Cotton provides the most abundant natural fiber for the textile *** mature cotton fiber largely consists of secondary cell walls with the highest proportion of cellulose and a small amount of hemicellulose and *** dissect the roles of hemicellulosic polysaccharides during fiber development,four IRREGULAR XYLEM 15(IRX15)genes,GhIRX15-1/-2/-3/-4,were functionally characterized in *** genes encode DUF579 domain-containing proteins,which are homologs of AtIRX15 involved in xylan *** four GhIRX15 genes were predominantly expressed during fiber secondary wall thickening,and the encoded proteins were localized to the Golgi *** GhIRX15 gene could restore the xylan deficient phenotype in the Arabidopsis irx15irx15l double *** of GhIRX15s in cotton resulted in shorter mature fibers with a thinner cell wall and reduced cellulose content as compared to the wild ***,GhIRX15-2 and GhIRX15-4 formed homodimers and *** addition,the GhIRX15s showed physical interaction with glycosyltransferases GhGT43C,GhGT47A and GhGT47B,which are responsible for synthesis of the xylan backbone and reducing end ***,the GhIRX15s can form heterocomplexes with enzymes involved in xylan modification and side chain synthesis,such as GhGUX1/2,GhGXM1/2 and *** findings suggest that GhIRX15s participate in fiber xylan biosynthesis and modulate fiber development via forming large multiprotein complexes.

To Analyze the sensitivity of RT-PCR Assays Employing s Gene Target Failure with Whole Genome sequencing Data during Third Wave by sARs-CoV-2 Omicron Variant

Introduction: Omicron is a highly divergent variant of concern (VOCs) of a severe acute respiratory syndrome sARs-CoV-2. It carries a high number of mutations in its spike protein hence;it is more transmissible in the community by immune evasion mechanisms. Due to mutation within s gene, most Omicron variants have reported s gene target failure (sGTF) with some commercially available PCR kits. such diagnostic features can be used as markers to screen Omicron. However, Whole Genome sequencing (WGs) is the only gold standard approach to confirm novel microorganisms at genetically level as similar mutations can also be found in other variants that are circulating at low frequencies worldwide. This Retrospective study is aimed to assess RT-PCR sensitivity in the detection of s gene target failure in comparison with whole genome sequencing to detect variants of Omicron. Methods: We have analysed retrospective data of sARs-CoV-2 positive RT-PCR samples for s gene target failure (sGTF) with TaqPath COVID-19 RT-PCR Combo Kit (ThermoFisher) and combined with sequencing technologies to study the emerged pattern of sARs-CoV-2 variants during third wave at the tertiary care centre, surat. Results: From the first day of December 2021 till the end of February 2022, a total of 321,803 diagnostic RT-PCR tests for sARs-CoV-2 were performed, of which 20,566 positive cases were reported at our tertiary care centre with an average cumulative positivity of 6.39% over a period of three months. In the month of December 21 samples characterized by the sGTF (70/129) were suggestive of being infected by the Omicron variant and identified as Omicron (B.1.1.529 lineage) when sequence. In the month of January, we analysed a subset of samples (n = 618) with sGTF (24%) and without sGTF (76%) with Ct values Conclusions: During the COVID-19 pandemic, it took almost more than 15 days to diagnose infection and identify pathogen by sequencing technology. Changes in the adult sex ratio of six duck species breeding populations over two decades

Despite all efforts,long-term changes in the adult sex ratios of breeding duck populations are still unclear;this uncertainty is especially true for male-bias populations,which are often under the scrutiny of researchers lacking convenient results for the active protection of endangered *** with male-bias populations are usually strongly affected by a decline in population size that leads to a higher extinction *** this study,we examined our long-term data of the abundance of breeding populations in six duck species(Mallard Anas platyrhynchos,Gadwall Mareca strepera,Red-crested Pochard Netta rufina,Common Pochard Aythya ferina,Tufted Duck Aythya fuligula,and Common Goldeneye Bucephala clangula)from fishponds in south Bohemia,Czechia,between 2004 and *** evidence was used to assess long-term changes in the adult sex ratio in these breeding populations and investigate the possible effects of the NAO index(North Atlantic Oscillation index)on them,indicating climate conditions in *** determined a long-term decrease of the proportion of females in the breeding season in two of the six examined species:Common Pochard and Red-crested Pochard,which is driven by the long-term increase in the number of males in contrast to the decreasing or stable number of females likely caused by different migration behaviours between females and *** the case of Common Pochard,in breeding populations,we estimated 60-65%of males in the early 2000s rising to 75-80%in the early ***,we establish no significant effects linked to climate conditions of the previous winter in these species as a crucial cause of the changes of the proportion of females in the breeding population.

Highly specific interactions between biomolecules,such as antigen-antibody,protein-ligand,or nucleic acid base pair complementary are on the basis of the organization of complex *** same principles may be tentatively used in molecular medicine for diagnosis and therapeutics.A molecule can be designed to selectively bind a protease and thereby inhibit the production of a peptide that forms toxic aggregates in the brain or an antibody may be produced to bind specifically to that peptide for detection or clearance ***,interference in biological systems is not that *** a start there is the inhibition of the physiological role of the protease;moreover,several cleavage fragments may be produced,which may continue to diverge due to putative post-translational modification and self-assembly processes,hiding the toxic target in a“soup”of peptide species varying in size,structure and chemical composition.A perspective of the current status and challenges in targeting peptide species for diagnosis and treatment in the context of Alzheimer’s disease is given.

