Knockdown of OLA1,a regulator of oxidative stress response,inhibits motility and invasion of breast cancer cells

To explore the role of a novel Obg-like ATPase 1 （OLA1） in cancer metastasis, small interference RNA （siRNA） was used to knockdown the protein, and the cells were subjected to in vitro cell migration and invasion assays. Knockdown of OLA 1 significantly inhibited cell migration and invasion in breast cancer cell line MDA-MB-231. The knockdown caused no changes in cell growth but affected ROs production. In wound-healing assays, decreased ROs in OLA1-knockdown cells were in situ asso- ciated with the cells＇ decreased motile morphology. Further, treatment ofN-acetylcysteine, a general ROs scavenger, blunted the motility and invasiveness of MDA-MB-231 cells, similar to the effect of OLAl-knockdown. These results suggest that knock- down of OLA1 inhibits breast cancer cell migration and invasion through a mechanism that involves the modulation of intracellular ROs levels.

The growing installation of natural gas fired power plants has increased the integration of natural gas and electricity sectors. This has driven the need investigate the interactions among them and to optimize energy resources management from a centralized planning perspective. Thus, a combined modeling of the reservoirs involved in electric power and gas systems and their locations on both networks are essential features to be considered in the operational planning of energy *** paper presents a modeling and optimization approach to the operational planning of electric power and natural gas systems, taking into account different energy storage facilities, such as water reservoirs, natural gas storages and line packs of pipelines. The proposed model takes advantage of captures both energy systems synergy and their associated networks. This approach identifies the interactions between the energy storage facilities and their economic impact over their optimal scheduling. The results show the benefits of an integrated operational planning of electric power and natural gas systems, the close interdependency between the energy resources stored in both systems, and the effects of a combined scheduling.

The growing demand for luminescent nanomaterials intended for various applications increases the necessity to develop and improve approaches to the creation of highly efficient nanosized *** current study,an approach to enhancing the efficiency of red luminescence of monoclinic Y_(2)O_(3):Eu^(3+)with the particle size of~20 nm by creating the oxygen-enriched environment during the gas-phase synthesis was *** investigate the effect of oxygen amount during the synthesis on characteristics of the phosphor,a series of nano structured Y_(2)O_(3):Eu^(3+)with the addition of 0-40 vol%O_(2) to the main buffer gas Ar was synthesized by laser *** is shown that the amount of added O_(2) exerts virtually no effect on the phase composition and particle size,but significantly improves the luminescent characteristics of Y_(2)O_(3):Eu^(3+).Thus,the addition of 30 vol%O_(2) leads to virtually a 20-fold growth in the photoluminescence(PL)intensity caused by ^(5)D_(0)→^(7)F_(0-4) transitions in Eu^(3+)and an increase in the absolute PL quantum yield from 3%to 53%(λex=395 nm)compared to the sample synthesized without O_(2).The addition of oxygen also improves the emission color coordinates from(0.571,0.320)to(0.630,0.322)due to the removal of a considerable fraction of oxygen *** synthesized nanopowders are shown to be highly stable:upon storage under ambient conditions for two *** yield(QY)of the samples decreases by less than 2%.It is expected that the key features underlying the proposed approach will be useful for various methods used to synthesize oxide nanophosphors.

Understanding crash contributing factors is essential in safety management and improvement. These factors drive investment decisions, policies, regulations, and other safety-related initiatives. This paper analyzes factors that contribute to crash occurrence based on two national datasets in the United states (CIss and NAss-CDs) for the years 2017-2022 and 2010-2015, respectively. Three taxonomies were applied to enhance understanding of the various crash contributing factors. These taxonomies were developed based on previous research and practice and involved different groupings of human factors, vehicle factors, and roadway and environmental factors. statistics for grouping the different types of factors and statistics for specific factors are provided. The results indicate that human factors are present in over 95% of crashes, roadway and environmental factors are present in over 45% of crashes, and vehicle factors are present in less than 2% of crashes. Regarding factors related to human error and vehicle maintenance, speeding is involved in over 25% of crashes, distraction is involved in over 20% of crashes, alcohol and drugs are involved in over 9% of crashes, and vehicle maintenance is involved in approximately 0.45% of crashes. Approximately 4.4% of crashes involve a driver who “looked but did not see.” Weather is involved in over 13% of crashes. Conclusions: The findings indicate that, consistent with previous research, human factors or human error are present in around 95% of crashes. Infrastructure and environmental factors contribute to about 45% of crashes. Vehicle factors contribute to only 1.67% - 1.71% of crashes. The results from this study could potentially be used to inform future safety management and improvement activities, including policy-making, regulation development, safe systems and systemic safety approaches to safety management, and other engineering, education, emergency response, enforcement, evaluation, and encouragement activities. The ability to perform against extreme corrosion environments in stainless steel depreciates under challenging conditions in industrial *** can be resolved by employing surface modification techniques to modify the surface with the required *** present study investigates the outcomes of friction stir processing of ss304 with CoCrFeCuTi high entropy alloy(HEA)as reinforcement and the property enhancements *** synthesized CoCrFeCuTi HEA exhibited irregular fragments with 17μm mean particle size and body centred cubic(BCC)*** microstructure of the processed surface revealed ultra-refined grains with a uniform distribution of HEA *** microstructures of the samples annealed at 400℃,550℃,700℃and 850℃for 2 h also showed significant grain growth and grain *** processed sample with HEA reinforcement annealed at 700℃exhibited enhanced microhardness and tensile strength by 63%and 0.88 times than that of the processed sample with HEA *** subsequent electrochemical corrosion test displayed better corrosion resistance for the HEA-processed and annealed *** corrosion tests at 900℃showcased excellent resistance for the HEA processed and annealed samples by 14.92%,12.75%,and 14.83%against the samples pre-annealed under three different salt mixtures,and the corroded surface was analyzed.

