Bioinformatics analysis and experimental validation of cystathionine-gamma-lyase as a potential prognosis biomarker in hepatocellular carcinoma

Background:Hepatocellular carcinoma(HCC)is a common malignant tumor with poor prognosis and high mortality *** cystathionine-gamma-lyase(CSE)plays an important role in the development of multiple tumors,the clinical implication and potential mechanisms of CSE in HCC development remain ***:In our study,the CSE expression in HCC was analyzed in Gene Expression Omnibus(GEO)and The Cancer Genome Atlas(TCGA)datasets and further conﬁrmed by RT-qPCR and immunohistochemistry assays in HCC ***,the associations between CSE expression and HCC malignancy as well as survival were analyzed in GSE14520 and validated in HCC ***,the biological functions of CSE in HCC cells was assessed by CCK-8,ﬂow cytometry and Western ***:Lower transcriptional and proteomic CSE expressions were found in HCC tissues in contrast to adjacent normal *** CSE mRNA expression was signiﬁcantly associated with advanced clinicopathological features and poor outcomes in HCC patients from public database and our *** univariate and multivariate analyses of GSE14520 data showed that CSE expression was an independent prognostic indicator for the overall survival(OS)and recurrence-free survival(RFS)of HCC *** vitro experiments further explained that CSE might trigger HCC cell apoptosis by ***:In summary,the present study identiﬁed the relationship between CSE expression and HCC malignancy as well as OS and RFS,indicating that CSE might be a potential prognostic biomarker and a novel therapeutic target for HCC.

Appendiceal bleeding caused by vascular malformation: A case report

BACKGROUND Acute lower gastrointestinal bleeding(LGIB)is a common occurrence in clinical ***,appendiceal bleeding is an extremely rare condition that can easily be overlooked and *** preoperative detection of appen-diceal bleeding often poses challenges due to the lack of related guidelines and consensus,resulting in controversial treatment *** SUMmaRY We presented a case of a 33-year-old female who complained of hematochezia that had lasted for 1 *** revealed continuous bleeding in the appen-diceal orifice.A laparoscopic appendectomy was performed immediately,and a pulsating blood vessel was observed in the mesangium of the appendix,accor-dingly,active bleeding into the appendicular lumen was *** examination revealed numerous hyperplastic vessels in the appendiceal mucosa and dilated capillary *** The preoperative detection of appendiceal bleeding is often challenging,colo-noscopy is extremely important,bowel preparation is not routinely recommend-ed for patients with acute LGIB or only low-dose bowel preparation is *** appendectomy is the most appropriate treatment for appendiceal bleeding.

Overexpression of auxin/indole-3-acetic acid gene MdIAA24 enhances Glomerella leaf spot resistance in apple(malus domestica)

Auxin is throughout the entire life process of plants and is involved in the crosstalk with other hormones,yet its role in apple disease resistance remains *** this study,we investigated the function of auxin/indole-3-acetic acid(IAA)gene Md IAA24 overexpression in enhancing apple resistance to Glomerella leaf spot(GLS)caused by Colletotrichum fructicola(Cf).Analysis revealed that,upon Cf infection,35S::Md IAA24 plants exhibited enhanced superoxide dismutase(SOD)and peroxidase(POD)activity,as well as a greater amount of glutathione(reduced form)and ascorbic acid accumulation,resulting in less H_(2)O_(2)and superoxide anion(O_(2)^(-))in apple ***,35S::Md IAA24 plants produced more protocatechuic acid,proanthocyanidins B1,proanthocyanidins B2 and chlorogenic acid when infected with *** Cf infection,35S::Md IAA24 plants presented lower levels of IAA and jasmonic acid(ja),but higher levels of salicylic acid(SA),along with the expression of related *** overexpression of Md IAA24 was observed to enhance the activity of chitinase andβ-1,3-glucanase in Cfinfected *** results indicated the ability of Md IAA24 to regulate the crosstalk between IAA,ja and SA,and to improve reactive oxygen species(ROS)scavenging and defense-related enzymes *** jointly contributed to GLS resistance in apple.

