Immediate implantation of ultrafine fiber slow-release system based on cell electrospinning to induce osteogenesis of mesenchymal stem cells

This study presents the development and evaluation of a poly(3-hydroxybutyrate-co-4-hydroxybutyrate)(P34HB)ultrafine fiber slow-release system for in vivo osteogenic induction of human umbilical cord mesenchymal stem cells(HUCMsCs).Utilizing dual-nozzle and cell electrospinning techniques,the system encapsulates L-ascorbic acid-2-phosphate magnesium(AsP),βB-glycerophosphate sodium and dexamethasone(DEX)within the'fbers,ensuring sustained osteogenic *** scaffold's morphology,characterization,hydrophilicity,mechanical properties and cellular behavior were *** subcutaneous implantation in rabbits was con-ducted to observe its ectopic osteogenic induction *** fabricated P34HB ultrafine fiber slow-release *** ization *** distribution of HUCMsCs and inducing components within the scaffold,with no chemical reactions affecting the active *** vitro tests showcased a prolonged release of DEX and AsP,while biocompatibility assays highlighted the scaffold's suitability for cellular *** Red,type I collagen,and osteopontin(OPN)staining verifed the scaffold's potent osteogenic induction effect on ***,immediate implantation into New Zealand White rabbits led to significant new bone formation within 8 *** findings underscore the system's potential for immediate in vivo implantation without prior in vitro induction,marking a promising advancement in bone tissue engineering.

Molecular mechanism of Xuebijing in treating pyogenic liver abscess complicated with sepsis

BACKGROUND:Xuebijing(XBJ)can alleviate the inflammatory response,improve organ function,and shorten the intensive care unit(ICU)stay in patients with pyogenic liver abscess(PLA)complicated with sepsis,but the molecular mechanisms have not been *** study aimed to explore the molecular mechanism of XBJ in treating PLA complicated with sepsis using a network pharmacology ***:The active ingredients and targets of XBJ were retrieved from the ETCM *** targets related to PLA and sepsis were retrieved from the GeneCards,PharmGKB,DisGenet,Online Mendelian Inheritance in Man(OMIM),Therapeutic Targets Database(TTD),and DrugBank *** targets of PLA complicated with sepsis were mapped to the targets of XBJ to identify potential treatment ***-protein interaction networks were analyzed using the sTRING *** treatment targets were imported into the Metascape platform for Gene Ontology(GO)functional enrichment and Kyoto encyclopedia of Genes and Genomes(KEGG)pathway enrichment *** docking was performed to validate the interactions between active ingredients and core ***:XBJ was found to have 54 potential treatment targets for PLA complicated with ***-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor(TNF)were identifi ed as core *** enrichment analysis revealed important pathways,including the interleukin-17(IL-17)signaling pathway,the TNF signaling pathway,the nuclear factor-kappa B(NF-κB)signaling pathway,and the Toll-like receptor(TLR)signaling *** docking experiments indicated stable binding between XBJ active ingredients and core ***:XBJ may exert therapeutic eff ects on PLA complicated with sepsis by modulating signaling pathways,such as the IL-17,TNF,NF-κB,and TLR pathways,and targeting IL-1β,IL-6,and TNF.

The OsCLV2s-OsCRN1 co-receptor regulates grain shape in rice

The highly conserved CLV–WUs negative feedback pathway plays a decisive role in regulating stem cell maintenance in shoot and floral meristems in higher plants,including Arabidopsis,rice,maize,and ***,we find significant natural variations in the OsCLV2c,OsCLV2d,and OsCRN1 loci in a genome-wide association study of grain shape in ***2a,OsCLV2c,OsCLV2d,and OsCRN1 negatively regulate grain length–width ratio and show distinctive geographical distribution,indica–japonica differentiation,and artificial selection ***,OsCLV2a and OsCRN1 interact biochemically and genetically,suggesting that the two components function in a complex to regulate grain shape of ***,the genetic contributions of the haplotypes combining OsCLV2a,OsCLV2c,and OsCRN1 are significantly higher than those of each single gene alone in controlling key yield *** findings identify two groups of receptor-like kinases that may function as distinct co-receptors to control grain size in rice,thereby revealing a previously unrecognized role of the CLV class genes in regulating seed development and proposing a framework to understand the molecular mechanisms of the CLV–WUs pathway in rice and other crops.

Experimental and numerical simulation study on the erosion behavior of the elbow of gathering pipeline in shale gas field

During the production period of shale gas, proppant particles and rock debris are produced together,which will seriously erode the elbows of gathering pipelines. In response to this problem, this paper takes the elbow of the gathering pipeline in the Changning shale Gas Field as an example to test the erosion rate and material removal mechanism of the test piece at different angles of the elbow through experiments and compares the four erosion models with the experimental results. Through analysis, it is found that the best prediction model for quartz sand-carbon steel erosion is the Oka model. Based on the Oka model, FLUenT software was used to simulate and analyze the law of erosion of the elbow of the gas gathering pipeline under different gas flow velocities, gas gathering pressure, particle size, length of L1,and bending directions of the elbow. And a spiral pipeline structure is proposed to reduce the erosion rate of the elbow under the same working conditions. The results show that this structure can reduce erosion by 34%.

