We aimed to characterize NDM-5-producing Enterobacteriaceae from aquatic products in Guangzhou,China.A total of 196 intestinal samples of grass carp collected in 2019 were screened for carbapenemase genes.Characterization of bla_(NDM-5) positive isolates and plasmids was determined by antimicrobial susceptibility testing,conjugation experiments,Illumina HiSeq,and Nanopore sequencing.One Citrobacter freundii and six Escherichia coli strains recovered from seven intestinal samples were verified as bla_(NDM-5) carriers(3.57%,7/196).The bla_(NDM-5) genes were located on the lncX3(n=5),lncHI2(n=1),or lncHI2-lncF(n=1)plasmids.All bla_(NDM-5)-bearing plasmids were transferred by conjugation at frequencies of~10^(-4)-10^(-6).Based on sequence analysis,the lncHI2 plasmid pHNBYF33-1 was similar to other bla_(NDM-5)-carrying lncHI2 plasmids deposited in GenBank from Guangdong ducks.In all lncHI2 plasmids,bla_(NDM-5)was embedded in a novel transposon,Tn7057(IS3000-△ISAba125-IS5-△ISAba125-bla_(NDM-5)-bleMBL-trpF-tat-△dct-IS26-△umuD-△ISKox3-IS3000),which was identical to the genetic structure surrounding bla_(NDM-5)found in some IncX3 plasmids.The lncHI2-lncF hybrid plasmid pHNTH9F11-1 was formed by homologous recombination of the bla_(NDM-5)-carrying lncHI2 plasmid and a heavy-metal-resistant IncF plasmid through△Tn1721 To the best of our knowledge,this is the first report on the characterization of bla_(NDM-5)-bearing plasmids in fish in China.The lncHI2 plasmid pHNBYF33-1 may be transmitted from ducks,considering the common duck-fish freshwater aquaculture system in Guangdong.Tn7051 is likely responsible for the transfer of bla_(NDM-5) from lncX3 to lncHI2 plasmids in Enterobacteriaceae,resulting in the expansion of transmission vectors of bla_(NDM-5).

During a 2018 antimicrobial resistance surveillance of Escherichia coli isolates from diarrheal calves in Xinjiang Province,China,an unexpectedly high prevalence(48.5%)of fosfomycin resistance was observed.This study aimed to reveal the determinants of fosfomycin resistance and the underlying transmission mechanism.Polymerase chain reaction(PCR)screening showed that all fosfomycin-resistant E.coli carried the fosA3 gene.Pulsed-field gel electrophoresis(PFGE)and southern blot hybridization revealed that the 16 fosA3-positive isolates belonged to four different PFGE patterns(i.e.,A,B,C,D).The fosA3 genes of 11 clonally related strains(pattern D)were located on the chromosome,while others were carried by plasmids.Whole-genome and long-read sequencing indicated that the pattern D strains were E.coli O101:H9-ST10,and the pattern C,B,and A strains were O101:H9-ST167,O8:H30-ST1431,and O101:H9 with unknown ST,respectively.Among the pattern C strains,the blaCTX-M-14 gene was co-localized with the fosA3 gene on the F18:A-:B1 plasmids.Interestingly,phylogenetic analysis based on core genome single nucleotide polymorphisms(cgSNPs)showed that the O101:H9-ST10 strains were closely related to a Australian-isolated Chroicocephalus-origin E.coli O101:H9-ST10 strain producing CTX-M-14 and FosA3,with a difference of only 11 SNPs.These results indicate possible international dissemination of the high-risk E.coli clone O101:H9-ST10 by migratory birds.

A privileged strategy has been developed for the precise construction of enantioenriched azaspiro polycyclic scaffolds.Structurally diverse azaspiro polycycles bearing multiple contiguous stereocenters are obtained with excellent results(up to 99:1 e.r.,>95:5 d.r.) via sequential enantioselective four-component Ugi reactions/post-Ugi transformations with substrates containing prerequisite functional groups in the presence of anionic stereogenic-at-cobalt(Ⅲ) complexes.

