Extracellular CIRP dysregulates macrophage bacterial phagocytosis in sepsis

In sepsis, macrophage bacterial phagocytosis is impaired, but the mechanism is not well elucidated. Extracellular cold-inducible RNA-binding protein (eCIRP) is a damage-associated molecular pattern that causes inflammation. However, whether eCIRP regulates macrophage bacterial phagocytosis is unknown. Here, we reported that the bacterial loads in the blood and peritoneal fluid were decreased in CIRP^(−/−) mice and anti-eCIRP Ab-treated mice after sepsis. Increased eCIRP levels were correlated with decreased bacterial clearance in septic mice. CIRP−/− mice showed a marked increase in survival after sepsis. Recombinant murine CIRP (rmCIRP) significantly decreased the phagocytosis of bacteria by macrophages in vivo and in vitro. rmCIRP decreased the protein expression of actin-binding proteins, ARP2, and p-cofilin in macrophages. rmCIRP significantly downregulated the protein expression of βPIX, a Rac1 activator. We further demonstrated that STAT3 and βPIX formed a complex following rmCIRP treatment, preventing βPIX from activating Rac1. We also found that eCIRP-induced STAT3 phosphorylation was required for eCIRP’s action in actin remodeling. Inhibition of STAT3 phosphorylation prevented the formation of the STAT3-βPIX complex, restoring ARP2 and p-cofilin expression and membrane protrusion in rmCIRP-treated macrophages. The STAT3 inhibitor stattic rescued the macrophage phagocytic dysfunction induced by rmCIRP. Thus, we identified a novel mechanism of macrophage phagocytic dysfunction caused by eCIRP, which provides a new therapeutic target to ameliorate sepsis.

The current study examines the significant determinants of electricity consumption and identifies an appropriate model to forecast the electricity price *** main contribution is focused on eastern region of Saudi Arabia,a relatively hottest geographical area full of energy resources but with different electricity consumption *** relative irrelevance of temperature as predicting factor of electricity consumption is quite surprising and contradicts the previous *** the eastern region,electricity price has negative association with electricity *** comparing traditional and machine learning,it is found that machine learning techniques offer better *** the machine learning techniques,the support vector machine has the lowest errors in forecasting the electricity ***,the support vector machine approach is used to forecast the trend of carbon emissions caused by electricity *** findings have policy implications and offer valuable suggestions to policymakers while addressing the determinants of electricity consumption and forecasting electricity prices.

Performance of a three-phase shunt active power filter(SAPF)relies on the capability of the controller to track the reference ***,designing an accurate current controller is crucial to guarantee satisfactory SAPF *** paper presents a model predictive current controller(MPCC)for a low-cost,four-switch,shunt active power filter for power quality improvement.A four-switch,B4,converter topology is adopted as an SAPF,hence offering a simple,robust,and low-cost *** addition,to further reduce overall cost,only two interfacing filter inductors,instead of three,are used to eliminate switching current *** proposed SAPF model MPCC is detailed for implementation,where simulation and experimental results validate effectiveness of the proposed control algorithm showing a 20%improvement in total harmonic distortion compared with a conventional hysteresis band current controller.

For the treatment of MRSA-infected wounds,the spatiotemporally sequential delivery of antibacterial and anti-inflammatory drugs is a promising *** this study,ROS-responsive HA-PBA/PVA(HPA)hydrogel was prepared by phenylborate ester bond cross-linking between hyaluronic acid-grafted 3-amino phenylboronic acid(HA-PBA)and polyvinyl alcohol(PVA)to achieve spatiotemporally controlled release of two kinds of drug to treat MRSA-infected *** hydrophilic antibiotic moxifloxacin(M)was directly loaded in the *** hydrophobic curcumin(Cur)with anti-inflammatory function was first mixed with Pluronic F127(PF)to form Cur-encapsulated PF micelles(Cur-PF),and then loaded into the HPA *** to the different hydrophilic and hydrophobic nature of moxifloxacin and Cur and their different existing forms in the HPA hydrogel,the final HPA/M&Cur-PF hydrogel can achieve different spatiotemporally sequential delivery of the two *** addition,the swelling,degradation,self-healing,antibacterial,anti-inflammatory,antioxidant property,and biocompatibility of hydrogels were ***,in the MRSA-infected mouse skin wound,the hydrogel-treated group showed faster wound closure,less inflammation and more collagen *** experiments further confirmed that the hydrogel promoted better repair by reducing inflammation(TNF-a)and promoting vascular(VEGF)*** conclusion,this HPA/M&CurPF hydrogel that can spatiotemporally sequential deliver antibacterial and anti-inflammatory drugs showed great potential for the repair of MRSA-infected skin wounds.

Blade Wrap Angle Impact on Centrifugal Pump Performance:Entropy Generation and Fluid-Structure Interaction Analysis

The centrifugal pump is a prevalent power equipment widely used in different engineering patterns,and the impeller blade wrap angle significantly impacts its performance.A numerical investigation was conducted to analyze the influence of the blade wrap angle on flow characteristics and energy distribution of a centrifugal pump evaluated as a low specific speed with a value of *** study investigates six impellermodels that possess varying blade wrap angles(95°,105°,115°,125°,135°,and 145°)that were created while maintaining the same volute and other geometrical *** investigation of energy loss was conducted to evaluate the values of total and entropy generation rates(TEG,EGR).The fluid-structure interaction was considered numerically using the software tools ANSYS Fluent and *** elastic structural dynamic equation was used to estimate the structural response,while the shear stress transport k–ωturbulence model was utilized for the fluid domain *** findings suggest that the blade wrap angle has a significant influence on the efficiency of the *** impeller featuring a blade wrap angle of 145°exhibits higher efficiency,with a notable increase of 3.76%relative to the original *** in the blade wrap angle impact the energy loss,shaft power,and pump *** model with a 145°angle exhibited a maximum equivalent stress of 14.8MPa and a total deformation of 0.084 *** results provide valuable insights into the intricate flow mechanism of the centrifugal pump,particularly when considering various blade wrap angles.

The current study attempts to compare the hybrid artificial intelligence models to forecast the environmental externalities in Saudi *** have used the denoising based artificial intelligence models to construct hybrid *** comparing the denoising techniques,the CSD-based denoising has ***,we have used the CSD-based hybrid ***-ANN and CSD-RNN are used for denoising-based artificial intelligence models,whereas CSD-ARIMA is used for denoising-based traditional *** these models are used to check and compare their performance in terms of level and direction of prediction for PM_(10).The results show that the CSD-based ANN model has a higher predictability for PM_(10) levels in Saudi Arabia due to low error values and higher Dstat *** comparing original and forecasted data,the superiority of CSD-ANN is evident in predicting the PM_(10) in Saudi ***,this hybrid model can predict the environmental externalities for non-linear and highly noised ***,the findings can be useful in achieving the sustainable development goal.
