** and other proteins involved in the biosynthesis and signaling of BRs are well understood from molecular genetics and phenotypic analysis in Arabidopsis thaliana;however,knowledge of the molecular functions of these genes in other plant species,especially cereal crop plants,is *** this manuscript,we comprehensively review functional studies of BR genes in Arabidopsis,maize,rice,Setaria,Brachypodium,and soybean to identify conserved and diversified functions across plant species and to highlight cases for which additional research is in *** performed phylogenetic analysis of gene families involved in the biosynthesis and signaling of BRs and re-analyzed publicly available transcriptomic *** trees coupled with expression data provide a valuable guide to supplement future research on BRs in these important crop species,enabling researchers to identify gene-editing targets for BR-related functional studies.

Projected Changes in extreme event Indices for Alaska

As climate has warmed in recent decades, Alaska has experienced a variety of high-impact extreme events that include heat waves, wildfires, coastal storms and freezing rain. Because the warming is projected to continue, it is essential to consider future changes when planning adaptation actions and building resilience. In this study, we synthesize information on future changes in extreme events in Alaska from an ensemble of regional climate model simulations performed as part of Arctic-CORDeX (Coordinated Regional Climate Downscaling experiment). A set of 13 extreme event indices, based on those developed by the World Climate Research Programme’s expert Team on Climate Change Detection and Indices (eTCCDI), are evaluated from the Arctic-CORDeX output for Alaska. Of the 13 indices, six pertain to temperature, five to total precipitation, one to wind and one to snow. The results for locations in seven different climate zones of Alaska include large increases (5˚C - 10˚C) in the temperature thresholds for the five hottest and coldest days of the year, and large increases in warm spell duration and decreases in cold spell duration. Changes in the cold day temperature threshold are generally larger than the changes in the hot day temperature threshold, consistent with the projections of a stronger warming in winter than in summer in Alaska yearly maximum 1-day and 5-day precipitation amounts as well as the yearly number of consecutive wet days are projected to increase at all locations. The indices for heavy snow days and high-wind days show mixed changes, although the results indicate increases in heavy snow days at the more northern locations and increases in windy days at coastal locations. The changes in the extreme event indices continue through 2100 under the higher-emission (RCP 8.5) emission scenario, while the changes generally stabilize under the lower-emission (RCP 4.5) scenario. .

Clinical characteristics of young patients with early Barrett’s neoplasia

BACKGROUND esophageal adenocarcinoma(eAC)and high-grade dysplasia(HGD)may appear in young patients with Barrett's esophagus(Be).However,characteristics of Barrett's-related neoplasia in this younger population remain *** To identify clinical characteristics that differ between young and old patients with early-stage Barrett's-related *** We conducted a retrospective analysis of a prospectively maintained database comprised of consecutive patients with early-stage eAC(pT1)and HGD at a tertiary-referral center between 2001 and *** characteristics,drug and risk factor exposures,clinicopathological staging of eAC/HGD and treatment outcomes[complete eradication of neoplasia(Ce-N),complete eradication of intestinal metaplasia(Ce-IM),recurrence of neoplasia and recurrence of intestinal metaplasia]were *** analyses were performed to identify factors that differed significantly between older and younger(≤50 years)*** We identified 450 patients with T1 eAC and HGD(74%and 26%,respectively);45(10%)were≤50 *** to the older group,young patients were more likely to present with ongoing gastroesophageal reflux disease(GeRD)symptoms(55%vs 38%,P=0.04)and to be obese(body mass index>30,48%vs 32%,P=0.04).Multivariate logistic regression analysis showed that young patients were significantly more likely to have ongoing GeRD symptoms[odds ratio(OR)2.00,95%confidence interval(CI)1.04-3.85,P=0.04]and to be obese(OR 2.06,95%CI 1.07-3.98,P=0.03)whereas the young group was less likely to have a smoking history(OR 0.39,95%CI 0.20-0.75,Ped to the old ***,there were no significant differences regarding tumor histology,Ce-N,Ce-IM,recurrence of neoplasia and recurrence of intestinal metaplasia(mean follow-up,44.3 mo).CONCLUSION While guidelines recommend Be screening in patients>50 years of age,younger patients should be considered for screening endoscopy if they suffer from obesity and GeRD symptoms.