In Vitro Propagation and Artificial Seed Production of Fritillaria cirrhosa D. Don, an Endangered Medicinal Plant

Salicylic acid positively regulates maize defenses against lepidopteran insects

In response to insect attack,plants use intricate signaling pathways,including phytohormones,such as jasmonate(ja),ethylene(ET),and salicylic acid(SA),to activate ***(Zea mays)is one of the most important staple food crops around the *** studies have shown that the ja and ET signaling play important roles in maize defense against insects,but little is known about whether and how SA regulates maize resistance to insect *** this study,we ectopically expressed the NahG(salicylate hydroxylase)gene in maize plants(NahG maize)to block the accumulation of *** was found that compared with the wild-type(WT)maize,the NahG-maize exhibited decreased resistance to the generalist insects Spodoptera litura and Spodoptera frugiperda and the specialist Mythimna separata,and the compromised resistance in the NahG maize was associated with decreased levels of defensive metabolites benzoxazinoids(Bxs)and chlorogenic acid(CA).Quantification of simulated *** feedinginduced ja,ja-isoleucine conjugate(ja-Ile),and ET in the WT and NahG maize indicated that SA does not regulate ja or ja-Ile,but positively controls *** provide evidence suggesting that the SA pathway does not crosstalk with the ja or the ET signaling in regulating the accumulation of Bxs and *** analysis revealed that the bHLH,ERF,and WRKY transcription factors might be involved in SAregulated *** study uncovers a novel and important phytohormone pathway in maize defense against lepidopterous larvae.

针对车载导航模式单一、无法根据情境差异动态改变交互策略导致用户体验不足的问题,本文提出了一种支持根据不同情境自适应车载导航交互策略的情境引擎模型。结合以用户为中心的设计流程(UCD)与情境感知理论,定义了汽车人机交互情境,并通过对典型驾驶用户进行跟车调研,提取关键情境要素,阐述了导航情境下情境引擎模型的自适应交互策略及原理,将用户对不同信息的需求程度映射至交互表现层的详细程度。进行了贴合驾驶员不同需求层级的导航HMI信息框架梳理与组件设计,对两个典型导航使用情境进行了交互界面设计与分析。结果表明,情境引擎能对各驾驶情境下的驾驶员需求进行自适应交互,可为智能座舱驾驶交互体验的优化与研发提供借鉴。

To the editor:Allogeneic hemopoietic stem cell transplantation(allo-HSCT)represents the only long-term survival treatment choice for patients with myelodysplastic syndromes(MDS)and MDS/myeloproliferative neoplasm(MPN).Unfortunately,post-hemopoietic stem cell transplantation(HSCT)relapse remains a cause of treatment failure and the results of salvage treatments are *** better conditioning regimens is urgently *** studies have shown synergistic antileukemic effects between decitabine(DEC)and idarubicin(IDA).[1]In an attempt to design a conditioning strategy with very low toxicity but considerable myelosuppressive activity and potential immune-enhancing effects for patients with high-risk MDS and MDS/MPN,we combined DEC and IDA with busulfan,cyclophosphamide,andudarabine for a modied myeloablative regimen in this prospective,multicenter cohort study(hereafter referred to as the“DEC/IDA study”).

基于BERT与生成对抗的民航陆空通话意图挖掘

针对民航陆空通话领域语料难以获取、实体分布不均,以及意图信息提取中实体规范不足且准确率有待提升等问题,为了更好地提取陆空通话意图信息,提出一种融合本体的基于双向转换编码器(bidirectional encoder representations from transformers,BERT)与生成对抗网络(generative adversarial network,GAN)的陆空通话意图信息挖掘方法,并引入航班池信息对提取的部分信息进行校验修正,形成空中交通管制(air traffic control,ATC)系统可理解的结构化信息。首先,使用改进的GAN模型进行陆空通话智能文本生成,可有效进行数据增强,平衡各类实体信息分布并扩充数据集;然后,根据欧洲单一天空空中交通管理项目定义的本体规则进行意图的分类与标注;之后,通过BERT预训练模型生成字向量并解决一词多义问题,利用双向长短时记忆(bidirectional long short-term memory,BiLSTM)网络双向编码提取上下句语义特征,同时将该语义特征送入条件随机场(conditional random field,CRF)模型进行推理预测,学习标签的依赖关系并加以约束,以获取全局最优结果;最后,根据编辑距离(edit distance,ed)算法进行意图信息合理性校验与修正。对比实验结果表明,所提方法的宏平均F_(1)值达到了98.75%,在民航陆空通话数据集上的意图挖掘性能优于其他主流模型,为其加入数字化进程奠定了基础。
