This study aims to investigate the role of metal regulatory transcription factor 1(MTF1)-mediated metal response in cadmium(Cd)-induced cerebellar injury,and to evaluate the antagonistic effects of nanoselenium(Nano-Se)against Cd toxicity.A total of 80 chicks(1 d old,male,Hy-Line Variety White)were randomly allocated to 4 treatment groups for 3 months:the control group(fed with a basic diet,n=20),the Nano-Se group(basic diet with 1 mg/kg nano-Se 1 mg/kg Nano-Se in basic diet,n=20),the NanoSe+Cd group(basic diet with 1 mg/kg Nano-Se and 140 mg/kg Cd Cl_(2),n=20)and the Cd group(basic diet with 140 mg/kg Cd Cl_(2),n=20).The results of the experiment showed that the Purkinje cells were significantly decreased with their degradation and indistinct nucleoli after Cd exposure.Moreover,exposure to Cd caused a significant accumulation of Cd and cupper.However,the contents of Se,iron,and zinc were decreased,thereby disturbing the metal homeostasis in the cerebellum.The Cd exposure also resulted in high levels of malondialdehyde(MDA)and down regulation of selenoprotein transcriptome.Furthermore,the expressions of MTF1,metallothionein 1(MT1),MT2,zinc transporter 3(ZNT3),ZNT5,ZNT10,zrt,irt-like protein 8(ZIP8),ZIP10,transferrin(TF),ferroportin 1(FPN1),ATPase copper transporting beta(ATP7B),and copper uptake protein 1(CTR1)were inhibited by Cd exposure.However,all these changes were significantly alleviated by the supplementation of Nano-Se.This study proved that Cd could disorder metal homeostasis and induce oxidative stress,whereas Nano-Se could relieve all these negative effects caused by Cd via activating the MTF1-mediated metal response in the cerebellum of chicken.

Planetary health risks in urban agriculture

Urban agriculture is gaining recognition for its potential contributions to environmental resilience and climate change adaptation,providing advantages such as urban greening,reduced heat island effects,and decreased air pollution.Moreover,it indirectly supports communities during weather events and natural disasters,ensuring food security and fostering community cohesion.However,concerns about planetary health risks persist in highly urbanized and climate-affected areas.Employing electronic databases such as Web of Science and PubMed and adhering to Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines,we identified 55 relevant papers to comprehend the planetary health risks associated with urban agriculture,The literature review identified five distinct health risks related to urban agriculture:(1)trace metal risks in urban farms;(2)health risks associated with wastewater irrigation;(3)zoonotic risks;(4)other health risks;and(5)social and economic risks.The study highlights that urban agriculture,while emphasizing environmental benefits,particularly raises concerns about trace metal bioaccumulation in soil and vegetables,posing health risks for populations.Other well studied risks included wastewater irrigation and backyard livestock farming.The main limitations in the available literature were in studying infectious diseases and antibiotic resistance associated with urban agriculture.

Design and Implementation of Hand Gesture Detection System Using HM Model for Sign Language Recognition Development

Gesture detection is the primary and most significant step for sign language detection and sign language is the communication medium for people with speaking and hearing disabilities. This paper presents a novel method for dynamic hand gesture detection using Hidden Markov Models (HMMs) where we detect different English alphabet letters by tracing hand movements. The process involves skin color-based segmentation for hand isolation in video frames, followed by morphological operations to enhance image trajectories. Our system employs hand tracking and trajectory smoothing techniques, such as the Kalman filter, to monitor hand movements and refine gesture paths. Quantized sequences are then analyzed using the Baum-Welch Re-estimation Algorithm, an HMM-based approach. A maximum likelihood classifier is used to identify the most probable letter from the test sequences. Our method demonstrates significant improvements over traditional recognition techniques in real-time, automatic hand gesture recognition, particularly in its ability to distinguish complex gestures. The experimental results confirm the effectiveness of our approach in enhancing gesture-based sign language detection to alleviate the barrier between the deaf and hard-of-hearing community and general people.

Klippel Trenaunay Syndrome with Angiokeratoma Circumscriptum Naeviforme and Bilateral Congenital Anorchia: A Rare Association

Klippel-Trenaunay syndrome (KTS) is not a common congenital vascular abnormality. A trio of capillary malformation, venous varicosities, and bony or soft-tissue hypertrophy define this syndrome. Significant morbidities associated with this illness include bleeding, deep vein thrombosis, and embolic consequences. Angiokeratoma circumscriptum naeviforme (ACN) is indeed a congenital variant of angiokeratoma that appears as a hyperkeratotic plaque on the lower extremity. Bilateral congenital anorchia (BCA) is the total lack of testicular tissue in a male with a normal phenotype and karyotype. KTS has been linked to ACN. Here we presented an 8-year-old male child who came with a swollen left thigh and the right side of his face with overlying blackish nodules on his left thigh and scrotum. The patient was diagnosed as KTS with angiokeratoma circumscriptum naeviforme and bilateral congenital anorchia based on his history, imaging studies and the typical clinical features of the disease.