Effectiveness of Acupoint Application in Patients with Pharyngeal Pain: Evidence from CHUNBO, A Prospective Real-World Study

Objective: To assess the outcomes after acupoint application in patients with pharyngeal pain in a real-world settings, and analyze the characteristics of effective population and prescription characteristics of acupoint application. Methods: Based on CHUNBO platform, patients with pharyngeal pain who were candidates for acupoint application on the basis of physician-evaluation, were enrolled in a nationwide, prospective, 69-week multicenter observational study from August 2020 to February 2022. Propensity score matching(PSM) was used to match the confounding factors and the association rules were used to analyze the characteristics of effective population and prescription characteristics of acupoint application. Outcome assessments included the disappearance rate of pharyngeal pain(within 3, 7, and 14 days), disappearance time of pharyngeal pain, as well as adverse events. Results: Of 7,699 enrolled participants, 6,693(86.9%) received acupoint application and 1,450(21.7%) with non-acupoint application. After PSM, there were 1,004 patients each in the application group(AG) and non-application group(NAG). The disappearance rate of pharyngeal pain in the AG at 3, 7, and 14 days were all higher than those in the NAG(P<0.05). The disappearance time of pharyngeal pain in the AG were shorter than that in the NAG(logrank P<0.001, hazard ratio=1.51, 95% confidence interval: 1.41–1.63). The median age of effective cases was 4 years, mainly 3–6 years old(40.21%). The disappearance rate of pharyngeal pain in the application group with tonsil diseases was 2.19 times higher than that in the NAG(P<0.05). The commonly used acupoints for the effective cases were Tiantu(RN 22), Shenque(RN 8) and Dazhui(DU 14). The commonly used herbs for the effective cases were Natrii sulfas, Radix et Rhizoma Rhei, and Herba Ephedrae. Among them, Natrii sulfas was applied to RN 8 most frequently(support 84.39%). In the research and development of new silicon pixel detectors,a collimated monoenergetic charged-particle test beam equipped with a high-resolution pixel-beam telescope is crucial for prototype verification and performance evaluation.When the beam energy is low,the effect of multiple Coulomb scattering on the measured resolution of the Device Under Test(DUT)must be considered to accurately evaluate the performance of the pixel chips and detectors.This study aimed to investigate the effect of multiple Coulomb scattering on the measured resolution,particularly at low beam energies.Simulations were conducted using Allpix^(2) to study the effects of multiple Coulomb scattering under different beam energies,material budgets,and telescope layouts.The simulations also provided the minimum energy at which the effect of multiple Coulomb scattering could be ignored.Compared with the results of a five-layer detector system tested with an electron beam at DESY,the simulation results were consistent with the beam test results,confirming the reliability of the simulations.

C_(Ar)-Br bond cleavage via cooperative EnT/NHC catalysis: mild access to indolines

The cooperative visible-light mediated and NHC-catalyzed dearomative arylation-acylation of N-(2-bromobenzoyl)indoles with aldehydes was developed, affording the corresponding highly functionalized polycyclic indolines in moderate to good yields with exclusive diastereoselectivities. Mechanism investigations support the activation of CAr-Br bond via visible-light mediated energy transfer(EnT) and the following single-electron-transfer(SET) with Breslow intermediate under NHC catalysis to give key aryl radical and ketyl radical. This protocol demonstrates the cleavage of CAr-Br bond under mild cooperative EnT/NHC catalysis.

Efficient photocatalytic NADH regeneration with Rh-loaded Z-scheme mediator-free system

Nicotinamide adenine dinucleotide(NADH)regeneration is necessary for the sustainable application of enzymatic industry.The Rh-based complex[Cp^(*)Rh(bpy)(H)]+has been widely used as an important mediator in NADH regeneration systems,but it is limited by complexity and high cost.Here,a Z-scheme was constructed by loading Rh onto carbon nitride nanosheets/carbon nitride quantum dots(CN-CNQD).The resultant catalyst achieved a high yield of NADH in a mediator-free(M-free)system of 0.283 mmol L^(−1) g^(−1) min^(−1),which is 5.29 times that of pure CN.ADH enzyme introduction experiments confirmed that the enzyme active product 1,4-NADH could reach 34.21%selectivity in the M-free system.Mechanism research revealed that the heterojunction between CNs and CNQDs improved the NADH regeneration activity in the traditional M-involved system,while Rh loading was proved to optimize the yield and selectivity of 1,4-NADH in M-free system.The immobilized Rh shows more competitiveness than[Cp^(*)Rh(bpy)(H)]^(+).This study contributes to the construction of an M-free system for further application in greener,lower-cost enzymatic processes.