This paper considers the random coefficient autoregressive model with time-functional variance noises,hereafter the RCA-TFV *** first establish the consistency and asymptotic normality of the conditional least squares estimator for the constant *** semiparametric least squares estimator for the variance of the random coefficient and the nonparametric estimator for the variance function are constructed,and their asymptotic results are reported.A simulation study is presented along with an analysis of real data to assess the performance of our method in finite samples.

Exogenous testosterone therapy on choroid in androgen deficiency

AIM:To investigate the effects of exogenous testosterone treatment on the choroidal parameters in patients with androgen ***:Right eyes of 24 patients with androgen deficiency and 31 healthy volunteers were included in the *** eyes were scanned for subfoveal choroidal thickness(sFCT),choroidal vascularity index(CVI),choroidstromal area(C-sA),choroid-luminal area(C-LA),choroidstromal to luminal area ratio(CsLR),and the choroidal parameters within central 1500μm of the macula(CVI1500,C-LA1500,C-sA1500,and CsLR1500)by enhanced-depth imaging optical coherence tomography(EDI-OCT)at baseline,6th and 18th weeks of the exogenous testosterone ***:The mean sFCT values of the androgen deficient groups and healthy controls were 307.7±27.0 and 303.2±37.2μm(P=0.8).However,CVI,C-sA,CsLR,CVI1500,C-LA1500,and CsLR1500 were significantly different between the groups(all Psit after exogenous testosterone treatment,sFCT,CVI,C-LA,and C-sA were significantly decreased,and these parameters returned to baseline levels at the 18th-week visit(all P>0.05).However,CVI1500 and LA1500 significantly increased at the end of the follow-up period(PsION:CVI is lower in androgen-deficient patients than in healthy *** alterations in the choroid during the testosterone peak are transient in the treatment of patients with androgen ***,the increase in CVI within the central 1500μm of the macula persists even after 4mo.

Gapless Genome Assembly of ZH8015 and Preliminary Multi-Omics Analysis to Investigate ZH8015's Responses Against Brown Planthopper Infestation

Accurate genomic information is essential for advancing genetic breeding research in specific rice *** study presented a gapless genome assembly of the indica rice cultivar Zhonghui 8015(ZH8015)using Pac Bio HiFi,Hi-C,and ONT(Oxford Nanopore Technologies)ultra-long sequencing technologies,annotating 43037 gene ***,utilizing this genome along with transcriptomic and metabolomic techniques,we explored ZH8015's response to brown planthopper(BPH)*** transcriptomic sampling indicated significant changes in gene expression levels around 48 h after BPH *** analysis revealed particularly significant alterations in genes related to reactive oxygen species scavenging and cell wall *** results demonstrated marked increases in levels of several monosaccharides,which are components of the cell wall and dramatic changes in flavonoid *** association analysis identified differentially expressed genes associated with key metabolites,shedding light on ZH8015's response to BPH *** summary,this study constructed a reliable genome sequence resource for ZH8015,and the preliminary multi-omics results will guide future insect-resistant breeding research.

Rational design of robust and transparent superrepellent surfaces for long-term marine antifouling

superhydrophobic surfaces have demonstrated exceptional efficacy in combatting biofouling contaminations of optical devices and equipment in marine applications. However, the fabrication of highly transparent superhydrophobic materials remains a formidable challenge due to the inherent trade-off between surface roughness for superhydrophobicity and optical ***, we design a robust and transparent superhydrophobic coating(si-POss) embedded silica nanoparticles(200 nm) with fluorinated polyhedral oligomeric silsesquioxanes(F-POss) and zinc pyrithione(ZPT). The si-POss coating exhibits excellent water repellence toward diverse liquids and optical transmittance exceeding 90% in the visible spectrum. Moreover, the si-POss coating sustains long-term anti-bacterial(> 99.11%) and anti-algal effects for over 30 days, accompanied by mechanical,chemical, and thermal stability. This research asserts that the si-POss coating with outstanding combined characteristics holds significant potential for marine applications, particularly in self-cleaning and antifouling endeavors.

Na_(3)P interphase reduces Na nucleation energy enabling stable anode-less sodium metal batteries

sodium metal batteries(sMBs)are rising as viable alternatives to lithium-ion systems due to their superior energy density and sodium's relative ***,sMBs face significant impediments,particularly the exceedingly high negative-to-positive capacity ratios(N/P ratios)which severely encumber energy density and hinder their practical ***,a novel nucleophilic Na_(3)P interphase on aluminum foil has been designed to significantly lower the nucleation energy barrier for sodium atom deposition,resulting in a remarkable reduction of nucleation overpotential and efficient mitigation of dendritic growth at high sodium deposition of 5 mA h cm^(−2).The interphase promotes stable cycling in anode-less sMB configurations with a low N/P ratio of 1.4 and high cathode mass loading of 11.5 mg cm^(−2),and demonstrates a substantial increase in high capacity retention of 92.4%after 500 cycles even under 1 C rate *** innovation signifies a promising leap forward in the development of high-energy-density,anode-less sMBs,offering a potential solution to the longstanding issues of cycle stability and energy efficiency.
