Given.the importan.e of vas.ularization.in.n.rve regen.ration. we hypothes.zed that en.blin. the growth of blood ves.els.from the s.rroun.in. tis.ue in.o the regen.ratin. n.rve within.the tn.C would help elimin.te n.crotic proces.es.an. lead to improved regen.ration. In.this.s.udy, we reported the application.of macros.opic holes.in.o the tubular walls.of s.lk-bas.d tn.Cs.an. compared the various.features.of thes. improved s.lk^(+) tn.Cs.with the tubes.without holes.s.lk^(–) tn.Cs. an. autologous.n.rve tran.plan.s.in.an.8-mm s.iatic n.rve defect in.rats. us.n. a combin.tion.of micro-computed tomography an. his.ological an.lys.s. we were able to prove that the us. of s.lk^(+) tn.Cs.in.uced the growth of blood ves.els.from the adjacen. tis.ue to the in.ralumin.l n.ovas.ular formation. A s.gn.fican.ly higher n.mber of blood ves.els.in.the s.lk^(+) group was.foun. compared with autologous.n.rve tran.plan.s.an. s.lk^(–), accompan.ed by improved axon.regen.ration.at the dis.al coaptation.poin. compared with the s.lk^(–) tn.Cs.at 7 weeks.pos.operatively. In.the 15-mm(critical s.ze) s.iatic n.rve defect model, we again.obs.rved a dis.in.t in.rowth of blood ves.els.through the tubular walls.of s.lk^(+) tn.Cs. but without improved fun.tion.l recovery at 12 weeks.pos.operatively. Our data proves.that macroporous.tn.Cs.in.reas. the vas.ular s.pply of regen.ratin. n.rves.an. facilitate improved axon.l regen.ration.in.a s.ort-defect model but n.t in.a critical-s.ze defect model.