Irradiating solids with ultrashort laser pulses is known to initiate femtosecond timescale magnetization ***,sub-femtosecond spin dynamics have not yet been observed or ***,we explore ultrafast light-driven spin dynamics in a highly nonresonant strong-field *** state-of-the-art ab initio calculations,we predict that a nonmagnetic material can transiently transform into a magnetic one via dynamical extremely nonlinear spin-flipping processes,which occur on attosecond timescales and are mediated by cascaded multi-photon and spin–orbit *** are nonperturbative nonresonant analogs to the inverse Faraday effect,allowing the magnetization to evolve in very high harmonics of the laser frequency(*** up to the 42nd,oscillating at~100 attoseconds),and providing control over the speed of magnetization by tuning the laser power and ***,we show that even for linearly polarized driving,where one does not intuitively expect the onset of an induced magnetization,the magnetization transiently oscillates as the system interacts with *** response is enabled by transverse light-driven currents in the solid,and typically occurs on timescales of~500 attoseconds(with the slower femtosecond response suppressed).An experimental setup capable of measuring these dynamics through pump–probe transient absorption spectroscopy is *** results pave the way for attosecond regimes of manipulation of magnetism.

Background:Hepatocellular carcinoma(HCC)is the most common cause of cancer-related death in saudi *** study aimed to investigate the patterns of HCC and the effect of TNM staging,Alfa-fetoprotein(AFP),and Child-Turcotte Pugh(CTP)on patients’overall survival(Os).Methods:A retrospective analysis was conducted on 43 HCC patients at a single oncology center in saudi Arabia from 2015 to *** patients had to fulfill one of the following criteria:(a)a liver lesion reported as definitive HCC on dynamic imaging and/or(b)a biopsy-confirmed ***:The mean patient age of all HCC cases was 66.8 with a male-to-female ratio of 3.3:*** patients were stratified into two groups:viral HCC(n=22,51%)and non-viral HCC(n=21,49%).Among viral-HCC patients,55%were due to HBV and 45%due to *** was diagnosed in 79%of *** and sex did not significantly statistically differ in Os among viral and non-viral HCC patients(p-value>0.05).About 65%of patients had tumor size>5 cm during the diagnosis,with a significant statistical difference in Os(p-value=0.027).AFP was>400 ng/ml in 45%of the *** was a statistically significant difference in the Os in terms of AFP levels(p-value=0.021).A statistically significant difference was also observed between the CTP score and Os(p-value=0.02).CTP class B had the longest *** was the most common treatment provided to HCC patients followed by sorafenib *** was a significant statistical difference in Os among viral and non-viral HCC patients(p-value=0.008).Conclusions:The most common predictors for Os were the underlying cause of HCC,AFP,and tumor *** having non-viral etiology,a tumor size>5 cm,an AFP>400 ng/mL,and a CTP score class C were all negatively associated with Os.

Dietary phenolic compounds （PCs） have been receiving intercst lor their presumcd roles ill disease prevention. However, there is a lack of studies on the underlying molecular mechanisnls. In this regard, in vitro metabolomic approaches are suitable for the investigation o1 the molecular changes in response to PC exposure. Up to date, the biological effects of PCs have only been cxalnined for PCs from rosemary （Rosmarinus officinalis）, olive oil, and resVCl-atrol using cell-based mctabolomic approach, although transcriptomic and/or proteomic studies have also been conducted in the same in vi[ro cell experiment in some cases. Our integral analysis of the reviewed studies suggest that PCs may be involved not only in basic cellular processes or macro-and micro-nutrient meta- bolism, but also in specific metabolic pathways that have been thoroughly investigated. These modulated pathways could have a clinical impact on neurodegenerativc diseases, type 2 diabetes, cancer, and cardiovascular diseases. In conclusion, the in vitro metabolomic approaches provide additional information of the molecular mechanisms involved in disease risk reduction of dietary PCs. In order to elucidate the mechanisms of action of PCs, more metabolomic cell-based studies are nccdcd and testing the physiological conjugated forms of PCs in these cell systems could be of special interest.

The Damage spreading(Ds)method allows the investigation of the effect caused by tiny perturbations,in the initial conditions of physical systems,on their final stationary or equilibrium *** damage(D(t))is determined during the dynamic evolution of a physical system and measures the time dependence of the difference between a reference(unperturbed)configuration and an initially perturbed *** this paper we first give a brief overview of Monte Carlo simulation results obtained by applying the Ds *** model systems under study often exhibit a transition between a state where the damage becomes healed(the frozen phase)and a regime where the damage spreads arriving at a finite(stationary)value(the damaged phase),when a control parameter is finely *** kinds of transitions are actually true irreversible phase transitions themselves,and the issue of their universality class is also ***,the attention is focused on the propagation of damage in magnetic systems placed in confined *** influence of interfaces between magnetic domains of different orientation on the spreading of the perturbation is also discussed,showing that the presence of interfaces enhances the propagation of the ***,the critical transition between propagation and nonpropagation of the damage is *** all cases,the determined critical exponents suggest that the Ds transition does not belong to the universality class of Directed Percolation,unlike many other systems exhibiting irreversible phase *** result reflects the dramatic influence of interfaces on the propagation of perturbations in magnetic systems.
