A peptide of 17 aminoacids from the N-terminal region of maize plastidial transglutaminase is essential for chloroplast targeting

Transglutaminases (TGases, EC 2.***.2.13) catalyse posttranslational modification of proteins by establishing ε-(γ-glutamyl) links and covalent conjugation of polyamines. In plants, the functionality of these enzymes is scarcely known. The maize transglutaminase gene (tgz), the only cloned plant TGase, produces major alterations in thylakoid membrane architecture when the transglutaminase (chlTGZ) protein was over-expressed in tobacco chloroplasts, significantly increasing the number of grana stacked layers. Here we demonstrate that nuclear transformation of rice plants starting from a tgz gene truncated in 17 N-terminal aas (tgzt) non altered chloroplast thylakoid structures. F3 transformed plants were analysed for TGase activity, chlTGZ presence and tgzt transcription levels. Transformed plants exhibited double the in vitro TGase activity of the non-transformed plants. Immunoblot and quantitative RT-PCR analysis results of tgzt-rice plants grown under different illumination periods revealed that chlTGZ maintains its differential expression depending on the light regime. Nevertheless, the maize protein was localised by confocal microscopy in the cell wall of transformed rice cells. TEM analyses of the transformed cells showed normal, non-altered chloroplast thylakoid structures with the maize protein preferentially located in the cell walls. The results confirmed that the tgz eliminated sequence is essential for chloroplast targeting, being its absence sufficient to the lack of protein expression in its original plastidal compartment. Interestingly, the immunolocalization of a putative endogenous rice TGase protein is also showed. These data give further information on plant TGase functionality and its relationship to photosynthetic membranes.

