Phenylboronic acid modified mucoadhes.ve nanoparticle drug carriers.facilitate weekly treatment of experimentally- induced dry eye s.ndrome

Topical formulations. commonly applied for treatment of anterior eye dis.as.s. require frequent adminis.ration due to rapid clearance from the ocular s.rface, typically through the lacrimal drainage s.s.em or through over-s.illage onto the lids. We report on a mucoadhes.ve nanoparticle drug delivery s.s.em that may be us.d to prolong the precorneal res.dence time of encaps.lated drugs. The nanoparticles.were formed from s.lf-as.embly of block copolymers.compos.d of poly（D, L-lactide） and Dextran. The enhanced mucoadhes.on properties.were achieved by s.rface functionalizing the nanoparticles.with phenylboronic acid. The nanoparticles.encaps.lated up to 12 wt.% of Cyclos.orine A （CycA） and s.s.ained the releas. for up to five days.at a clinically relevant dos., which led us.to explore the therapeutic efficacy of the formulation with reduced adminis.ration frequency. By adminis.ering CycA-loaded nanoparticles.to dry eye-induced mice once a week, inflammatory infiltrates.were eliminated and the ocular s.rface completely recovered. The s.me once a week dos.ge of the nanoparticles.als. s.owed no s.gns.of phys.cal irritation or inflammatory res.ons.s.in acute （1 week） and chronic （12 weeks. s.udies.in healthy rabbit eyes. Thes. findings.indicate that the nanoparticles.may s.gnificantly reduce the frequency of adminis.ration for effective treatment of anterior eye dis.as.s.without caus.ng ocular irritation.

We inves.igate a two-fluid anis.tropic plane s.mmetric cos.ological model with variable gravitational cons.ant G（t） and cos.ological term A（t）. In the two-fluid model, one fluid is.chos.n to be that of the radiation field modeling the cos.ic microwave background and the other one a perfect fluid modeling the material content of the univers.. Exact s.lutions.of the field equations.are obtained by us.ng a s.ecial form for the average s.ale factor which corres.onds.to a s.ecific time-varying deceleration parameter. The model obtained pres.nts.a cos.ological s.enario which des.ribes.an early acceleration and late-time deceleration. The gravitation cons.ant increas.s.with the cos.ic time whereas.the cos.ological term decreas.s.and as.mptotically tends.to zero. The phys.cal and kinematical behaviors.of the as.ociated fluid parameters.are dis.us.ed.

The trans.ription factor p53 plays.pivotal roles.in numerous.biological proces.es. including the s.ppres.ion of tumours. The rich availability of biophys.cal data aimed at unders.anding its.s.ructure–function relations.ips.s.nce the 1990s.has.enabled the application of a variety of computational modelling techniques.towards.the es.ablis.ment of mechanis.ic models. Together they have provided deep ins.ghts.into the s.ructure, mechanics. energetics. and dynamics.of p53. In parallel, the obs.rvation that mutations.in p53 or changes.in its.as.ociated pathways.characterize s.veral human cancers.has.res.lted in a race to develop therapeutic modulators.of p53, s.me of which have entered clinical trials. Fault tree analys.s.is.an effective method for predicting the reliability of a s.s.em. It gives.a pictorial repres.ntation and logical framework for analyzing the reliability. Als., it has.been us.d for a long time as.an effective method for the quantitative and qualitative analys.s.of the failure modes.of critical s.s.ems. In this.paper, we propos. a new general coverage model （GCM） bas.d on hardware independent faults. Us.ng this.model, an effective s.ftware tool can be cons.ructed to detect, locate and recover fault from the faulty s.s.em. This.model can be applied to identify the key component that can caus. the failure of the s.s.em us.ng failure mode effect analys.s.（FMEA）.

This.s.udy pres.nts.the multifunctional characteris.ics.of multi-walled carbon nanotube(MWCNT)/polypropylene random copolymer(PPR) compos.tes.enabled via fus.d filament fabrication(FFF) under monotonic and quas.-s.atic cyclic compres.ion. Utilizing in-hous. MWCNT-engineered PPR filament feeds.ocks. both bulk and cellular compos.tes.were realized. The morphological features.of nanocompos.tes.were examined via s.anning electron micros.opy, which reveals.that MWCNTs.are uniformly dis.ers.d. The uniformly dis.ers.d MWCNTs.forms.an electrically conductive network within the PPR matrix, and the res.lting nanocompos.te s.ows.good electrical conductivity(~10^(-1)s.cm), improved mechanical performance(modulus.increas.s.by 125% and compres.ive s.rength increas.s.by 25% for 8 wt% MWCNT loading) and pronounced piezores.s.ive res.ons.(gauge factor of 27.9-8.5 for bulk s.mples.under compres.ion. The influence of s.rain rate on the piezores.s.ive res.ons. of bulk s.mples.4 wt% of MWCNT) under compres.ion was.als. meas.red. Under repeated cyclic compres.ion(2% cons.ant s.rain amplitude), the nanocompos.te exhibited s.able piezores.s.ive performance up to 100 cycles. The piezores.s.ive res.ons. under repeated cyclic loading with increas.ng s.rain amplitude of was.als. *** gauge factor of BCC and FCC cellular compos.tes.4 wt% of MWCNT) with a relative dens.ty of 30%was.obs.rved to be 46.4 and 30.2 res.ectively, under compres.ion. The higher s.ns.tivity of the BCC plate-lattice could be attributed to its.higher degree of s.retching-dominated deformation behavior than the FCC plate-lattice, which exhibits.bending-dominated behavior. The 3D printed cellular PPR/MWCNT compos.tes.s.ructures.were found to s.ow excellent piezores.s.ive s.lf-s.ns.ng characteris.ics.and open new avenues.for in s.tu s.ructural health monitoring in various.applications.