Serum Zinc and Copper Level in Juvenile Idiopathic Arthritis (JIA) Patients and Its Correlation with Disease Duration-A Tertiary Hospital Study

Background: Juvenile Idiopathic Arthritis (JIA) is the most prevalent rheumatic disease in children. It is associated with abnormal levels of serum zinc (Zn) and copper (Cu) as during inflammation serum copper concentration increases and zinc decreases. Objective: To assess the serum Zn and Cu levels in different sub-types of JIA patients and their correlation with the disease duration. Methods: This cross-sectional study was conducted over twelve months at the Pediatric Rheumatology Division, Department of Paediatrics, Bangabandhu Sheikh Mujib Medical University. Sixty-nine JIA cases that fulfilled the International League of Association for Rheumatology (ILAR) criteria were taken as cases and age and sex-matched healthy children were considered as controls. The serum Zn and Cu tests were done using the spectrophotometric method with INDIKO PLUS Drug Analyzer. Data were recorded in a pre-designed questionnaire. Data were checked, verified and analyzed manually where continuous variables were analyzed using unpaired t-test and categorical variables using the ANOVA test. Pearson’s correlation coefficient test was used to see the correlation of serum zinc and copper levels with disease duration. Results: Boys were predominant in both case and control groups, with the majority within the 10 to 16-year-age group. Enthesitis-related arthritis (ERA) was the most common subtype followed by sJIA, Oligo JIA, Poly JIA (RF-) and unclassified subtypes. Disease duration was found less than 12 months in 30.4% of JIA patients. Serum analysis revealed a statistically significant reduction in mean zinc levels and increased copper levels in JIA patients compared to controls. This study observed a negative correlation between serum zinc levels and disease duration, whereas serum copper levels exhibited a positive correlation with disease duration. Acute Lymphoblastic Leukemia Presenting as Systemic Juvenile Idiopathic Arthritis: Experience from Bangladesh

Background: Acute lymphoblastic leukemia (ALL), the most common paediatric malignancy, is a heterogeneous hematologic disease. ALL patients may present with isolated and persistent osteo-articular complaints, lower incidence of hepatomegaly, splenomegaly or lymphadenopathy without clear laboratory features, and misdiagnosed as systemic juvenile idiopathic arthritis (sJIA). Methods: This was a single center cross sectional study over a period of 4 years. Clinic laboratory profiles of 39 ALL children were compared with 39 age and sex-matched sJIA cases. Result: Among 39 ALL patients 89.7% were initially misdiagnosed as sJIA upon clinical presentation. Majority (66.7%) of ALL patients had oligo-articular joint involvement. In sJIA, small joints of the hands were most commonly involved. The total WBC count was significantly higher in ALL patients (p-value 0.0065). CRP and LDH values between the two groups showed significant differences (p-value 0.00006 and 0.00001 respectively). Conclusion: The presentation of leukemia with arthralgia or arthritis makes the diagnosis difficult for the physicians. The diagnosis of sJIA must be made with caution keeping the possibility of haematological malignancy in mind.

目的镉(Cadmium,Cd)是环境中广泛存在蓄积性强的重金属污染物,被美国毒物管理委员会列为第六位危及动物健康的有毒物质,已严重危害到了生态环境和人类健康。环境生态系统通过生物富集作用,将土壤中的镉通过食物链引入到动物体内,放大了镉的毒害作用,特别对生态系统中的野生鸟类及家禽危害更为严重。目前研究表明,镉可通过血脑屏障,损伤神经系统,导致认知功能障碍等神经系统疾病发生,但机制尚不明确。本研究的目的是揭示镉暴露对禽类大脑毒性损伤机理。材料和方法本研究选用80只1日龄海兰白种鸡,随机分为4组:对照组、35 mg/kg CdC12、70mg/kg CdC12和140mg/kg CdC12组,饲养90天。观察大脑病理组织学变化,检测组织中金属元素含量及其转运蛋白的表达水平、氧化应激参数变化、MTF1金属反应、血脑屏障、Wnt/β-catenin信号通路、内质网应激、自噬和凋亡相关标志物水平。结果组织病理学评价和尼氏染色结果显示,镉暴露致大脑组织神经元肿胀变性、数量减少,轴突和树突缺失,细胞质空泡变性,纹状体损伤,毛细血管直径增宽,内质网和线粒体肿胀,自噬小体形成;镉暴露致大脑组织氧化还原稳态失衡,ROS、MDA和HO水平升高,GPX、CAT、T-SOD活性和T-AOC水平降低,引发氧化应激;镉暴露致大脑组织金属转运蛋白(DMT1、ZIP8、ZIP10、ZNT3、ZNT5和ZNT6)表达上调,转录因子MTF1及其下游靶点(MT1、MT2、FPN1、ATOX1和XIAP)的表达下调,导致大脑镉积累增加和其他金属(锌,铁,硒,铬,钼,铅)浓度的改变;转录组分析表明,Cx43可能是介导镉诱导星形胶质细胞向A1亚型的转化的潜在靶点;基因集富集分析显示Wnt/β-catenin信号转导轴减弱,降低Wnt7A、FZD 4和β-catenin的蛋白表达,上调TNF-α表达,抑制IL-10表达,诱导炎症的产生和BBB功能障碍,表现为受损的紧密连接和粘附连接的形成;镉暴露致大脑组织GRP78、PERK、eIF-2α表达上调,触发内质网应激;激活Mfn2、PINK1,促进自噬小体形成,Beclin1、LC3B、p62表达上调,诱导鸡脑组织自噬;镉暴露激活大脑神经元细胞Caspase依赖性凋亡信号通路,表明镉暴露诱导神经元凋亡。结论镉暴露会导致金属稳态失衡,鸡大脑组织形态和细胞器损伤,金属转运蛋白和MTF1相关因子异常表达,引发内质网应激、自噬、神经炎症应答和BBB功能障碍。本研究阐明了镉致鸡大脑毒性效应机理,为后续筛选缓解镉致鸡神经毒性作用靶点提供了理论依据。

