BACKGROUND: Triptolide (TPT) is a diterpenoid triepoxide extracted from the Chinese herb Tripterygium wilfordii Hook. F. It exhibits potent immunosuppressive and anti-inflammatory properties. This study was undertaken to investigate its effects on prolongation of islet allograft survival in rodents. Additionally, we investigated whether TPT would be toxic to islet function in vivo. METHODS: We transplanted BALB/c islets to either chemically induced diabetic C57BL/6 mice or spontaneously diabetic non-obese diabetic (NOD) mice. TPT was injected within 2 weeks or continuously, until rejection, in the two combinations. Then, we evaluated the toxicity of TPT on islet function by daily injection to naive BALB/c or diabetic BALB/c that was cured by syngeneic islet transplantation under the kidney capsule. Mice injected with cyclosporine A (CsA) or vehicle served as controls. Intraperitoneal glucose tolerance tests (IPGTTs) performed at 4 and 8 weeks in the naive BALB/c group, and at 2, 4, 6, and 8 weeks in the syngeneic transplanted group. RESULTS: The medium survival time of islets allograft from TPT treated C57BL/6 and NOD recipients were 28.5 days (range 24-30 days, n=10) and 33.0 days (range 15-47 days, n=6), respectively, and they were significantly different from those of the vehicle treated controls, which were 14.0 days (range 13-16 days, n=6) and 5.0 days (range 4-10 days, n=6), respectively (all P<0.0001). The IPGTT demonstrated that there was no difference between the TPT treated and vehicle treated groups, either in the normal or syngeneic transplanted islet BALB/c mice. However, CsA injection impaired islet function in both normal and syngeneic transplanted mice as early as 4 weeks. CONCLUSION: TPT prolonged islets allograft survival in a chemically induced diabetic or an autoimmune diabetic murine model without impairment of islet function. Background In order to acquire a high quality image with a low radiation dose, prospective electrocardiogram （ECG）-triggered computed tomography coronary angiography （CTCA） requires a stable heart rate （HR）〈 65 beats/min. Esmolol has the advantage of reduc-ing HR. The objective of this article is to assess the value of intravenous esmolol treatment before prospective ECG-tr/ggered high-pitch spiral acquisition for CTCA. Methods From March 2013 to june 2013, 313 patients underwent prospective ECG-triggered CTCA. Two hundred and thirty two of them received esmolol before angiography. We retrospectively analyzed clinical characteristics, esmolol dose, radiation exposure dose, and the change in HR and blood pressure in these 232 patients. Results A total of 232 patients with a HR 〉 65 beats/rain before CTCA examination received intravenous esmolol treatment （mean dose of 57.26±15.39 rag）, The mean initial HR （HR1）, slowest HR （HR2）, and the HR 30 min after HR2 （HR3） were 75.06± 5.59, 60.75 ±4.00, and 75.54 ± 5.96 beats/min, respectively （HR1 vs. HR2, P 〈 0.0001; HRI vs. HR3, P = 0.377）. The mean time from esmolol administration to HR2 was 24.25 ± 4.97 s and the mean effective radiation dose was 2.28 ± 0.02 mSv. Conclusions HR could be rapidly controlled at an optimum level with intravenous esmolol before prospective ECG-triggered high-pitch spiral acquisition for CTCA. Consequently, the patients received a very low radiation dose.

The multi-physics instrument(MPI)is the first user cooperative instrument at the China Spallation Neutron Source(CSNS).It was designed to explore the structures of complex materials at multiple scales based on the neutron total scattering *** imposes the requirements for the detector,including a high detection efficiency to reduce the measurement time and a large solid angle coverage to cover a wide range of momentum *** satisfy these demands,a large-area array of 3He-filled linear position-sensitive detectors(LPSDs)was constructed,each with a diameter of 1 inch and pressure of 20 *** uses an orbicular layout of the detector and an eight-pack module design for the arrangement of 3He LPSDs,covering a range of scattering angles from 3°to 170°with a total detector area of approximately 7 *** detector works in air,which is separated from the vacuum environment to facilitate installation and *** characteristics of the MPI detector were investigated through Monte Carlo(MC)simulations using Geant4 and experimental *** results suggest that the detectors are highly efficient in the wavelength range of the MPI,and an efficiency over 25%is achievable for above 0.1 A neutrons.A minimal position resolution of 6.4 mm full width at half maximum(FWHM)along the tube length was achieved at a working voltage of 2200 V,and a deviation below 2 mm between the real and measured positions was attained in the beam *** detector module exhibited good consistency and an excellent counting rate capacity of up to 80 kHz,which satisfied the requirements of experiments with a high event *** of its operation over the past year have shown that the detector works steadily in sample experiments,which allows the MPI to serve the user program successfully.