快速城镇化进程中,人口大规模流失引发乡村人地关系失衡,加剧了村庄空心化问题。科学测度乡村空心化、提出以乡村振兴为导向的空心村整治策略,具有重要的现实意义。论文以黑龙江省拜泉县为例,基于人口转移率测度了乡村空心化水平,运用熵权TOPsIs法评价了村庄发展潜力水平,将二者结合识别了村庄发展类型,提出差异化的空心村整治策略。研究结果显示:(1)拜泉县乡村空心化水平均值为0.6529,空心化程度明显。乡镇层面的空心化处于中度和高度水平,村庄以高度和重度空心化为主。农村人口流失主要归因于城乡建设不均衡、农村就业机会少、农村社会保障水平偏低等。(2)村庄发展潜力均值为0.2279,整体水平偏低且空间分布差异明显,县城周边、沿主要交通干线以及主要河流的村庄发展潜力水平普遍高于偏远地区的村庄。产业兴旺(0.5026)水平显著高于乡风文明(0.2703)、生态宜居(0.1472),治理有效(0.0317)、生活富裕(0.0482)水平较低,成为村庄发展潜力的短板。(3)拜泉县村庄类型划分为Ⅰ类——高潜力低空心化、Ⅱ类——高潜力高空心化、Ⅲ类——低潜力低空心化、Ⅳ类——低潜力高空心化4类,整体以Ⅱ类——高潜力高空心化为主,且县域南部村庄发展状态好于北部地区。未来村庄发展应以乡村振兴战略为指引,重心向Ⅰ类、Ⅱ类以及南部村镇转移,并完善宅基地、生态用地、耕地等相关制度体系,促进村庄内部结构调整;培育合作社、家庭农场等新型经营主体,探索特色产业规划与升级道路;构建“中心镇—中心村—农民社区—村庄”的新型村镇体系,加强城乡互动,促进要素聚集、资源共享;同时加大环境整治,增强社会保障,吸引人才回归,实现空心化治理与县域乡村振兴。

Astrophysical s-factor and reaction rate for 15N(p,γ)16O within the modified potential cluster model

We study radiative p^(15)N capture on the ground state of ^(16)O at stellar energies within the framework of a modified potential cluster model(MPCM)with forbidden states,including low-lying *** investigation of the ^(15)N(p,γ0)^(16)O reaction includes the consideration of ^(3)s_(1) resonances due to E1 transitions and the contribution of the ^(3)P_(1) scattering wave in the p+^(15)N channel due to the ^(3)P_(1)→^(3)P_(0)M1 *** calculated the astrophysical low-energy s-factor,and the extrapolated s(0)turned out to be within 34.7−40.4 keV·*** important role of the asymptotic constant(AC)for the ^(15)N(p,γ0)16O process with interfering ^(3)s_(1)(312)and ^(3)s_(1)(962)resonances is elucidated.A comparison of our calculation for the s-factor with existing experimental and theoretical data is addressed,and a reasonable agreement is *** reaction rate is calculated and compared with the existing *** has negligible dependence on the variation of AC but shows a strong impact of the interference of ^(3)s_(1)(312)and ^(3)s_(1)(962)resonances in reference to the CNO Gamow windows,especially at low *** estimate the contribution of cascade transitions to the reaction rate based on the exclusive experimental data from ***.C.85,065810(2012).The reaction rate enhancement due to the cascade transitions is observed from T_(9)>0.3 and reaches the maximum factor~1.3 at T_(9)=*** present the Gamow energy window and a comparison of rates for radiative proton capture reactions ^(12)N(p,γ)^(13)O,^(13)N(p,γ)^(14)O,^(14)N(p,γ)^(15)O,and ^(15)N(p,γ)^(16)O obtained in the framework of the MPCM and provide the temperature windows,prevalence,and significance of each process.

The autophagy protein Atg9 functions in glia and contributes to parkinsonian symptoms in a Drosophila model of Parkinson’s disease

Parkinson’s disease is a progressive neurodegenerative disease characterized by motor deficits,dopaminergic neuron loss,and brain accumulation ofα-synuclein aggregates called Lewy *** in protein degradation pathways,such as autophagy,has been demonstrated in neurons as a critical mechanism for eliminating protein aggregates in Parkinson’s ***,it is less well understood how protein aggregates are eliminated in glia,the other cell type in the *** the present study,we show that autophagy-related gene 9(Atg9),the only transmembrane protein in the autophagy machinery,is highly expressed in Drosophila glia from adult *** from immunostaining and live cell imaging analysis reveal that a portion of Atg9 localizes to the trans-Golgi network,autophagosomes,and lysosomes in ***9 is persistently in contact with these *** glial atg9 reduces the number of omegasomes and autophagosomes,and impairs autophagic substrate *** suggests that glial Atg9 participates in the early steps of autophagy,and hence the control of autophagic ***,loss of glial atg9 induces parkinsonian symptoms in Drosophila including progressive loss of dopaminergic neurons,locomotion deficits,and glial *** findings identify a functional role of Atg9 in glial autophagy and establish a potential link between glial autophagy and Parkinson’s *** results may provide new insights on the underlying mechanism of Parkinson’s disease.
