Yeast artificial chromosomes(YACs) are important tools for sequencing,gene cloning,and transferring large quantities of genetic information.However,the structure and activity of YAC chromatin,as well as the unintended impacts of introducing foreign DNA sequences on DNA-associated biochemical events,have not been widely explored.Here,we showed that abundant genetic elements like TATA box and transcription factor-binding motifs occurred unintentionally in a previously reported datacarrying chromosome(d Chr).In addition,we used state-of-the-art sequencing technologies to comprehensively profile the genetic,epigenetic,transcriptional,and proteomic characteristics of the exogenous d Chr.We found that the data-carrying DNA formed active chromatin with high chromatin accessibility and H3K4 tri-methylation levels.The d Chr also displayed highly pervasive transcriptional ability and transcribed hundreds of noncoding RNAs.The results demonstrated that exogenous artificial chromosomes formed chromatin structures and did not remain as naked or loose plasmids.A better understanding of the YAC chromatin nature will improve our ability to design better data-storage chromosomes.

The widespread adoption of lithium-ion batteries has been driven by the proliferation of portable electronic devices and electric vehicles,which have increasingly stringent energy density requirements.Lithium metal batteries(LMBs),with their ultralow reduction potential and high theoretical capacity,are widely regarded as the most promising technical pathway for achieving high energy density batteries.In this review,we provide a comprehensive overview of fundamental issues related to high reactivity and migrated interfaces in LMBs.Furthermore,we propose improved strategies involving interface engineering,3D current collector design,electrolyte optimization,separator modification,application of alloyed anodes,and external field regulation to address these challenges.The utilization of solid-state electrolytes can significantly enhance the safety of LMBs and represents the only viable approach for advancing them.This review also encompasses the variation in fundamental issues and design strategies for the transition from liquid to solid electrolytes.Particularly noteworthy is that the introduction of SSEs will exacerbate differences in electrochemical and mechanical properties at the interface,leading to increased interface inhomogeneity—a critical factor contributing to failure in all-solidstate lithium metal batteries.Based on recent research works,this perspective highlights the current status of research on developing high-performance LMBs.

Multiplexed sequencing relies on specific sample labels,the barcodes,to tag DNA fragments belonging to different samples and to separate the output of the sequencers.However,the barcodes are often corrupted by insertion,deletion and substitution errors introduced during sequencing,which may lead to sample misassignment.In this paper,we propose a barcode construction method,which combines a block error-correction code with a predetermined pseudorandom sequence to generate a base sequence for labeling different samples.Furthermore,to identify the corrupted barcodes for assigning reads to their respective samples,we present a soft decision identification method that consists of inner decoding and outer decoding.The inner decoder establishes the hidden Markov model(HMM)for base insertion/deletion estimation with the pseudorandom sequence,and adapts the forward-backward(FB)algorithm to output the soft information of each bit in the block code.The outer decoder performs soft decision decoding using the soft information to effectively correct multiple errors in the barcodes.Simulation results show that the proposed methods are highly robust to high error rates of insertions,deletions and substitutions in the barcodes.In addition,compared with the inner decoding algorithm of the barcodes based on watermarks,the proposed inner decoding algorithm can greatly reduce the decoding complexity.

Physicochemical principles of hydrogen metallurgy in blast furnace

Based on the stoichiometric method and the free energy minimization method,an ideal model for the reduction of iron oxides by carbon and hydrogen under blast furnace conditions was established,and the reduction efficiency and theoretical energy consumption of the all-carbon blast furnace and the hydrogen-rich blast furnace were compared.The results show that after the reduction reaction is completed at the bottom of the blast furnace,the gas produced by reduction at 1600℃still has a certain excessive reduction capacity,which is due to the hydrogen brought in by the hydrogen-rich blast as well as the excess carbon monoxide generated by the reaction of the coke and the oxygen brought in by the blast.During the process of the gas with excessive reduction capacity rising from the bottom of the blast furnace and gas reduction process,the excessive reduction capacity of the gas gradually decreases with the increase in the dydrogen content in the blast.In the all-carbon blast furnace,the excess gas reduction capacity is the strongest,and the total energy consumption per ton of iron reduction is the lowest.This shows that,for the current operation mode of the blast furnace,adding hydrogen in the blast furnace cannot reduce the consumption of carbon required for reduction per ton of iron,but rather increases the consumption of carbon.