Growth phase dependent changes in the structure and protein composition of nucleoid in Escherichia coli

The genomic DNA of bacteria is highly compacted in a single or a few bodies known as nucleoids. Here, we have isolated Escherichia coli nucleoid by sucrose density gradient centrifugation. The sedimentation rates, structures as well as protein/DNA composition of isolated nucleoids were then compared under various growth phases. The nucleoid structures were found to undergo changes during the cell growth; i. e., the nucleoid structure in the stationary phase was more tightly compacted than that in the exponential phase. In addition to factor for inversion stimulation(Fis), histone-like nucleoid structuring protein(H-NS), heat-unstable nucleoid protein(HU) and integration host factor(IHF) here we have identified, three new candidates of E. coli nucleoid, namely DNA-binding protein from starved cells(Dps), host factor for phage Qβ(Hfq) and suppressor of td- phenotype A(Stp A). Our results reveal that the major components of exponential phase nucleoid are Fis, HU, H-NS, Stp A and Hfq, while Dps occupies more than half of the stationary phase nucleoid. It has been known for a while that Dps is the main nucleoid-associated protein at stationary phase. From these results and the prevailing information, we propose a model for growth phase dependent changes in the structure and protein composition of nucleoid in E. coli.

Oblique collisional effects of dust acoustic waves in unmagnetized dusty plasma

Effects of oblique collisions of the dust acoustic(DA)waves in dusty plasma are studied by considering unmagnetized fully ionized plasma.The plasma consists of inertial warm negatively charged massive dusts,positively charged dusts,superthermal kappa distributed electrons,and isothermal ions.The extended Poincaré–Lighthill–Kuo(e PLK)method is employed for the drivation of two-sided Korteweg–de Vries(KdV)equations(KdVEs).The Kd V soliton solutions are derived by using the hyperbolic secant method.The effects of superthermality index of electrons,temperature ratio of isothermal ion to electron,and the density ratio of isothermal ions to negatively charged massive dusts on nonlinear coefficients are investigated.The effects of oblique collision on amplitude,phase shift,and potential profile of right traveling solitons of DA waves are also studied.The study reveals that the new nonlinear wave structures are produced in the colliding region due to head-on collision of the two counter propagating DA waves.The nonlinearity is found to decrease with the increasing density ratio of ion to negative dust in the critical region.The phase shifts decrease(increase)with increasing the temperature ratio of ion to electron(κe).The hump(compressive,κeκec)solitons are produced depending on the angle(θ)of oblique collision between the two waves.

The Methodology of Probe Design with Better Resolution and Less Resistive Donut Probe to Achieve the Best Performance

Probes are the interface between microsystems and bio-cells.The ideal interface is one-to-one interface.Though various research groups have been able to establish some sort of interfaces after many years of research,they are very crude.Neurons are millions in numbers,whereas the prostheses successfully built so far have only a few hundred probes at best.Creating an ef- fective interface is still far away.Though we have micro-and nano-technologies,we couldn't build a prosthesis with an effective resolution.Main reasons behind it are the type of probe being used and the poor design of the probe.To address this problem,we developed a methodology to design a probe and an array of probes with better resolution and less resistive donut probe.This methodology helps us to design a probe optimizing all the parameters.We presented our methodology through a design that is capable of 70 μm penetration inside the tissue.The tissue heating by our designed probe is only 0.411 °C.We also characterized the donut probe,which could be used by any research group to design a donut probe of their specific need.
