Association between low molecular polypeptide 7 single nucleotide polymorphism and response to therapy in hepatitis C virus infection

AIM: To investigate the relationship between low molecular polypeptide-7 (LMP-7) gene polymorphism and response to interferon (IFN) therapy in chronic hepatitis C virus (hCV) patients. METhODS: LMP-7 polymorphism at codon 49 with nucleotide substitution from A to C was amplified in 104 chronic hCV patients of genotype 4. The amplicons were digested with restriction endonuclease Bsm I and the produced restriction fragment length polymorphism was analyzed. Patients received IFN + regional blood volume therapy for 48 wk and the frequency of thissingle nucleotide polymorphism (SNP) was statistically correlated with treatment response. The exclusion criteria for these patients were stated by the national health program for treating viral hepatitis. Main exclusion criteria included co-infection with hepatitis B virus or schistosomiasis, thyroid dysfunction, uncontrolled diabetes mellitus, history of long term drug or alcohol intake and autoimmune hepatitis. Multivariate analyses were done to correlate LMP-7 SNP plus several factors such as age, gender, weight, serum alpha-fetoprotein (AFP) and alanine aminotransferase levels, liver activity, fibrosis score and viral load with response to therapy. RESULTS: The data presented in this study clearly demonstrated statistically significant differences between sustained virological response (SVR) (defined as the absence of hCV RNA levels in the patient's sera at least 6 mo after discontinuation of treatment) and non-response (NR) (where hCV RNA levels in the patient's sera never become undetectable for 6 mo during or after treatment). Variables were described as odds ratio with 95%CI. The data were considered significant if P values were ≤ 0.05; highly significant if P highly significant if P howed that 91.7% of patients carrying LMP-7 C/C allele were associated with SVR, while the other two genotypes C/A and A/A were associated with NR patients, 83.3% and 64.3% respectively, showing that g

Immunotherapy becomes a promising line of treatment for breast cancer(BC)however,its success rate is still ***:The study was designed to optimize the condition for producing an effective dendritic cell(DCs)based immunotherapy by using DCs and T lymphocytes together with tumor-inﬁltrating lymphocytes(TILs)and tumor-inﬁltrating DCs(TIDCs),treated with anti-PD1 and anti-CTLA4 monoclonal *** mixture of immune cells was co-cultured with autologous breast cancer cells(BCCs)isolated from 26 BC ***:There was a signiﬁcant upregulation of CD86 and CD83 on DCs(p=0.001 and 0.017,respectively),similarly upregulation of CD8,CD4 and CD103 on T cells(p=0.031,0.027,and 0.011,respectively).While there was a signiﬁcant downregulation of FOXP3 and combined ***8 expression on regulatory T cells(p=0.014 for both).Increased CD8/Foxp3 ratio(phere was a signiﬁcant increase in interferon-γ(IFN-γ,phydrogenase(LDh,p=0.02),and a signiﬁcant decrease in vascular endothelial growth factor(VEGF,ph ligand 1(PDL-1)were downregulated in BCCs(ph),similarly cytotoxic T lymphocyte antigen-4(CTLA4,p=0.02),Programmed cell death 1(PD-1,ph immune checkpoint inhibitors could produce a potent and effective BC ***,these data should be validated on an experimental animal model to be transferred to the clinical setting.

Mathematical Evaluation Changes in Rheological and Mechanical Properties of Pears during Storage under Variable Conditions

In order to better design, fabricate and control pear handling machine, we should take into account mechanical and rheological properties of pear fruits as related to handling process. The changes in rheological properties of pears stored at 5, 15, 25 ℃ and variable （fluctuating） temperature for 12 days were evaluated in terms of elasticity and viscosity parameters using creep tests. The elasticity and viscosity parameters in creep tests in general decreased with increase in storage time both under constant and variable storage conditions. For the variable storage condition, a bulk mean temperature calculated to account for a series combination of storage time and temperature to which the pears subjected. The changes in rheological properties due to variable storage temperature were described as a function of storage time. The result indicated that except the viscosity parameter of the Maxwell component of the four-element model, it was possible to describe the changes in rheological properties as a function of storage time, which are better physical parameters to estimate the quality of pears.

