In recent years,multiple advances have been made in understanding the photosynthetic machinery in model *** transfer to horticultural important fruit crops is challenging and time-consuming due to restrictions in gene editing tools and prolonged life ***,we characterize a gene encoding a PetM domain-containing protein in *** CRISPR/Cas9 knockout lines of the PetM showed impairment in the chloroplastic electron transport rate(ETR),reduced CO_(2) assimilation,and reduction of carotenoids and chlorophylls(Chl)under several light ***,growth-condition-dependent elevation or repression of Chl a/b ratios and de-epoxidation states were identified,underlining possible impairment compensation ***,under low light and glasshouse conditions,there were basal levels in CO_(2) assimilation and ETR,indicating a potential role of the PetM domain in stabilizing the cytochrome b6f complex(Cb6f)under higher light irradiance and increasing its quantum *** suggests a potential evolutionary role in which this domain might stabilize the site of the Cb6f regulating ratios of cyclic and linear electron transport and its potential importance during the conquest of terrestrial ecosystems during which plants were exposed to higher ***,the results are discussed with regard to metabolism and their implication to photosynthesis from an agronomic perspective.

The ion transfer reaction of 2-benzoylpyridine-thiosemicarbazone (hl), which has antimicrobial and antifungal properties and anticancer activity, has been studied to determine its lipophilicity by cyclic voltammetry at the water/1,2-dichloroethane (1,2-DCE) interface. The physicochemical parameters such as standard partition coefficient (lgPI)andthestandardGibbsenergyoftransfer("G0tr,,wI "o)oftheprotonatedformoftheligandwere measured asa function of pH in aqueous phase. The protonated form of the ligand exhibited reversible or quasi-reversible voltammograms at the1,***,pKa1andpKa2,weredeterminedspectrophotometrically and were found to be 12.14 and 3.24, respectively. The standard Gibbs energy of transfer ("G0tr,,wN" o) and the partition coefficient of neutral species (lgPN) were also determined by the shake-flask method. The standard Gibbs energy of transfer of this compound across the water/1,2-DCE interface was evaluated as the quantitative measure of its lipophilicity. The difference between lgPI and lgPN was related to the degree of charge delocalization and was used to evaluate qualitatively the lipophilicity of the ligand.

In order to obtain better anti-cancer compounds, nine glycosylated derivatives were designed and synthe- sized using diosgenin as starting material. Their structures were confn-med by 1H NMR, 13C NMR and MS spectra. The anti-cancer activities of intermediate compounds 5a-5i and the target compounds 6a-6i were investigated against human leukemia HEL, K562, hl60 and melanoma wm9 cell lines via MTT method. The bioassay results show that these derivatives possess good inhibitory activities against the four cancer cell lines. Furthermore, these derivatives show better inhibitory activities against K562 than against other cell lines, and most derivatives show better inhibitory activities than the parental material. Moreover, compounds 6a--6i are more active than their inter- mediates 5a--5i when against these ceils. The above results demonstrate the effects of glycosylation on 3-OH of di- osgenin and acetylation of the sugar moiety on their antitumor activities.

