Objective:to determine the morphological and molecular characterization of Acanthamoeba isolates from contact lens paraphernalia in Malaysia and to investigate their pathogenic potential based on the physiological ***:One hundred and eighty contact lens wearers donated their contact lens,lens storage cases and lens solutions between2018 and *** samples were inoculated onto 1.5%nonnutrient agar plates for 14 *** chain reaction(PCR)was performed and the amplified PCR products were sequenced and compared with the published sequences in *** pathogenic potential of positive isolates was further tested using temperature-tolerance and osmo-tolerance *** species were categorized into three distinct morphological groups established by Pussard and ***:Acanthamoeba was successfully isolated from 14(7.8%)culture-positive samples in which 11 belong to morphological groupⅡand 3 belong to morphological groupⅢ,*** sequencing of 18 S ribosomal RNA gene led to the identification of the t4 genotype in all the isolated *** vitro assays revealed that 9(64.3%)Acanthamoeba isolates were able to grow at 42℃and 1 M mannitol and were thus considered to be highly ***:to the best of our knowledge,this is the first report identifying the Acanthamoeba genotype and their pathogenic potential among contact lens wearers in *** potentially pathogenic t4 genotype isolated in this study is the most predominant genotype responsible for human ocular infection ***,increasing attention should be aimed at the prevention of contamination by Acanthamoeba and the disinfection of contact lens paraphernalia.

Role of urine studies in asymptomatic febrile neutropenic patients presenting to the emergency department

BACKGROUND: the role of urine studies in the detection of urinary tract infection(UtI) in febrile neutropenic patients with urinary symptoms(having a urinary catheter or having a positive urine analysis) is inarguable. However, the evidence is scarce regarding the indication for urine studies in asymptomatic(i.e., without urinary symptoms) patients with febrile neutropenia(FN) presenting to the emergency department(ED). the aim of this study is to evaluate the need for obtaining urine studies in asymptomatic febrile neutropenic ***: this was a retrospective cohort study conducted on adult cancer patients who presented to the ED with FN and had no urinary symptoms. We included all ED presentations of eligible patients between January 2013 and September 2018. Student's t-test and Wilcoxon rank-sum test were used for continuous data, while Chi-square and Fisher's exact tests were used for categorical data. Participants were divided into two groups based on their urine culture(UC) results: negative and positive UCs. two cut-offs were used for positive UC results: ≥10~5 cfu/m L and ≥10~4 cfu/m ***: We included 284 patients in our study. the age of our patient population was 48.5±18.5 years. More than two-thirds(68.7%) of patients had severe neutropenia, while only 3.9% and 9.9% of the patients had positive UCs at ≥10~5 cfu/m L and ≥10~4 cfu/m L, respectively. UCs were expectedly positive in most patients with urinalysis(UA) abnormalities. However, 27.3% and 32.1% of patients with positive UCs at ≥10~5 cfu/m L and ≥10~4 cfu/m L respectively had a normal UA. CONCLUSIONS: In our study, the incidence of UtI in adult febrile neutropenic cancer patients who present to the ED without urinary symptoms is low. Consequently, routine urine testing may not be warranted in this population, as it adds unnecessary financial burdens on the patients and delays timely management.

An effective vaccine against group A streptococcus(GAS)is highly desirable for definitive control of GAS *** the present study,two variants of amphiphilic chitosan nanoparticles-based GAS vaccines were *** vaccines were primarily composed of encapsulated KLH protein(a source of t helper cell epitopes)and lipidated M-protein derived B cell peptide epitope(lipoJ14)within the amphiphilic structure of *** only difference between themwas one of the nanoparticles vaccines received additional surface coating with poly(I:C).the formulated vaccines exhibited nanosized particles within the range of 220–240 *** uptake study showed that nanoparticles vaccine without additional poly(I:C)coating has greater uptake by dendritic cells and macrophages compared to nanoparticles vaccine that was functionalized with poly(I:C).Both vaccines were found to be safe in mice and showed negligible cytotoxicity against HEK293 *** immunization in mice,both nanoparticle vaccines produced high antigen-specific antibodies titres that were regulated by a balanced th1 and th2 response compared to physical *** antibodies elicited high opsonic activity against the tested GAS ***,our data demonstrated that amphiphilic chitosan nanoparticles platform induced a potent immune response even without additional inclusion of poly(I:C).

