To Analyze the Sensitivity of RT-PCR Assays Employing S Gene Target Failure with Whole Genome Sequencing Data during Third Wave by SARS-CoV-2 Omicron Variant

Introduction: Omicron is a highly divergent variant of concern (VOCs) of a severe acute respiratory syndrome SARS-CoV-2. It carries a high number of mutations in its spike protein hence;it is more transmissible in the community by immune evasion mechanisms. Due to mutation within S gene, most Omicron variants have reported S gene target failure (SGTF) with some commercially available PCR kits. Such diagnostic features can be used as markers to screen Omicron. However, Whole Genome Sequencing (WGS) is the only gold standard approach to confirm novel microorganisms at genetically level as similar mutations can also be found in other variants that are circulating at low frequencies worldwide. This Retrospective study is aimed to assess RT-PCR sensitivity in the detection of S gene target failure in comparison with whole genome sequencing to detect variants of Omicron. Methods: We have analysed retrospective data of SARS-CoV-2 positive RT-PCR samples for S gene target failure (SGTF) with TaqPath COVID-19 RT-PCR Combo Kit (ThermoFisher) and combined with sequencing technologies to study the emerged pattern of SARS-CoV-2 variants during third wave at the tertiary care centre, Surat. Results: From the first day of December 2021 till the end of February 2022, a total of 321,803 diagnostic RT-PCR tests for SARS-CoV-2 were performed, of which 20,566 positive cases were reported at our tertiary care centre with an average cumulative positivity of 6.39% over a period of three months. In the month of December 21 samples characterized by the SGTF (70/129) were suggestive of being infected by the Omicron variant and identified as Omicron (B.1.1.529 lineage) when sequence. In the month of January, we analysed a subset of samples (n = 618) with SGTF (24%) and without SGTF (76%) with Ct values Conclusions: During the COVID-19 pandemic, it took almost more than 15 days to diagnose infection and identify pathogen by sequencing technology. Under optimal process conditions,pyrolysis of polyolefins can yield ca.90 wt%of liquid product,i.e.,combination of light oil fraction and heavier *** this work,the experimental findings reported in a selected group of publications concerning the non-catalytic pyrolysis of polyolefins were collected,reviewed,and compared with the ones obtained in a continuously operated bench-scale pyrolysis *** process parameters were used for the pyrolysis of waste and virgin counterparts of high-density polyethylene,low-density polyethylene,polypropylene and a defined mixture of those(i.e.,25:25:50 wt%,respectively).To mitigate temperature drops and enhance heat transfer,an increased feed intake is employed to create a hot melt plastic *** 1.5 g·min^(-1) feed intake,1.1 L·min^(-1) nitrogen flow rate,and a moderate pyrolysis temperature of 450℃,the formation of light hydrocarbons was favored,while wax formation was limited for polypropylene-rich *** of virgin plastics yielded more liquid(maximum 73.3 wt%)than that of waste plastics(maximum 66 wt%).Blending polyethylenes with polypropylene favored the production of liquids and increased the formation of gasoline-range *** products were mainly composed of C3 hydrocarbons,and no hydrogen production was detected due to moderate pyrolysis temperature.

Changes in the adult sex ratio of six duck species breeding populations over two decades

Outcome of patients with traumatic cranial nerve palsy admitted to a university hospital in Nepal

Background Cranial nerve palsy (CNP) is a common complication of traumatic brain injury (TBI). Despite a high incidence of TBI in Nepal (382 per 100,000), literature on the specific management and outcome of CNP is lacking. This study aimed to examine the outcomes of TBI patients involving single versus multiple *** A retrospective chart review of 170 consecutive TBI patients admitted to the tertiary neurosurgical center in Nepal between April 2020 and April 2022 was conducted. Demographic, clinical, and etiological characteristics; imaging findings; and management strategies were recorded, compared, and analyzed using descriptive statistics. The Glasgow Outcome Scale Extended (GOSE) was used to measure the outcomes in two groups of patients (single and multiple CNP) at 3 *** Out of 250 eligible patients, 80 were excluded and CNP was noted in 29 (17.1%) of the remaining 170. The median age was 34.9 years, and falls (60.6%) were the most common cause of trauma. TBI severity was categorized based on GCS: mild (82.4%), moderate (15.9%), and severe (1.8%). Cranial nerve involvement was seen in 29 (17.05%) patients: single cranial nerve involvement in 26 (89.65%) and multiple nerve involvement in 3 (10.34%). The most common isolated cranial nerve involved was the oculomotor nerve (37.9%). CT findings revealed a maximum of skull fractures with no significant association between CNP and CT *** CNP is a common consequence of TBI with the most common etiology being falls followed by RTA. Single CNP was more common than multiple CNP with no significant difference in the outcome in the 3-month GOSE score. Further research is needed to determine the burden of traumatic CNP and establish specific management guidelines for different types of CNP.

快速城镇化进程中,人口大规模流失引发乡村人地关系失衡,加剧了村庄空心化问题。科学测度乡村空心化、提出以乡村振兴为导向的空心村整治策略,具有重要的现实意义。论文以黑龙江省拜泉县为例,基于人口转移率测度了乡村空心化水平,运用熵权TOPSIS法评价了村庄发展潜力水平,将二者结合识别了村庄发展类型,提出差异化的空心村整治策略。研究结果显示:(1)拜泉县乡村空心化水平均值为0.6529,空心化程度明显。乡镇层面的空心化处于中度和高度水平,村庄以高度和重度空心化为主。农村人口流失主要归因于城乡建设不均衡、农村就业机会少、农村社会保障水平偏低等。(2)村庄发展潜力均值为0.2279,整体水平偏低且空间分布差异明显,县城周边、沿主要交通干线以及主要河流的村庄发展潜力水平普遍高于偏远地区的村庄。产业兴旺(0.5026)水平显著高于乡风文明(0.2703)、生态宜居(0.1472),治理有效(0.0317)、生活富裕(0.0482)水平较低,成为村庄发展潜力的短板。(3)拜泉县村庄类型划分为Ⅰ类——高潜力低空心化、Ⅱ类——高潜力高空心化、Ⅲ类——低潜力低空心化、Ⅳ类——低潜力高空心化4类,整体以Ⅱ类——高潜力高空心化为主,且县域南部村庄发展状态好于北部地区。未来村庄发展应以乡村振兴战略为指引,重心向Ⅰ类、Ⅱ类以及南部村镇转移,并完善宅基地、生态用地、耕地等相关制度体系,促进村庄内部结构调整;培育合作社、家庭农场等新型经营主体,探索特色产业规划与升级道路;构建“中心镇—中心村—农民社区—村庄”的新型村镇体系,加强城乡互动,促进要素聚集、资源共享;同时加大环境整治,增强社会保障,吸引人才回归,实现空心化治理与县域乡村振兴。
