Effect of Nonenzyma.ic a.tioxida.ts on Sperm Motility a.d Surviva. Rela.ive to Free Ra.ica.s a.d a.tioxida.t Enzymes of Chilled-Stored Ra. Semen

The effect of inclusion of three a.tioxida.ts (Vita.in E, cysteine a.d gluta.hione) in ra. semen extender on the relea.e of a.tioxida.t enzymes a.d free ra.ica.s wa. studied. a.3 × 3 fa.toria. experiment wa. conducted to test effects of supplementing ra. semen extender with Vita.in E (1, 5 a.d 10 IU), cysteine (1, 5 a.d 10 mM) or gluta.hione (0.5, 1 a.d 2 mM) on the sperm surviva. a.d relea.e of superoxide dismuta.e (SOD), gluta.hione peroxida.e (GPX) a.d thioba.bituric a.id rea.tive substa.ces (TBa.S). Eighty eja.ula.es of eight fertile Na.di ra.s were collected, a.sessed for the gross exa.ina.ion a.d the good eja.ula.es (≥90% motility) were pooled a.d sperm count wa. a.sessed. Therefore ten extenders;control (C) a.d 9 trea.ed (C plus a.tioxida.t) were tested. Extended semen wa. stored a. 5°C for 96 h, exa.ined for motility a.d surviva. a.d sperm cells were sepa.a.ed from pla.ma. sonica.ed, homogenized a.d exposed to the determina.ions of SOD, GPX a.d TBa.S. The highest sperm surviva. wa. found in diluents conta.ning 5 IU Vita.in E/ml, 1 a.d 2 mM gluta.hione (55.5% surviva.), while the lowest surviva. wa. found in 10 mM cysteine (11.1%). TBa.S concentra.ion wa. highest (P a. other trea.ments, however, Vita.in E a.d gluta.hione exhibited low va.ues. Contra.iwise, a.tivities of SOD a.d GPX increa.ed (P a.d semina. pla.ma.in diluents conta.ning 5 IU E, 1 mM a.d 2 mM gluta.hione. The enzyma.ic a.tivities were genera.ly higher in semina. pla.ma.tha. in sperm cells. It wa. concluded tha. supplementing ra. semen extender during chilled stora.e with 5 IU Vita.in E per ml or 1 - 2 mM gluta.hione enha.ced sperm surviva. a.d reduced free ra.ica.s.
