Assessment of Heritable Variation and Best Combining Genotypes for Grain Yield and Its Attributes in Bread Wheat

Grain yield in wheat is the resultant of several plant attributes. It is very important to assess heritable variation involved in the inheritance of these attributes in addition to find the best combining genotypes. For this purpose, the present study involving 5 × 5 full diallel analysis was performed. Twenty F1 hybrids along with their parents (9797, 9801, 9802, Chakwal-50 and Chakwal-86) were planted in field using randomized complete block design (RCBD) with three replications in the research area of Department of Plant Breeding and Genetics, University of Agriculture, Faisalabad during 2015-2016. Plant characters like plant height, flag leaf area, spike length, No. of fertile tillers per plant, No. of grains per spike, No. of spikelets per spike, 1000 grain weight and grain yield per plant were studied in this experiment. Mean squares due to general combining ability (GCA) were highly significant for all the traits except for spike length for which GCA effects were significant. Mean squares due to specific combining ability (SCA) and reciprocal combining ability (RCA) were highly significant for all the characters studied. GCA variance was higher than the SCA variance for spike length and No. of grains per spike exhibiting the predominant role of additive genetic variation in the inheritance of these traits. However, for the characters like plant height, flag leaf area, No. of fertile tillers per plant, No. of spikelets per spike, 1000 grain weight and grain yield per plant, the value of SCA variance was higher than the value of GCA variance showing non additive gene action for these traits. The variety Chakwal-50 proved to be the best general combiner for plant height, spike length, No. of spikelets per spike, No. of grains per spike and grain yield per plant. The cross, 9802 × Chakwal-86 was the best specific combination for grain yield and most of the yield attributes. Coccidiosis is an endemic problem in broilers inflicting disastrous losses to worldwide poultry *** are generally effective in controlling the infectious diseases;the subunit vaccines effectiveness can hopefully be improved with concurrent use of probiotics as assessed in this experiment using an Eimeria tenella *** mapped protein-1(IMP 1)is a novel immunogenic protein of Apicomplexans,including in Eimeria *** performance of the cumulative effect of probiotics and EtIMP1 C(Part of Eimeria tenella immune mapped protein1) was evaluated based on various parameters such as an intestinal lesion,oocyst scores,feed conversion ratio,and organ *** were also analyzed on both immunological and hematological *** EtIMP1 C and probiotics administered group showed less intestinal lesion,decreased oocyst shedding,satisfactory feed conversion ratio(FCR) and improved hematological parameters as compared to EtIMP1 C emulsified with FCA ***,there was no statistically significant difference between the two groups(P>0.05),in aspect of lesion and oocyst scores,as well as immunological and hematological *** experimental work showed that probiotics could be a good hope due to its antioxidant,immunomodulatory and growth promoting effect against poultry coccidiosis alone or in combination with vaccines,including IMP1 C based vaccine,but further studies are required to formulate its dose with the vaccine as well as a different strain of probiotics effect against coccidiosis.

Dendritic cells(DCs) play a pivotal role to amplify antigen-specific immune *** that sensitize T cells via antigen-presentation by DCs could enhance the capacity of host immunity to fight *** this study,we tested the immunogenic profiles of chicken DCs towards Glyceraldehyde-3-phosphate dehydrogenase from Eimeria acervulina(EaGAPDH).Immunoblot analysis showed that recombinant EaGAPDH(rEaGAPDH) protein was successfully recognized by rat sera generated against *** and internalisation of rEaGAPDH by chicken splenic-derived DCs(chSPDCs) was confirmed by immunofluorescence *** cytometry revealed that chSPDCs upregulated MHCII,CD 1.1,CD11 c,CD80,and CD86 cell-surface ***,mRNA expressions of DC maturation biomarkers(CCL5,CCR7,and CD83) and TLR signalling genes(TLR15 and MyD88) were also upregulated whereas those of Wnt signalling were non-significant compared to negative *** treatment induced IL-12 and IFN-γ secretion in chSPDCs but had no effect on IL-10 and TGF-β.Likewise,DC-T cell co-culture promoted IFN-γ secretion and the level of IL-4 was *** of T cells and their differentiation into CD3+/CD4+T cells were triggered in chSPDCs-T cells co-culture *** together,rEaGAPDH could promote Th1 polarization by activating both host DCs and T cells and sheds new light on the role of this important molecule which might contribute to the development of new DCs-based immunotherapeutic strategies against coccidiosis.

Previously,it was found that several proteins of Haemonchus contortus were involved in the stimulation of the host immune ***,the information about the selection of superlative antigens with immunogenic efficacies on host DCs is *** the current study,the stimulatory effects of five recombinant proteins(elongation factor-1α,arginine kinase,ES-15,ES-24,and ADP-ribosylation factor 1) of *** on the maturation of goat monocyte-derived dendritic cells(md-DCs) were *** proteins were purified separately in *** expression and incubated with isolated goat peripheral blood mononuclear cells(PBMC).Immunofluorescence assay(IFA) results confirmed the binding of these molecules to the md-DC’s surface as compared to control *** the flow cytometry analysis,recombinant proteins induced md-DC stimulation via the up-regulation of the expression of the costimulatory molecule(CD80) and MHC-Ⅱ.Quantitative RT-PCR data showed a significant increase in the expression of specific genes of the WNT and toll-like receptor(TLR) signaling *** result of ELISA indicated the higher levels of cytokine(IL-10,IL-12,IFN-y,and TNF-α) secretion in the md-DC compared to the negative(pET-32 a His-Tag) and blank(PBS)control *** data gives valuable support in the selection of potential antigens for future studies on the immunomodulation of the host against the infection of ***.