目的:通过随机对照研究,评估针刺在智利地区居丧人群中对其创伤应激障碍的疗效以及安全性。方法:病例来源经由智利蝴蝶基金会(Fundación Mariposas)宣传推介,于智利当地南美中医学院下属门诊部招募符合纳入标准的90名创伤应激障碍患者,共纳入90名患者,脱落2名,最终纳入有效病例88例。通过随机数字法将研究对象分为针刺组(44例)、对照组(44例)。治疗方案针刺组予基础治疗+针刺治疗;对照组予基础治疗+安慰针治疗。基础治疗基础治疗主要为心理动力治疗,包括认知行为治疗(CBT)、Beck认知治疗以及集体心理治疗等。包括集体倾诉与倾听、识别自动性想法与认知错误以及互相共情与抚慰三部分。每次2小时,每周1次,总共12次治疗。针刺组选穴:选取百会、四神聪为主穴,配穴选取五脏俞。操作:操作环境在针灸门诊或其他安静舒适的环境。采用苏州产环球牌一次性管针。嘱受试者取舒适俯卧位,术者手指及腧穴皮肤消毒后,以管针弹入进针。头部穴位平刺进针0.3～0.5寸,背部腧穴直刺进针1.0～1.5寸。进针后稍事捻转,得气即止,不行补泻或导气手法。留针30分钟后取针,留针期间予红外治疗仪照射以避免患者受凉。每周治疗2次,总共治疗10次。对照组选穴与定位同针刺组操作:操作环境在针灸门诊或其他安静舒适的环境。采用苏州产环球牌一次性带管平头针,嘱受试者取舒适俯卧位,术者手指及腧穴皮肤消毒后,以管针弹针尾。使针头轻压患者皮肤但不刺入身体。包括头部与背部穴位均稍事捻转,使患者有被针刺感,但未有针实际刺入穴位得气。术者稍事捻转后嘱患者勿轻易活动,保持30分钟不留针,期间同予红外治疗仪照射患者背部避免受凉。每周治疗2次,总共治疗10次。评价时点及指标分别在治疗前(第1次治疗前)、治疗中(第5次治疗后)、治疗结束(第10次治疗结束后)。采用CORE-10量表评分来观察不同时点组内及组间疗效差异,同时对CORE-10量表内不同维度进行分层分析。同时记录研究中所出现不良反应及副作用。统计将收集到的病例信息,统计计数组间的差异性比较根据数据类型及分布,合理选用(校正)卡方检验;等级资料选用秩和检验。计量资料中,若符合正态分布且方差齐,则以均数加减标准差(x±s)进行统计学描述,并采用t检验(其中组间比较采用独立样本t检验,组内强采用配对t检验)。若不符合正态分布,则以四分位数(R(P25,P50,P75))进行统计学描述,采用秩和检验(Kruskal-Wallis),组内两两比较采用秩和检验(Wilcoxon),组间两两比较采用秩和检验(Mann-Whitney)。多时点的计量资料采用重复测量的方差分析,各时点组间比较采用多变量方差分析。所有检验为双侧检验,检验水平设为α=0.05。结果:1.基线本研究共纳入患者90例,有效病例88例,脱落2例,脱落率为2.2%。其中针刺组44例,对照组44例。两组患者治疗前一般基线比较,在年龄(P=0.259)、性别(P=0.338)、婚姻状况(P=0.103)、病程(P=0.831)、与死者亲缘关系(P=0.933)、亡者死因(P=0.125)和受教育背景(P=0.692)方面均无明显差异,具有可比性。***-10总评分经Mauchly球形度检验,两组指标方差不齐(P=0.001<0.05),组内效应经Greenhouse-Geisser检验,时间维度上有统计学意义(P=0.000<0.05),提示随时间变化两组患者病情都有所缓解。时间*组别维度上亦有统计学意义(P=0.000<0.05),提示时间与组别两个维度具有交互作用。主体间效应检验见两组在组别维度上有统计学意义(P=0.024<0.05)。提示两组指标随时间变化趋势不同,两组疗效有差异。针刺组治疗前后整体结局指标减分较对照组更甚。***-10焦虑维度评分经Mauchly球形度检验,两组指标方差不齐(P=0.001<0.05),组内效应经Greenhouse-Geisser检验,时间维度上有统计学意义(P=0.000<0.05),提示随时间变化两组患者病情都有所缓解。时间*组别维度上亦有统计学意义(P=0.000<0.05),提示时间与组别两个维度具有交互作用。主体间效应检验见两组在组别维度上有统计学意义(P=0.000<0.05)。提示两组指标随时间变化趋势不同,两组疗效有差异。针刺组治疗前后整体结局指标减分较对照组更甚。***-10抑郁维度评分经Mauchly球形度检验,两组指标方差不齐(P=0.001<0.05),组内效应经Greenhouse-Geisser检验,时间维度上有统计学意义(P=0.000<0.05),提示随时间变化两组患者病情都有所缓解。时间*组别维度上亦有统计学意义(P=0.000<0.05),提示时间与组别两个维度具有交互作用。主体间

Health systems are paradigmatic examples of human organizations that blend a multitude of different professional and disciplinary features within a critically performance environment. Communication failure and defective processes in health systems have a tremendous impact in society, both in the financial and human aspects. Traditionally, health systems have been regarded as linear hierarchic structures. However, recent developments in the sciences of complexity point out to health systems as complex entities governed by non-linear interaction laws, self-organization and emergent phenomena. In this work we review some aspects of complexity behind health systems and how they can be applied to improve the performance of healthcare organizations.

Melatonin Avoids Anatomofunctional Changes Associated to Aging in a Rat Model

Melatonin is a hormone synthesized and released primarily by the pineal gland. Its secretion fol-lows a circadian rhythm with a peak overnight. Its secretion is initiated approximately to the three months of age and continues to rise during the childhood. Previous to the puberty there is a de-crease of melatonin secretion that continues until the old age. Melatonin has effects in the body and acts through at least four mechanisms: membrane receptors, orphan nuclear receptors, calmodulin and free radicals. It has been suggested that aging can be a consequence of the oxidation of cells that eventually become vulnerable to injury and die. This work reviews the antioxidant effects of melatonin in a rodent model, on the formation of free radicals, on the MAP2 protein expression and on the electrophysiology of the hippocampus at different ages. The results indicate that melatonin maintains in a “best” state to the experimental animals compared to controls. It suggests the use of melatonin as a therapy to prevent or delay the aging effects on the cells.