Influence of fiber posts on the fracture resistance of endodontically treated premolars with different dental defects

This study aimed to evaluate the influence of quartz fiber post placement on the fracture resistance of endodontically treated premolars with different dental defects under dynamic loading.Fifty extracted single-rooted mandibular premolars were randomized into five groups.Each group was prepared according to numbers of residual walls ranged from 0 to 4.Then each group was divided into two subgroups with one restored with quartz fiber posts and the other without posts.In no-post groups,gutta percha point 2 mm below cemento-enamel junction was removed.Composite resin was adapted to the well and used to shape the core directly.Each tooth was restored with a complete metal crown.Dynamic loading was carried out in a masticatory simulator with a nominal load of 50 N at 2 Hz for 300 000 loading cycles.Then a quasi-statically load was applied in a universal testing machine 306 to the long axis with a crosshead speed of 1 mm？min21until fracture.Data were analyzed with one-way analysis of variance and pairwise comparison（P,0.05）.No specimens failed during dynamic loading.The fracture resistance enhanced with the increase of numbers of coronal walls and the differences were significant（P,0.05）.Placement of fiber posts had a significant effect when fewer than two walls remained（P,0.05）,but it had no significant influence in groups with two,three or four walls（P.0.05）.Fiber post did not change failure mode,and the fracture pattern was mainly favorable.More dentin walls need to be retained in clinic.When no less than two walls remained,a fiber post is not always necessary.

HepG2.2.15-derived exosomes facilitate the activation and fibrosis of hepatic stellate cells

BACKGROUND The role of exosomes derived from HepG2.2.15 cells,which express hepatitis B virus(HBV)-related proteins,in triggering the activation of LX2 liver stellate cells and promoting liver fibrosis and cell proliferation remains elusive.The focus was on comprehending the relationship and influence of differentially expressed microRNAs(DE-miRNAs)within these exosomes.AIM To elucidate the effect of exosomes derived from HepG2.2.15 cells on the activation of hepatic stellate cell(HSC)LX2 and the progression of liver fibrosis.METHODS Exosomes from HepG2.2.15 cells,which express HBV-related proteins,were isolated from parental HepG2 and WRL68 cells.Western blotting was used to confirm the presence of the exosomal marker protein CD9.The activation of HSCs was assessed using oil red staining,whereas DiI staining facilitated the observation of exosomal uptake by LX2 cells.Additionally,we evaluated LX2 cell proliferation and fibrosis marker expression using 5-ethynyl-2′-deoxyuracil staining and western blotting,respectively.DE-miRNAs were analyzed using DESeq2.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathways were used to annotate the target genes of DE-miRNAs.RESULTS Exosomes from HepG2.2.15 cells were found to induced activation and enhanced proliferation and fibrosis in LX2 cells.A total of 27 miRNAs were differentially expressed in exosomes from HepG2.2.15 cells.GO analysis indicated that these DE-miRNA target genes were associated with cell differentiation,intracellular signal transduction,negative regulation of apoptosis,extracellular exosomes,and RNA binding.KEGG pathway analysis highlighted ubiquitin-mediated proteolysis,the MAPK signaling pathway,viral carcinogenesis,and the toll-like receptor signaling pathway,among others,as enriched in these targets.CONCLUSION These findings suggest that exosomes from HepG2.2.15 cells play a substantial role in the activation,proliferation,and fibrosis of LX2 cells and that DE-miRNAs within these exosomes contribute to the underlying mechanisms.

Influence of the Acoustic Liner in Large Eddy Simulation of Longitudinal Thermoacoustic Instability in a Model Annular Combustor

Although extensive efforts have been made to dampen the thermoacoustic instability,successfully controlling the pressure oscillations in modern gas turbines or aeroengines remains challenging.The influence of the acoustic liner on the longitudinal thermoacoustic mode in a model annular combustor is investigated by Large Eddy Simulation(LES) in this work.The result of the self-excited longitudinal thermoacoustic instability without the liner agrees well with the frequency and acoustic analysis of the pressure mode based on experimental data.Three different bias flow velocities of the liner located downstream of the combustor are then simulated.The results reveal that the existence of the liner influences not only the acoustic field but also the flow field.When the bias velocity is large,it leads to intense turbulence-induced fluctuations,and the pressure oscillation is modulated intermittently.It shows that the weak coupling between flow and pressure oscillations plays a significant role in the onset of the intermittency of a thermoacoustic system.Based on the dynamic analysis of the thermoacoustic system with the acoustic liner,this intermittency is caused by the influence of the flow field on the flame-acoustic coupling.Finally,a low-order modeling method based on Van der Pol(VdP) oscillator with additive stochastic forcing is conducted to reproduce the evolving dynamics of the thermoacoustic system.Although the numerical cases demonstrated in this work are relatively simpler than those in a practical combustion system,the results are helpful for us to understand the effect of the acoustic liner and show the attractive potential to apply this device to suppress thermoacoustic instability.