纤维素基纳米纤维因其优异的生物相容性和物理化学特性而备受关注,并被广泛应用。该研究介绍了一种简单易行的静电纺丝工艺及其制备得到的纤维素基纳米材料。采用经典的纤维素-铜氨溶液,从而避免使用更复杂的化学溶剂组合。进一步将聚环氧乙烷(polyethylene oxide,PEO)引入二元聚合物体系,改善了静电纺丝过程的稳定性以及最终材料的性能,此过程不涉及有机溶剂。研究了纤维素来源和质量分数以及PEO配方对纺丝性能和纤维形貌的影响,并使用铜氨溶液成功制备了直径为130～382 nm的纤维,X射线光电子能谱(X-ray photoelectron spectroscopy,XPS)分析证实了纳米纤维中铜的存在。铜氨溶液对纳米纤维形态内部由纤维素Ⅰ向纤维素Ⅲ晶体结构的转变存在显著影响。同时,纳米纤维膜体现出对金黄色葡萄球菌和大肠杆菌优异的抗菌特性。

As an important zoonotic protozoan,Toxoplasma gondii(***) has spread around the world,leading to infections in one-third of the *** is still no effective vaccine or medicine against *** recombinant antigens entrapped within nanospheres have benefits over traditional *** the present study,we first expressed and purified *** proteasome subunit alpha type 1(TgPSAl),then encapsulated the recombinant TgPSAl(rTgPSA1) in chitosan nanospheres(CS nanospheres,rTgPSA1/CS nanospheres) and incomplete Freund’s adjuvant(IFA,rTgPSA1/IFA emulsion).Antigens entrapped in CS nanospheres reached an encapsulation efficiency of 67.39%,and rTgPSA1/CS nanospheres showed a more stable release profile compared to rTgPSA1/IFA emulsion in *** vivo,Thl-biased cellular and humoral immune responses were induced in mice and chickens immunized with rTgPSA1/CS nanospheres and rTgPSA1/IFA emulsion,accompanied by promoted production of antibodies,IFN-γ,IL-4,and IL-17,and modulated production of *** with rTgPSA1/CS nanospheres and rTgPSA1/IFA emulsion conferred significant protection,with prolonged survival time in mice and significantly decreased parasite burden in ***,our results also indicate that rTgPSA1/CS nanospheres could be used as a substitute for rTgPSA1/IFA emulsion,with the optimal administration route being intramuscular in mass ***,the results of this study indicate that rTgPSA1/CS nanospheres represent a promising vaccine to protect animals against acute toxoplasmosis.

Neuroprotective Effect of Phyllanthus acidus L. on Learning and Memory Impairment in Scopolamine-Induced Animal Model of Dementia and Oxidative Stress: Natural Wonder for Regulating the Development and Progression of Alzheimer’s Disease

Nature is the best source of complementary and alternative medicine. The plant Phyllanthus acidus (PA) L. has been used traditionally in pain, inflammatory and oxidative stress related disorders. In this consequence, methanolic extract of PA (MEPA) was selected to explore the ability of this plant to enhance cognitive function, brain antioxidant enzymes and anti-acetylcholinesterase activity which can be used for the treatment of oxidative stress related disorders like Alzheimer’s disease (AD). The purpose of this study was to investigate the neuroprotective effect of MEPA on learning and memory impairment in scopolamine-induced rats of dementia and oxidative stress. Treatment with MEPA (i.e., 100 and 200 mg/kg b.w.) was investigated in scopolamine-treated Swiss albino male rats for 14 days and its neuroprotective effects were examined using Elevated Plus Maze (EPM) test, Passive Avoidance (PA) test, Novel Object Recognition (NOR) test, Morris Water Maze (MWM) test as well as level of antioxidant enzymes such as catalase (CAT), super oxide dismutase (SOD), glutathione reductase (GSR), glutathione-S-transferase (GST), reduced glutathione (GSH), glutathione peroxidase (GSH-Px), lipid peroxidation (TBARS) contents and acetylcholinesterase (AChE) activity in rat brain tissue homogenates. Administration of MEPA significantly (P th and 14th day compared to the disease control and control group in the EPM test. Spontaneous coronary dissection presenting with reperfusion arrhythmia: A case report

Spontaneous coronary dissection is more commonly reported in females and is an important differential diagnosis for acute coronary syndrome. Accelerated idioventricular rhythm has been reported before with reperfusion post myocardial ischemia. We report a case of accelerated idioventricular rhythm in a patient with spontaneous coronary artery dissection. A 45-year-old Caucasian female presented with left sided chest pain radiating to the neck and palpitations. Admission ECG showed accelerated idioventricular rhythm. Troponin I peaked at 0.5 ng/ml. Coronary angiography showed mid to distal left anterior descending artery dissection with adequate distal flow. Patient was initially medically managed with aspirin, metoprolol, intravenous heparin and eptifibatide infusions but continued to have symptoms of unstable angina. She underwent successful percutaneous coronary intervention with 2 drug eluting stents and was discharged back home symptom free on dual platelet therapy. Spontaneous coronary artery dissection is an important differential diagnosis for acute coronary syndrome especially in younger females. Accelerated idioventricular rhythm can be a presentation of coronary dissection and may indicate instability. Early percutaneous coronary intervention should be considered in such patients.