One of the many problems that are considered to be NP-Hard is the Multiple Sequence Alignment one that initially requires, as for any other of its siblings, a specific encoding schema and design of the main functionalities of the heuristics algorithm being implemented and executed. this paper intends to discuss our proposed generic implementation of the Simulated Annealing, inspired for the procedure of cooling and shaping methods of metals. In our algorithm, we attempted to add some executions tracing functionalities in order to help later analysis for initial parameters tuning. On another hand, we also tried to get closer in our attempt to mimic the cooling of metals, but giving it an option to run under different cooling schedules. We proposed a few schedules that seemed to be studied and/or used in many algorithm implementations.

the Internet of things (Iot) is a technological revolution that has changed everything we do and given us a new perspective on our daily lives, but despite the fact that numerous publications have focused on characterizing the many edges and technologies that make up an Iot system, the Iot ecosystem is still seen as too complex to be recognized as a stand-alone environment due to its significant diversity;hence, the objective of this research is to address such a complex environment in a way that highlights its components and distinguishes them both individually and in relation to their broader context. therefore, the definition of Iot and its emergence were discussed and organized around the timeline of Internet development phases demonstrating that Iot has been a need that has accompanied the presence of the Internet since its early stages, and then its growth and impact were discussed and highlighted with estimates and numbers. On the technical side, each of the following groups, Iot components, protocols, and architectures, was defined, discussed, and grouped in such a way that their intergroup organization, as well as their placement and contribution to the overall ecosystem, was highlighted. this, in addition to the various examples mentioned throughout the discussion, will provide the reader with a better understanding of the Internet of things and how deeply it has become entwined in our daily lives and routines as a result of its numerous applications.

以船舶艉轴承为研究对象,基于质量守恒边界条件,编制相应的润滑仿真程序,研究艉轴承的润滑性能,探讨轴承润滑性能随轴承运行参数(如转速、轴承宽度、外载荷及空穴压力等)变化的规律。研究表明:质量守恒边界条件相比于雷诺边界条件,更加接近实际情况;空穴压力的增大并不总是有损轴承的承载力;随着空穴压力和轴承宽度的增大,偏心率和最大油膜压力都减小;随着外载荷的增加,油膜压力和空穴区域大小逐渐增大,呈现出很强的非线性趋势;随着转速的增大,油膜压力峰值减小;空穴区域随转速增大而增大,其位置会向最大轴承间隙处移动。

Arsenic trioxide induces regulatory functions of plasmacytoid dendritic cells through interferon-α inhibition

Arsenic trioxide(As2O3)is recently found to have therapeutic potential in systemic sclerosis(SSc),a life-threatening multi-system fibrosing autoimmune disease with type I interferon(IFN-I)*** activated plasmacytoid dendritic cells(pDCs)are responsible for IFN-I secretion and are closely related with fibrosis establishment in *** this study,we showed that high concentrations of As2O3 induced apoptosis of pDCs via mitochondrial pathway with increased BAX/BCL-2 ratio,while independent of reactive oxygen species ***,at clinical relevant concentrations,As2O3 preferentially inhibited IFN-αsecretion as compared to other cytokines such as tNF-α,probably due to potent down-regulation of the total protein and mRNA expression,as well as phosphorylation of the interferon regulatory factor7(IRF7).In addition,As2O3 induced a suppressive phenotype,and in combination with cytokine inhibition,it down-regulated pDCs’capacity to induce CD4+t cell proliferation,thl/th22 polarization,and B cell differentiation towards ***,chronically activated pDCs from SSc patients were not resistant to the selective IFN-αinhibition,and regulatory phenotype induced by ***,our data suggest that As2O3 could target pDCs and exert its treatment efficacy in SSc,and more autoimmune disorders with IFN-I signature.

BACKGROUND Numerous studies investigated cell-based therapies for myocardial infarction(MI).the conflicting results of these studies have established the need for developing innovative approaches for applying cell-based therapy for *** studies on animal models demonstrated the potential of fresh,uncultured,unmodified,autologous adipose-derived regenerative cells(UAADRCs)for treating acute *** contrast,studies on the treatment of chronic MI(CMI;>4 wk post-MI)with UA-ADRCs have not been published so *** several methods for delivering cells to the myocardium,retrograde delivery into a temporarily blocked coronary vein has recently been demonstrated as an effective *** to test the hypothesis that in experimentally-induced chronic myocardial infarction(CMI;>4 wk post-MI)in pigs,retrograde delivery of fresh,uncultured,unmodified,autologous adipose-derived regenerative cells(UA-ADRCs)into a temporarily blocked coronary vein improves cardiac function and *** the left anterior descending(LAD)coronary artery of pigs was blocked for 180 min at time point ***,either 18×106 UA-ADRCs prepared at“point of care”or saline as control were retrogradely delivered via an over-the-wire balloon catheter placed in the temporarily blocked LAD vein 4 wk after t0(t1).Effects of cells or saline were assessed by cardiac magnetic resonance(CMR)imaging,late gadolinium enhancement CMR imaging,and post mortem histologic analysis 10 wk after t0(t2).RESULtS Unlike the delivery of saline,delivery of UA-ADRCs demonstrated statistically significant improvements in cardiac function and structure at t2 compared to t1(all values given as mean±SE):Increased mean LVEF(UA-ADRCs group:34.3%±2.9%at t1 vs 40.4±2.6%at t2,P=0.037;saline group:37.8%±2.6%at t1 vs 36.2%±2.4%at t2,P>0.999),increased mean cardiac output(UA-ADRCs group:2.7±0.2 L/min at t1 vs 3.8±0.2 L/min at t2,P=0.002;saline group:3.4±0.3 L/min at t1 vs 3.6±0.3 L/min at t2,P=0.798),increased mean mass of the