In Vitro Propagation of Echeveria elegans, a Species of the Flora Endangered Mexican

The genus Echeveria has about 143 species that are unique to the Americas, 117 of which are presented in the Mexican Republic, mainly in the states of Hidalgo, Puebla and Oaxaca. Propagation can be done by cuttings, leaf cuttings or seeds, but these methods are insufficient to avoid the predation of many species. NOM-059-ECOL-2001 shows Echeveria elegans as species reported endangered. The objectives in this study were achieved in vitro propagation from axillary buds, as an alternative to multiplication and preservation species. We evaluated different concentrations and combinations of two growth regulators, 6-BAP （2.22, 4.44, 6.66 and 8.88 μM） and u-NAA （1.35 and 2.70 μM） on shoot formation from axillary buds and two culture media with different concentrations of MS salts, to achieve root plants grown in vitro. It was found by combining 6.66 μM of 6-BAP and 1.35μM of α-NAA favored the formation and development of new shoots. In the rooting stage, the best results are achieved with the treatment containing 50% of MS salts. Then the plants were transplanted into greenhouses and achieved a successful acclimatization. During this last phase of development, there were no phenotypic changes or presence of somaclonal variants.

On Second Riesz Φ-Variation of Normed Space Valued Maps

In this article we present a Riesz-type generalization of the concept of second variation of normed space valued functions defined on an interval [a,b]R. We show that a function f [a,b], where X is a reflexive Banach space, is of bounded second Φ-variation, in the sense of Riesz, if and only if it can be expressed as the (Bochner) integral of a function of bounded (first) $\Phi$-variation. We provide also a Riesz lemma type inequality to estimate the total second Riesz-Φ-variation introduced.

In the 1990s, misoprostol (synthetic prostaglandin E1 analogue) was found to be an effective abortive agent when taken orally and became widely used in Latin America as a means to terminate unwanted pregnancies. A variety of congenital anomalies have been observed among the children of women who ingested misoprostol, but failed to terminate their pregnancy. We report here eight years of experience in Panama with the detection and follow-up of the malformations seen in infants associated with the use of misoprostol prostaglandin during the first trimester of pregnancy. During the period between April 1995 and March 2003, we identified 63 infants at the Panama’s Children’s Hospital who were exposed to misoprostol while in the womb and who were born with malformations. These infants were evaluated by a team of neonatologists, geneticists, cardiologists, ophthalmologists, and radiologists.

Melatonin changes the electrical spontaneous activity of hippocampal rat neurons at different ages

Melatonin, the pineal indole is characterized by being a compound that crosses all cell membranes and which has been attributed to several mechanisms of action. Among these it is the ability to reduce free radicals, thereby reducing the potential aging and cell death. Studies in different age Wistar rats have shown that chronic application of melatonin, in the hippocampus, reduces the concentration of free radicals and keeps its architecture. This study showed that melatonin increases the firing rate and favors the presence of bursting activity in animals of different ages. It is suggested that melatonin conserved hippocampal cells in good anatomical and physiological condition probably as a result of the elimination of free radicals.

Structured Perovskite-Based Oxides: Use in the Combined Methane Reforming

The behavior of metallic structured perovskite-based catalysts was evaluated in the combined methane reforming reaction with CO2-O2. The reaction conditions were established by varying the reaction temperature and reactor input composition in the range of 650 to 850℃ and CH4/CO2 ratio 1 to 5, respectively. The results of the catalytic tests at 750℃ showed a positive effect of the metallic structure, producing higher conversions and H2/CO ratios in the products compare to that obtained with the powder catalyst.