Autoimmune diseases(AID)encompass a diverse array of conditions characterized by immune system dysregulation,resulting in aberrant responses of B cells and T cells against the body’s own healthy tissues.Plant extracellular vesicles(PEVs)are nanoscale particles enclosed by phospholipid bilayers,secreted by plant cells,which facilitate intercellular communication by transporting various bioactive molecules.Due to their nanoscale structure,safety,abundant sources,low immunogenicity,high yield,biocompatibility,and effective targeting of the colon and liver,PEVs are regarded as a promising platform for the treatment of AID.This review provides a comprehensive summary of PEV biogenesis,physicochemical and biological properties,internalization mechanisms,isolation methods,and their applications in various diseases,with a specific focus on their potential roles in AID.Additionally,we propose engineering approaches and administration methods for PEVs.Finally,we present an overview of the advantages and challenges associated with utilizing PEVs for the treatment of AID.By gaining a comprehensive understanding of PEVs,we anticipate the development of innovative therapeutic strategies for AID.Natural and engineered PEVs hold substantial promise as a valuable resource for innovative technologies in AID treatment.

针对一类做重复运动的不确定全驱系统,提出了一种带有未知参数精确估计的迭代学习自适应滑模控制算法。通过设计跟踪误差的期望轨迹,放宽了对系统初值条件的限制,并使系统在初始时刻便位于滑模面上,消除了滑模到达阶段,有利于提高系统鲁棒性。通过构造一组低通滤波器并引入遗忘因子和归一化技术,对参数估计误差进行了重构,并利用重构结果进行了微分-差分型自适应律设计。基于Lyapunov方法设计了迭代学习自适应滑模控制算法,证明了闭环系统信号的有界性以及跟踪误差和参数估计误差的渐近收敛性。最后,将所提出的迭代学习自适应滑模控制算法应用于航天器的绕飞控制中,并通过数值仿真验证了设计结果的有效性。

In this paper,a new concept of forward-pumped random Raman fiber laser(RRFL)-based liquid refractive index sensing is proposed for the first time.For liquid refractive index sensing,the flat fiber end immersed in the liquid can act as the point reflector for generating random fiber lasing and also as the sensing head.Due to the high sensitivity of the output power of the RRFL to the reflectivity provided by the point reflector in the ultralow reflectivity regime,the proposed RRFL is capable of achieving liquid refractive index sensing by measuring the random lasing output power.We theoretically investigate the effects of the operating pump power and fiber length on the refractive index sensitivity for the proposed RRFL.As a proof-of-concept demonstration,we experimentally realize high-sensitivity half-open short-cavity RRFL-based liquid refractive index sensing with the maximum sensitivity and the sensing resolution of-39.88W/RIU and 2.5075×10^(-5) RIU,respectively.We also experimentally verify that the refractive index sensitivity can be enhanced with the shorter fiber length of the RRFL.This work extends the application of the random fiber laser as a new platform for highly-sensitive refractive index sensing in chemical,biomedical,and environmental monitoring applications,etc.

Sulfidated nano zero-valent iron particles were immobilized on ZSM-5 zeolite(Z/S-nZⅥ)and used for hexavalent chromium(Cr(Ⅵ))remediation.The performance of Z/S-nZⅥimproved with the increase in Cr(Ⅵ)concentration(han 60 mg/L.The adsorption behavior for Cr(Ⅵ)was different from that reported in previous studies.The improved performance can be tailored for increasing efficiency of nano zero-valent iron(nZⅥ)corrosion,while the degree of corrosion of nZⅥwas affected by the concentration of the pollutant as discussed by kinetics,X-ray diffraction(XRD)and X-ray photoelectron spectrometer(XPS)analyses.The experiments for the dissolution of ferrous ions and the dosage of adsorbent demonstrated that the critical layer in the liquid-solid system changed with the increase in the concentration of Cr(Ⅵ)(Cr(Ⅵ):Z/S-nZⅥ>0.6).Moreover,the removal mechanisms of Cr(Ⅵ)were elucidated through XRD,transmission electron microscopy(TEM)and XPS techniques.This results demonstrate that the species of chromium in the critical layer changed from Cr(Ⅲ)to Cr(Ⅵ)as the concentration of chromium increased from low to high.Furthermore,the critical layer was composed of Cr(Ⅵ),Fe(Ⅱ),O and H elements.Additionally,the experiments of coexisting ions and aging time confirmed that Z/S-nZⅥpossessed high selectivity and stability to ensure efficiency and cost-effectiveness in practical applications.
