Apelin.12 improves metabolic an. fun.tion.l recovery of rat heart after global ischemia

This work was design.d to explore efficacy of apelin.12 (A-12) as a cardioprotective agen. when.given.before ischemia or at reperfusion.usin. the isolated workin. heart model. Hearts of male Wistar rats were subjected to 30-min.stabilization.period followed by 35-min.global ischemia an. 30-min.reperfusion. A short-term in.usion.of Krebs-Hen.eleit buffer (KHB) con.tain.n. A-12 (35, 70, 140, 280 or 560 ?M) was ap-plied prior to ischemia (A-12-I) or at on.et of reperfusion.(A-12-R). KHB in.usion.was used as con.rol. A-12 in.usion. in.uced a dose-depen.en. in.rease in.recovery of coron.ry flow, con.ractile an. pump fun.tion.durin. reperfu-sion. with the largest augmen.ation.of these in.ices in.the A-12-I group. Both A-12 groups exhibited a sign.fican. reduction.of LV diastolic pressure rise durin. reperfusion.compared with con.rol. En.an.ed fun.tion.l recovery in.the A-12-I group was combin.d with a decrease in.LDH leakage in.perfusate on.early reperfusion.(by 36% vs. con.rol, p n.usion.of 140 ?M A-12 markedly in.reased myocardial ATP con.en., en.an.ed preservation.of the total aden.n. n.cleotide pool an. improved recovery of the en.rgy charge in.reperfused hearts. There was a tren. towards in.rease in.myocardial phosphocreatin. by the en. of re- perfusion.in.the A-12-I group;however this ben.fit did n.t reach statistical sign.fican.e. At the en. of reperfusion. myocardial lactate an. lactate/pyruvate ratio were on.average 5-fold lower in.A-12-I treated hearts compared with con.rol on.s an. did n.t differ sign.fican.ly from the in.tial values. Therefore, improved cardiac dysfun.tion.after I/R in.ury an. less cell mem-bran. damage in.uced by A-12 are associated with main.ain.n. high en.rgy phosphates, particularly ATP, in.reperfused myocardium. Chan.es in.en.rgy metabolism may play a role in.mechan.sms of cardioprotection.afforded by A-12 durin. I/R stress.

Backgroun.: An.lysis of Philadelphia (Ph) chromosome, a hallmark of chron.c myeloid leukemia (CML) plays an.importan. role in.disease mon.torin. of the targeted drug Imatin.b. Apart from Ph, gen.mic imbalan.es such as addition.l chromosomal abn.rmalities (ACAs) of major route occur durin. tran.formation.of the disease an. show n.gative impact on.progn.sis. Objective: The presen. study was carried out to in.estigate frequen.ies of ACAs, gen.mic deletion., complex Ph varian.s an. their progn.stic in.luen.es in.a large cohort of n.wly diagn.sed CML-CP (chron.c phase) an. CML-AP/BP (accelerated/blast phase). Material & Methods: Retrospective, sin.le in.titution.l study on.1367 cases of CML-CP an. 82 cases of CML-AP/BP between.2009 an. 2015, usin. con.en.ion.l cytogen.tics alon. with fluorescen.e in.situ hybridization. Results: Of the 1367 patien.s in.CML-CP, 1041 patien.s who completed 12 - 18 mon.hs of Imatin.b therapy showed complete cytogen.tic remission.(CCyR) rates of 76% an. 82% at 12 an. 18 mon.hs respectively. Imatin.b in.uced 81% an. 33% CCyR in.CML-AP an. CML-BP respectively. Frequen.ies of ACAs in.CML-CP, AP an. BP were 2%, 27% an. 67% respectively. Patien.s in.chron.c an. AP/BP phase with ACAs showed resistan.e to Imatin.b (p n.iden.e of gen.mic deletion. an. complex Ph varian.s was 21% an. 6.3% respectively with n. comparable differen.e of cytogen.tic respon.e to Imatin.b (p p n.a cohort of 112 patien.s in.CCyR, developmen. of n.w clon.l abn.rmalities, more frequen.ly trisomy 8 was detected in.Ph n.gative clon.. Con.lusion. Our data demon.trated that Imatin.b as a fron.lin. therapy had sign.fican.ly improved man.gemen. of CML. However, ACAs play an.importan. role in.resistan.e to Imatin.b, both in.chron.c an. acute phase, which may limit sole ABL targeted therapy.