Objective:To investigate the reproductive performance and milk quality to nutrition pre-pubertal plane in Kurdish female ***: Forty Kurdish female kids [aged (28.0±6.6) d and weighted (7.56±1.10) kg] were assigned randomly in pre-weaning period to one of two practical diets: low quality diet (LQD) [87 g CP/kg dray matter (DM) and 2.02 Mcal ME/kg DM], and high quality diet (hQD), (148 g CP/kg DM and 2.50 Mcal ME/kg DM). At weaning, from each group, one half of kids was separated randomly and allocated to LQD or hQD. Consequently, in post-weaning period, there were four treatment groups including: LQD pre and post-weaning (L-L), control group and LQD pre-weaning and hQD post-weaning (L-h);hQD pre-weaning and LQD post-weaning (h-L), hQD pre- and post-weanin (h-h). From 30 to 180 d of age, body weight and DM intake were determined every 2 ***:Results showed that the hQD treatment enhanced body weight and DM intake during pre-weaning period, in comparison with the LQD treatment (Ph-h treatment had higher DM intake compared with other kid's treatments. Kids fed the hQD treatment had greater withers height compared to kids on the LQD treatment at 90 d of age (Phe L-h and h-h groups weighed more and were younger at puberty. In the period of pre-pubertal, diet plan was not significantly affected milk yield and reproductive performance at the first ***:Overall, management strategies that have been used to availability of nutrition could increase growth and feed intake in Kurdish female kids. In addition, these strategic programs should be enhancing economic characteristics at the start of puberty of kid in goat husbandry.

A study was conducted to investigate the status of the water and sediment quality in the Chalan Beel——a major fresh water fish reservoir of the country for a period of one year from July 2007 to June 2008. The mean values of water quality parameters(depth: 214.73±152.22 cm, temperature 27.68±4.26℃, transparency 123±82 cm, p h 9.7±0.47, total alkalinity 137±42 mg/L, conductivity 307±147 μs/cm, total dissolved solids 183±89 mg/L, ammonia-N 0.27±0.39 mg/L, nitrate-N 0.09±0.07 mg/L, phosphate-P 2.01±2.53 mg/L) and sediment quality parameters(p h 7.21±0.35, organic matter 2.59±0.52%, total nitrogen 0.09±0.04%, available phosphorus 5.4±3.6 Meq./100 g and exchangeable potassium 0.43±0.14 Meq./100 g) were within the range recommended for most of the inland fishes of Bangladesh. Although the water and sediment quality parameters except ammonia and phosphate are in the suitable range, the overall results suggest that better management techniques should be practiced in order to overcome the declining trend of associated aquatic life(fauna and flora) of this important fresh water body of Bangladesh.

Role of Cell Surface Structures in Biofilm Formation by <i>Escherichia coli</i>

This study aims to understand the relationship between capabilities of Escherichia coli strains to form biofilm and serotype groups expressed on cell surface. Sixteen strains of E. coli were originally isolated from different food processing lines in different Moroccan cities. Strains serotyped based on their O (somatic), h (flagellar), and K (capsular) surface antigen profiles using different antiserums. Biofilm assays carried out in 96-well microtiter dishes using the method of O’Toole et al. Our results show that no clear relation observed between origin and serotype groups. In the other hand, we observed that not all studied strains were able to form biofilm. Furthermore, combination of antigens h40 and K11 appears to be involved in biofilm formation. In fact, the h antigen seems to be implicated in the placement of the bacterial cells near the surface and the K antigen may play a role in physicochemical interactions between bacteria and inert surface.

Flow cytometric detection of hepatitis C virus antigens in infected peripheral blood leukocytes: Binding and entry

AIM： We designed two synthetic-core-specific peptides core 1 （C1） and core 2 （C2）, and an E1-specific peptide （El）. We produced specific polyclonal antibodies against these peptides and used the antibodies for detection of hCV antigens on surface and within infected peripheral blood leukocytes. METhODS： Peripheral blood from a healthy individual who tested negative for hCV RNA was incubated with hCV type 4 infected serum for i h and 24 h at 37 ℃. Cells were stained by direct and indirect immunofluorescence and measured by flow cytometry. RESULTS： After 1 h of incubation, antibodies against C1, C2, and El detected hCV antigens on the surface of 27%, 26% and 73% of monocytes respectively, while 10%, 5% and 9% of lymphocytes were positive with anti-C1, anti-C2 and anti-E1 respectively. Only 1-3% of granulocytes showed positive staining with anti-C1, anti-C2 and anti E1 antibodies. After 24 h of incubation, we found no surface staining with anti-C1, anti-C2 or anti-E1. Direct immunostaining using anti-C2 could not detect intracellular hCV antigens, after 1 h of incubation with the virus, while after 24 h of incubation, 28% of infected cells showed positive staining. Only plus strand RNA was detectable intracellularly as early as 1 h after incubation, and remained detectable throughout 48 h post-infection. Interestingly, minus RNA strand could not be detected after 1 h, but became strongly detectable intracellularly after 24 h post-infection. CONCLUSION： Monocytes and lymphocytes are the preferred target cells for hCV infection in peripheral blood leukocytes. Our specific anti-core and anti-E1 antibodies are valuable reagents for demonstration of hCV cell cycle. Also, hCV is capable of infecting and replicating in peripheral blood mononuclear cells as confirmed by detection of minus strand hCV RNA as well as intracellular staining of core hCV antigen.