AIM: To identify plasma analytes using metabolomics that correlate with the diagnosis and severity of liver disease in patients with alcoholic hepatitis(AH).METHODS: We prospectively recruited patients with cirrhosis from AH(n = 23) and those with cirrhosis with acute decompensation(AD) from etiologies other than alcohol(n = 25). We used mass spectrometry to identify 29 metabolic compounds in plasma samples from fasted subjects. A receiver operating characteristics analysis was performed to assess the utility of biomarkers in distinguishing acute AH from alcoholic cirrhosis. Logistic regression analysis was performed to build a predictive model for AH based on clinical characteristics. A survival analysis was used to construct Kaplan Meier curves evaluating transplant-free ***: A comparison of model for end-stage liver disease(MELD)-adjusted metabolomics levels between cirrhosis patients who had AD or AH showed that patients with AH had significantly higher levels of betaine, and lower creatinine, phenylalanine, homocitrulline, citrulline, tyrosine, octenoyl-carnitine, and symmetric dimethylarginine. When considering combined levels, betaine and citrulline were highly accurate predictors for differentiation between AH and AD(area under receiver operating characteristics curve = 0.84). The plasma levels of carnitine [0.54(0.18, 0.91); P = 0.005], homocitrulline [0.66(0.34, 0.99); P tanoyl-carnitine [0.53(0.16, 0.90); P = 0.007] correlated with MELD scores in patients diagnosed with AH. Increased levels of many biomarkers(carnitine P = 0.005, butyrobetaine P = 0.32, homocitrulline P = 0.002, leucine P = 0.027, valine P = 0.024, phenylalanine P = 0.037, tyrosine P = 0.012, acetyl-carnitine P = 0.006, propionyl-carnitine P = 0.03, butyryl-carnitine P = 0.03, trimethyl-lisine P = 0.034, pentanoyl-carnitine P = 0.03, hexanoyl-carnitine P = 0.026) were associated with increased mortality in patients with AH. ＜正＞ Adenosine - mediated anti - inflammatory response in the lung involves A2A activation, which may lead to inhibition of Pro - I gene expression. Expression of iNOS,p65 subunit of NF - kappa B （p65） and A2A genes along with NO and Pro- I cytokines were assessed in A2A KO murine model of asthma. KO and WT mice were sensitized according to our published protocol （Fan and mustafa. Pulm Pharmacol Ther. 15:147, 2000）. A day after last challenge, HALF and lungs were collected for Pro - I gene expression. Ragweed （ RW） challenge in sensitized mice increased gene expression of both p65 and i - NOS of WT and KO as compared to the controls （p ＜ 0.01）. A2A expression was down - regulated by RW challenge in WT sensitized mice as compared to controls with no transcripts being detectable in KO. Pro - I cytokines （IL - 2 and IL - 4） and NO levels were also increased in KO challenged mice as compared to WT （p ＜ 0. 01）, with KO and WT having greater NO levels than their controls （p ＜ 0.01）. The data show that A2A down - regulation resulted in higher Pro - I gene expression of p65 leading to increased expression of i - NOS, NO and Pro - I cytokines in the lung, implying a role for A2A in Pro - I gene expression in this model.

目的:采用Brdu免疫组化标记技术观察经静脉移植人脐血间充质干细胞进入损伤脊髓组织的表达情况,探讨其促进损伤脊髓神经功能恢复的作用。方法:实验于2003-10/2005-03在苏州大学生物技术研究所干细胞实验室完成。①选取健康成年Wistar大鼠60只,随机数字表法分为假手术组、模型对照组、干细胞移植组,20只/组。脐血标本来自苏州大学附属第一医院产科,产妇及其家属均签署知情同意书。②自脐带抽取新鲜脐带血50～60mL,分离培养人脐血间充质干细胞,调整细胞浓度为5×109L-1待用。在细胞移植前48h,体外进行Brdu标记。③模型对照组、干细胞移植组大鼠采用重物压迫法建立脊髓损伤模型。麻醉后取俯卧位,于T11节段处行背后方正中切口,剥离椎旁肌,咬除T11棘突及椎板,显露4mm×5mm硬脊膜,将55g重砝码放置于硬脊膜表面1min,然后逐层缝合伤口。假手术组仅显露硬脊膜,然后缝合伤口。④造模后5d,干细胞移植组大鼠自后肢大隐静脉缓缓注入人脐血间充质干细胞悬液0.2mL～0.3mL(1×106个)。模型对照组、假手术组给予等量生理盐水。⑤分别于细胞移植后1d、1,2,3周,采用BassoBeatlieBresnahan(BBB)评分法和斜板试验法测定各组神经功能情况。于细胞移植后1,3周制作冰冻切片,检测各组脊髓组织内脐血间充质干细胞的分布情况。结果:①神经功能检测结果:脊髓损伤细胞移植后3周内,模型对照组、干细胞移植组大鼠后肢运动功能均有所恢复,但干细胞移植组恢复较快,于术后1周BBB评分和斜板试验结果即明显优于模型对照组[(10.71±6.24),(5.13±3.36)分,t=2.39,P<0.05;(51.67±5.22),(46.63±3.72)度,t=3.39,P<0.01],并维持至术后3周[(17.29±4.03),(11.25±5.01)分,t=3.89,P<0.01;(65.77±8.06),(57.05±4.61)度,t=4.07,P<0.01]。②脊髓组织中人脐血干细胞的鉴定:假手术组、模型对照组脊髓组织中未见Brdu阳性表达区。干细胞移植组于移植后1周脊髓损伤区可见大量棕褐色Brdu阳性表达的人脐血间充质干细胞存在,并持续至移植后3周,而非损伤区则始终无阳性表达。结论:Brdu对脐血间充质干细胞具有良好的标记作用,经静脉移植的人脐血间充质干细胞能够进入脊髓损伤区,并可以促进损伤脊髓神经功能的修复。
