Hypoxia and cytokines regulate carbonic anhydrase 9 expression in hepatocellular carcinoma cells in vitro

AIM: To study the expression of carbonic anhydrase(CA) 9 in human hepatocellular carcinoma(HCC) ***: We studied CA9 protein, CA9 m RNA and hypoxia-inducible factor-1 alpha(HIF-1α) protein levels in Hep3 B cells exposed in different parallel approaches. In one of these approaches, HCC cells were exposed to extreme in vitro hypoxia(24 h 0.1% O2) without or with interleukin(IL)-1, IL-6, tumor necrosis factoralpha(TNF-α) and transforming growth factor-beta(TGF-β) stimulation for the same hypoxic exposure time or exposed to normoxic oxygenation conditions without or with cytokine ***: The tumour cell line analysed showed a strong hypoxic CA9 m RNA expression pattern in response to prolonged severe hypoxia with cell-line specific patterns and a marked induction of CA9 protein in response to severe hypoxia. These results were paralleled by the results for HIF-1α protein under identical oxygenation conditions with a similar expression tendency to that displayed during the CA9 protein expression experimental series. Continuous stimulation with the cytokines, IL-1, IL-6, TNF-α and TGF-β, under normoxic conditions significantly increased the carbonic anhydrase 9 expression level at both the protein and m RNA level, almost doubling the CA9 m RNA and CA9 and HIF-1α protein expression levels found under hypoxia. The findings from these experiments indicated that hypoxia is a positive regulator of CA9 expression in HCC, and the four signal transduction pathways, IL-1, IL-6, TNF-α and TGF-β, positively influence CA9 expression under both normoxic and hypoxic ***: These findings may potentially be considered in the design of anti- cancer therapeutic approaches involving hypoxia-induced or cytokine stimulatory effects on expression. Objective： Over 50% of the world populations are infected with Helicobacter pylori （H. pylori）. Most subjects are asymptomatic; however, in 1994, H. pylori has been categorized as group I carcinogen. The aim of the study was to investigate the relationship between H. pylori infection and gastric cancer. Methods： Thirty gastric cancer patients （GCs） and 30 gastritis patients were enrolled in the study. H. pylori was cultured on non-selective and selective medias, infection density was assessed by quantitative culture. Antibiotic sensitivity testing was performed. PCR was done for the H. pylori 16S rRNA gene in addition to cagA, vacA and iceA genes. Results： H. pylori could be cultured from 100% of specimens obtained from all patients. The density of H. pylori was higher in cancer cases than in gastritis patients. The 16S rRNA was detected in all GC patients （100%） while it was only detected in 70% of gastritis patients. The cagA gene was found in 53.3% vs 13.3% of GC and gastritis patients, respectively. The vacA gene was present in all GC patients （by at least one of its alleles） while it was only found in 33.3% of gastritis patients. The vacA slml combination was the most predominant genotype in GC patients, while m2 was the commonest allele in gastritis patients （10%）. The iceA gene was found in 86.7% vs 40% of GC and gastritis patients, respectively. Simultaneous presence of multiple H. pyiori strains was proved, both phenotypically and genotypically. Conclusion： The development of GC is linked to infection with H. pylori harboring certain virulence genes. Higher infection density of H. pylori was found in GC patients. Co-existence of more than one strain of H. pylori in the same patient occurs in both malignant and benign lesions.

Mankind is facing an unprecedented threat of existence due to the antibiotic resistance developed by bacteria. The unripe fruits of Diospyros malabarica (Desr.) Kostel. (family: Ebenaceae) can be considered as one of the natural sources to tackle this issue. The present study is designed to assess the antimicrobial activity of D. malabarica seed and flesh ex-tracts. Herein, D. malabarica extracts were prepared using polar solvents (i.e., water and 70% ethanol) and their antimicrobial activity as well as in vivo toxicity was investigated. Their antibacterial activity was investigated against gram positive (Bacillus subtilis) and gram negative (Escherichia coli DH5α, and Salmonella typhi) bacteria at different time points. All the extracts showed the highest antibacterial activity after 2 hours of incubation. The aqueous seed extract showed the maximum zone of inhibition (i.e., ~13 mm) against Bacillus subtilis with a minimum inhibitory concentration (MIC) value of 2 μg/μl. The an-tibacterial propensity was also confirmed through trypan blue dye exclusion assay, CellToxTM Green assay, and lipid peroxidation (LPO) assay. On the other hand, the etha-nolic seed extract demonstrated higher antifungal activity through inhibition of mycelial growth. All the extracts showed excellent hemocompatibility against both human and rat red blood cells (RBCs). They also did not show any toxicity to rat liver and kidneys. Taken together, this study demonstrates that the aqueous and ethanolic extracts of D. malabarica seed and flesh could be an effective source of natural antimicrobial agents with no cytotox-ic activity.

Land Use and Land Cover Changes in Arid Region: The Case New Urbanized Zone, Northeast Cairo, Egypt

The spatial characteristics of land cover are useful for understanding the various impacts of human activity on the overall ecological conditions of the urban environment. The multi-temporal Landsat images (TM) between the years of 1990 and 2003 were used together with the Geographic Information System (GIS) techniques to evaluate the environmental changes in the area around Gabal El hamza and the surrounding urban expansion in the new urban cities at the northeast side of the Greater Cairo by using the post classification change detection technique and field investigation. Five major units were determined including: urban, cultivated land, Holocene sand dunes, Oligocene basalt and Miocene–Pleistocene sediments. The cultivated cover changed from 89.6 to 150.4 km2 for the years of 1990 and 2003 respectively. The urban area increased from 49.5 to 120.9 km2 with a great value of change reached 71.3 km2 . The basaltic exposures changed from 3 to 3.75 km2 . The sandy cover decreased from 68.9 to 60.1 km2 and the exposures of the rock units changed from 904.8 to 780.8 km2 with removing 124 km2 in 13 years. The total accuracy of the Landsat-derived land cover data was 95 and 92% for the years 1990 and 2003 respectively. Landsat TM thermal infrared data indicated that the surface temperature was strongly affected by the land cover changes.

Outcome of Renal Transplantation at ahmed Gasim Cardiac and Renal Transplant Centre in Sudan-2014

Background: Although not life-saving procedures like liver or heart transplantation, renal transplantation is the preferred treatment for many patients with end-stage renal disease because it improves patients’ quality of life, while other forms of renal replacement therapy (RRT) such as haemodialysis (HDX) shown to regain only 10% of the patient’s renal function [1]. Transplantation releases patients from the dietary and fluid restrictions and the physical constraints imposed by other forms of (RRT), patients become able to return to their normal activities. Patient’s 5-year survival is higher post transplant. Patients and methods: This is Descriptive, retro-prospective, cross sectional analytic and multicentre study done in ahmed Gasim Cardiac and Renal Transplant Centre including 105 patients. After meeting the Inclusion Criteria and exclusion Criteria, then cases were selected and written in constructed questionnaire. The aim of the study was to evaluate the outcome of renal transplantation in Sudan. Results: 105 cases were used with male predominate and ratio male to female ratio 3:1, the main age of presentation between 31 - 45 years, most of the patients discharge uneventful with good outcome. During one-year follow-up following renal transplantation. Patient survival 95.2 % graft survival 82.4%. Overall morbidity shows 70.5%. Conclusion: this study showed that the one-year patient survival of 95.2%. Actuarial one-year graft survival of 92.4%. Renal transplantation complications can be reduced by meticulous preoperative workup including good cardiovascular assessment, tissue mismatch and BMI good postoperative follow up for early detection of graft rejection and long-term complications and finally patient education.
