Replication of natural surface topographies to generate advanced cell culture substrates

Surface topographies of cell culture substrates can be used to generate in vitro cell culture environments similar to the in vivo cell niches. In vivo, the physical properties of the extracellular matrix (ECM), such as its topography, provide physical cues that play an important role in modulating cell function. Mimicking these properties remains a challenge to provide in vitro realistic environments for cells. Artificially generated substrates’ topographies were used extensively to explore this important surface cue. More recently, the replication of natural surface topographies has been enabling to exploration of characteristics such as hierarchy and size scales relevant for cells as advanced biomimetic substrates. These substrates offer more realistic and mimetic environments regarding the topographies found in vivo. This review will highlight the use of natural surface topographies as a template to generate substrates for in-vitro cell culture. This review starts with an analysis of the main cell functions that can be regulated by the substrate’s surface topography through cell-substrate interactions. Then, we will discuss research works wherein substrates for cell biology decorated with natural surface topographies were used and investigated regarding their influence on cellular performance. At the end of this review, we will highlight the advantages and challenges of the use of natural surface topographies as a template for the generation of advanced substrates for cell culture.

An intraoperative diagnosis is critical for precise cancer surgery. However, traditional intraoperative assessments based on hematoxylin and eosin(H&E) histology, such as frozen section, are time-,resource-, and labor-intensive, and involve specimen-consuming concerns. Here, we report a near-real-time automated cancer diagnosis workflow for breast cancer that combines dynamic full-field optical coherence tomography(D-FFoCT), a label-free optical imaging method, and deep learning for bedside tumor diagnosis during surgery. To classify the benign and malignant breast tissues, we conducted a prospective cohort trial. In the modeling group(n = 182), D-FFoCT images were captured from April 26 to June 20, 2018, encompassing 48 benign lesions, 114 invasive ductal carcinoma(IDC), 10 invasive lobular carcinoma, 4 ductal carcinoma in situ(DCIS), and 6 rare tumors. Deep learning model was built up and fine-tuned in 10,357 D-FFoCT patches. Subsequently, from June 22 to August 17, 2018, independent tests(n = 42) were conducted on 10 benign lesions, 29 IDC, 1 DCIS, and 2 rare tumors. The model yielded excellent performance, with an accuracy of 97.62%, sensitivity of 96.88% and specificity of 100%;only one IDC was misclassified. Meanwhile, the acquisition of the D-FFoCT images was non-destructive and did not require any tissue preparation or staining procedures. In the simulated intraoperative margin evaluation procedure, the time required for our novel workflow(approximately 3 min)was significantly shorter than that required for traditional procedures(approximately 30 min). These findings indicate that the combination of D-FFoCT and deep learning algorithms can streamline intraoperative cancer diagnosis independently of traditional pathology laboratory procedures.

Comparison between Care Strategies for Patients with osteoarthritis of the Hands Based on the Use of Joint Protection, Assistive Technology and Exercises

Background: osteoarthritis (oA) is a disabling disease that can affect 6% to 12% of the adult population and more than a third of people over 65 years of age. Purpose: To assess whether a group of people with hand osteoarthritis (hoA) who received different types of treatment improved their function after two years of follow-up. Method: The entire sample (n = 97) underwent three follow-up assessments regarding anthropometric parameters of the upper limbs and ability to perform functional activities. Subsequently, the sample was divided into two groups for the intervention periods, called the First Period (n = 73) and the Second Period (n = 24);the First Period kept the same protocol with orientations, and the Second Period went to an intervention with orientation strength exercises and use of orthosis. Findings: In the separate analysis of the three questions of the DASH pain module, no differences were found between the assessment moments for groups of guidelines, treatment, or symptoms. Significant effects were observed for F(2, 162) = 3.5, p = 0.033, η2 = 0.04, and interaction for moments and intervention F(2, 162) = 4.3, p = 0.016, η2 = 0.05. Implications: It can be concluded that only guidance treatment does not benefit patients with hand osteoarthritis. In contrast, guidance, exercise, and orthosis treatment can significantly improve the disease.