Several software reliability growth models （SRGM） have been.developed to mon.tor the reliability growth durin. the testin. phase of software developmen.. In.most of the existin. research available in.the literatures, it is con.idered that a similar testin. effort is required on.each debuggin. effort. However, in.practice, differen. types of faults may require differen. amoun.s of testin. efforts for their detection.an. removal. Con.equen.ly, faults are classified in.o three categories on.the basis of severity： simple, hard an. complex. This categorization.may be exten.ed to r type of faults on.the basis of severity. Although some existin. research in.the literatures has in.orporated this con.ept that fault removal rate （FRR） is differen. for differen. types of faults, they assume that the FRR remain. con.tan. durin. the overall testin. period. On.the con.rary, it has been.observed that as testin. progresses, FRR chan.es due to chan.in. testin. strategy, skill, en.iron.en. an. person.el resources. In.this paper, a gen.ral discrete SRGM is proposed for errors of differen. severity in.software systems usin. the chan.e-poin. con.ept. Then. the models are formulated for two particular en.iron.en.s. The models were validated on.two real-life data sets. The results show better fit an. wider applicability of the proposed models as to differen. types of failure datasets.

An.exten.ive theoretical set of atomic data for Rb XXIX in.a wide ran.e with L-shell electron.excitation. to the M-shell has been.reported. We have computed en.rgy levels for the lowest 113 fin. structure levels of Rb XXIX. The fully relativistic multicon.iguration.l Dirac-Fock method （MCDF） within.the framework of Dirac-Coulomb Hamilton.an.takin. quan.um electrodyn.mics （QED） an. Breit correction. in.o accoun. has been.adopted for calculation.. Radiative data are reported for electric dipole （El）, magn.tic dipole （MI）, electric quadrupole （E2）, an. magn.tic quadrupole （M2） tran.ition. from the groun. level, although calculation. have been.performed for a much larger n.mber of levels. To assess the accuracy of results, we performed an.logous calculation. usin. flexible atomic code （FAC）. Comparison. are made with existin. available results an. a good agreemen. has been.achieved. Most of the wavelen.ths calculated lie in.the soft x-ray （SXR） region. Lifetimes for all 113 levels have also been.provided for the first time. Addition.lly, we have provided the spectra for allowed tran.ition. from n.= 2 to n.= 3 within.the x-ray region.an. also compared our SXR photon.wavelen.ths with experimen.ally recogn.zed wavelen.ths. 目的我们在COS-1细胞的瞬时转染表达研究已发现大鼠肠道细胞膜上粘蛋白Muc3的羧基端区域在GSIVV的丝氨酸和甘氨酸之间被蛋白酶酶切,该酶切反应发生在内质网。细胞外的酶切片段与细胞膜上的片段通过非共价键连接在一起。方法和结果本研究表明该酶切反应不仅依赖G/SIVV位点的存在(G/S代表甘氨酸和丝氨酸的酶切点),也依赖SEA(sea—urchin.子蛋白,肠激酶和agrin.组件的更远的羧基段序列;被转染的细胞用tun.camycin.制n.糖基化不能破坏两个酶切片段的再连接,尽管蛋白酶切被部分阻断,一些非糖基化及非酶切的产物在细胞内聚集出现:Muc3结构域和它的酶切产物在转染细胞的细胞膜出现靶向定位,并且酶切位点突变了的突变体的细胞膜的靶向定位未有影响,消除n.型多聚糖的Muc3的细胞膜的靶向定位也未见影响。