Leishmaniasis is a set of diseases with a worldwide distribution that affects mainly economically underprivileged populations in developing countries. It has a major impact on public health, with a global cost of billions of dollars per year. The treatment and control of leishmaniasis vary according to the Leishmania species involved, which require reliable methods for species identification. Since most of the currently used methods have limitations, there is a need for assays that allow rapid, precise identification of the offending species. Real-time polymerase chain reactions in conjunction with dissociation curve analysis have been used to detect differences in the DNA composition of selected genes of Leishmania spp. Kinetoplast DNA is the main molecular target used because of its high copy number per parasite, but other targets have also been studied. As part of an effort to establish melting temperature standards for each target gene, we have reviewed the pertinent literature available in public databases, including Pub Med, Web of Science, Sci eLO and LILACS, using the keywords "Leishmania", "leishmaniasis", "realtime PCR", "melting temperature", and "melting curve", alone or in combination. After applying eligibility criteria, 27 articles were selected for analysis. A considerable variation in the methodologies analyzed was found regarding molecular targets, standardization of the methods, reproducibility and specificity. Because of this, statistical analysis was not performed. In most cases, the methods were able to differentiate the parasite at the subgenus level or few species regardless of the target chosen.

Transitioning from paternalism to empowerment of farmers in lowincome countries: Farming components to systems

Reasons for the initiation of farming systems/participatory approaches in non-Green Revolution（i.e., in more heterogeneous and less favourable production environments） areas in low-income countries is followed by a discussion of their *** phases are described along, with a brief description of some of the significant methods in which farmer participation has been sought, accessed and *** enabled/facilitated the evolutionary ***, over the last 40 years there has been a progress in making small-scale farmers much more visible in driving the agricultural research/development agenda through using the four basic stages embodied in the farming systems approach（i.e., descriptive, diagnosis, testing/evaluation and finally dissemination）, much still needs to be *** leads on to a discussion as to some of the barriers still inhibiting true farmer empowerment and why further, such empowerment is imperative, if the agricultural challenges of this century are to be addressed successfully, namely substantially increasing agricultural productivity in an ecologically sustainable *** the heterogeneity of the production environments and that many identified improvements are likely to be incremental rather than revolutionary in nature, this will require farmers＇ intimate involvement in their identification, evaluation and *** in the paper recognizes that there is increasing globalization/commercialization of agriculture and is predicated on the need to consider the whole farmer-research-development continuum involving multiple stakeholders（i.e., farmers, scientists, extension workers, input/output service providers and policymakers）.Such a continuum has been explicitly recognized in the operational plans for the recently reformed international agricultural research（i.e., CGIAR） *** paper concludes with a short discussion on the potential role of formal modelling.

A theoretical rigid-plastic analysis for the dynamic shear failure of beams under impulsive loading is presented when using a travelling plastic shear hinge model which tabes into account material strain hardening. The maximum dynamic shear strain and shear strain-rate can be predicted in addition to the permanent transverse deflections and other parameters. The conditions for the three modes of shear failure, i.e., excess deflection failure, excess shear strain failure and adiabatic shear failure are analyzed. The special case of an infinitesimally small plastic zone is discussed and compared with Nonaka's solution for a rigid, perfectly plastic material. The results can also be generalized to examine the dynamic response of fibre-reinforced beams.

Organic Gases in Fluid Inclusions of Ore Minerals and Their Constraints on Ore Genesis:A Case Study of the Changkeng Au-Ag Deposit,Guangdong,China

The newly discovered Changkeng Au-Ag deposit is a new type of sediment-hostedprecious metal deposit. Most of the previous researchers believed that the deposit was formed bymeteoric water convection. By using a high vacuum quadrupole gas mass spectrometric system, ninelight hydrocarbons have been recognized in the fluid inclusions in ore minerals collected from theChangkeng deposit. The hydrocarbons are composed mainly of saturated alkanes C_(1-4) and unsaturatedalkenes C_(2-4) and aromatic hydrocarbons, in which the alkanes are predominant, while the contentsof alkenes and aromatic hydrocarbons are very low. The sum alka/sum alke ratio of most samples ishigher than 100, suggesting that those hydrocarbons are mainly generated by pyrolysis of kerogens insedimentary rocks caused by water-rock interactions at medium-low temperatures, and themetallogenic processes might have not been affected by magmatic activity. A thermodynamiccalculation shows that the light hydrocarbons have reached chemical equilibrium at temperatureshigher than 200 deg C, and they may have been generated in the deep part of sedimentary basins(e.g., the Sanzhou basin) and then be transported by ore-forming fluids to a shallow position of thebasin via a long distance. Most of the organic gases are generated by pyrolysis of the type IIkerogens (kukersite) in sedimentary host rocks, only a few by microorganism activity. Thecompositions and various parameters of light hydrocarbons in gold ores are quite similar to those insilver ores, suggesting that the gold and silver ores may have similar metallogenic *** on the compositions of organic gases in fluid inclusions, the authors infer that the Changkengdeposit may be of a tectonic setting of continental rift.