The relationships between climate conditions and wood density in tropical forests are still poorly *** quantify spatial dependence of wood density in the state of Minas Gerais(MG,Brazil),map spatial distribution of density,and correlate density with climate variables,we extracted data from the Forest Inventory of Minas Gerais for 1988 trees scaled throughout the territory and measured wood density of discs removed from the *** variables were extracted from the database of the Ecological-Economic Zoning of Minas *** spatial analysis,tree densities were measured at 44 georeferenced sampling *** data were subjected to exploratory analysis,variography,cross-validation,model selection,and ordinary *** relationships between wood density and environmental variables were calculated using dispersion matrices,linear correlation,and *** density proved to be highly spatially dependent,reaching a correlation of 96%,and was highly continuous over a distance of 228 *** distribution of wood density followed a continuous gradient of 514-659 kg m^(−3),enabling corre-lation with environment *** was correlated with mean annual precipitation(−0.57),temperature(0.63),and evapotranspiration(0.83).Geostatistical methods proved useful in predicting wood density in native tropical forests with different climate *** results confirmed the sensitivity of wood density to climate change,which could affect future carbon stock in forests.

Effects of laying on of hands with and without a spiritual context on pain and functionality of older women with knee osteoarthritis:study protocol for a randomized controlled trial

Background： Laying on of hands （LooH） is a culturally-accepted therapy in several traditions. In Brazil, ＂Spiritism＂（third-largest religious tradition in number of followers） uses LooH with the name of ＂Spiritist Passe＂（SP）. However, there are few studies assessing SP＇s role in medical outcomes. The present study aims to investigate the effects of LooH, with and without a spiritual context, on pain, joint stiffness, and functional capacity of older women （≥60 years old） with knee osteoarthritis （KoA）. Methods/design： In this triple-blind, prospective randomized controlled trial, older women with KoA are assigned to three groups： LooH with a spiritual context group, LooH group without a spiritual context, and a control group receiving no intervention. Patients are assessed by a blinded researcher at baseline, 8 weeks, and 16 weeks. Pain, joint stiffness, and functional capacity are assessed using Western ontario and McMaster Universities osteoarthritis Index. other measures such as anxiety, depression, gait speed, and quality of life will also be assessed. To detect differences between groups, a post-intervention com- parison between the three groups and a mean change （post-pre） comparison among the three groups will be done using analysis of variance. All statistical analyses will be performed using an intention-to-treat approach and a per-protocol analysis. Discussion： A pragmatic design using SP, LooH without spiritual context, and no LooH may provide further scientific evidence on SP＇s feasibility and efficacy for KoA patients.

Seismic performance of composite moment-resisting frames achieved with sustainable CFST members

The main objective of the research presented in this paper is to study the bending behaviour of Concrete Filled Steel Tube （CFST） columns made with Rubberized Concrete （RuC）, and to assess the seismic performance of moment-resisting frames with these structural members. The paper describes an experimental campaign where a total of 36 specimens were tested, resorting to a novel testing setup, aimed at reducing both the preparation time and cost of the test specimens. Different geometrical and material parameters were considered, namely cross-section type, cross-section slenderness, aggregate replacement ratio, axial load level and lateral loading type. The members were tested under both monotonic and cyclic lateral loading, with different levels of applied axial loading. The test results show that the bending behaviour of CFST elements is highly dependent on the steel tube properties and that the type of infill does not have a significant influence on the flexural behaviour of the member. It is also found that Eurocode 4 is conservative in predicting the flexural capacity of the tested specimens. Additionally, it was found that the seismic design of composite moment- resisting frames with CFST columns, according to Eurocode 8, not only leads to lighter design solutions but also to enhanced seismic performance in comparison to steel frames.
