Role of retinoids in the prevention and treatment of colorectal cancer

Vitamin A and its derivatives, retinoids, have been widely studied for their use as cancer chemotherapeutic agents. With respect to colorectal cancer(CRC), several critical mutations dysregulate pathways implicated in progression and metastasis, resulting in aberrant Wnt/β-catenin signaling, gain-of-function mutations in K-ras and phosphatidylinositol-3-kinase/Akt, cyclooxygenase-2 over-expression, reduction of peroxisome proliferatoractivated receptor γ activation, and loss of p53 function. Dysregulation leads to increased cellular proliferation and invasion and decreased cell-cell interaction and differentiation. Retinoids affect these pathways by various mechanisms, many involving retinoic acid receptors(RAR). RAR bind to all-trans-retinoic acid(ATRA) to induce the transcription of genes responsible for cellular differentiation. Although most research concerning the chemotherapeutic efficacy of retinoids focuses on the ability of ATRA to decrease cancer cell proliferation, increase differentiation, or promote apoptosis; as CRC progresses, RAR expression is often lost, rendering treatment of CRCs with ATRA ineffective. Our laboratory focuses on the ability of dietary vitamin A to decrease CRC cell proliferation and invasion via RAR-independent pathways. This review discusses our research and others concerning the ability of retinoids to ameliorate the defective signaling pathways listed above and decrease tumor cell proliferation and invasion through both RAR-dependent and RAR-independent mechanisms.

Oxidative stress is prevalent among infertile men and is a significant cause of sperm DNA *** sperm DNA damage may reduce embryo quality and increase miscarriage rates,it is possible that untreated sperm oxidative stress may impair in vitro fertilization(IVF)live birth *** that the antioxidant Menevit is reported to reduce sperm DNA damage,it was hypothesized that men’s consumption of this supplement may alter IVF ***,a retrospective cohort study was conducted analyzing outcomes for couples undergoing their first fresh embryo *** were classified as controls if they were taking no supplements,health conscious controls if taking“general health”supplements,or Menevit *** with karyotype abnormalities,or cycles using donated,frozen and surgically extracted sperm were *** the final study cohort of 657 men,live birth rates were significantly higher in Menevit users than controls(multivariate adjusted odds ratio[OR]:1.57,95%confidence interval[Cl]:1.01-2.45,P=0.046),but not between controls taking no supplements and those using general health supplements,thereby suggesting that potential health conscious behavior in supplement users is unlikely responsible for the superior outcomes in Menevit ***,in a post hoc sensitivity analysis,live birth rates among Menevit users were statistically superior to controls for lean men(OR:2.73,95%Cl:1.18-6.28;P=0.019),not their overweight/obese counterparts(OR:1.29,95%Cl:0.75-2.22,P=0.37).The results of this large cohort study therefore support a positive association between men’s use of the Menevit antioxidant during IVF treatment and live birth rates,especially in lean individuals.

Adaptation Decision Support: An Application of System Dynamics Modeling in Coastal Communities

This research develops and applies a system dynamics(SD) model for the strategic evaluation of environmental adaptation options for coastal communities. The article defines and estimates asset-based measures for community vulnerability, resilience, and adaptive capacity with respect to the environmental, economic, social, and cultural pillars of the coastal community under threat. The SD model simulates the annual multidimensional dynamic impacts of severe coastal storms and storm surges on the community pillars under alternative adaptation *** calculation of the quantitative measures provides valuable information for decision makers for evaluating the alternative strategies. The adaptation strategies are designed model results illustrated for the specific context of the coastal community of Charlottetown, Prince Edward Island, Canada. The dynamic trend of the measures and model sensitivity analyses for Charlottetown—facing increased frequency of severe storms, storm surges, and sea-level rise—provide impetus for enhanced community strategic planning for the changing coastal *** research is presented as part of the International Community-University Research Alliance C-Change project ‘‘Managing Adaptation to Environmental Change in Coastal Communities: Canada and the Caribbean'' sponsored by the Social Science and Humanities Research Council of Canada and the International Development Resource Centre.

An Example Using Improved Lefschetz Duality

A theorem of Lambrechts and Stanley is used to find the rational cohomology of the complement of an embedding S^(4n-1)→ S^(2n)× S^m as a module and demonstrate that it is not necessarily determined by the map induced on cohomology by the embedding, nor is it a trivial extension. This demonstrates that the theorem is an improvement on the classical Lefschetz duality.

Understanding p53 functions through p53 antibodies

TP53 is the most frequently mutated gene across all cancer types. Our understanding of its functions has evolved since its discovery four decades ago. Initially thought to be an oncogene, it was later realized to be a critical tumour suppressor. A significant amount of our knowledge about p53 functions have come from the use of antibodies against its various forms. The early anti-p53 antibodies contributed to the recognition of p53 accumulation as a common feature of cancer cells and to our understanding of p53 DNA-binding and transcription activities. They led to the concept that conformational changes can facilitate p53’s activity as a growth inhibitory protein. The ensuing p53 conformational-specific antibodies further underlined p53’s conformational flexibility, collectively forming the basis for current efforts to generate therapeutic molecules capable of altering the conformation of mutant p53. A subsequent barrage of antibodies against post-translational modifications on p53 has clarified p53’s roles further, especially with respect to the mechanistic details and context-dependence of its activity. More recently, the generation of p53 mutation-specific antibodies have highlighted the possibility to go beyond the general framework of our comprehension of mutant p53-and promises to provide insights into the specific properties of individual p53 mutants. This review summarizes our current knowledge of p53 functions derived through the major classes of anti-p53 antibodies, which could be a paradigm for understanding other molecular events in health and disease.

The verbiage in variable message signs and traffic diversion during crash incidents

Purpose–WIth limited research on the effects of variable message sign(VMS)message content and verbiage on revealed driver behavior,this study aims to investigate how different verbiage of crash-related messages are related to the diversion ***/methodology/approach–Using ordered logit models,the associations of message verbiage with diversion rates during crash incidents were assessed using five years of VMS message history within a section of I-15 in the state of ***–A significant impact of message verbiage on the diversion rate was *** on the analysis results,the crash message verbiage with the highest diversion was found to be miles to crash 1“prepare to stop,”followed by crash location 1 delay information,miles to crash 1“use caution”1 lane of the crash,*** addition,the diversion rate was found to be correlated to some roadway characteristics(*** in mainline,weather condition and light condition)along with the temporal *** limitations/implications–These findings could be used by transportation agencies(*** department of transportation[DOTs])to make informed decisions about choosing the message verbiage during future crash *** study also revealed that higher diversion rates are associated with a shorter distance between the crash location and VMS device location,recommending increasing the number of VMS devices,particularly in crash-prone areas.

From Democracy to Bureaucracy： The Baojia in Nationalist Thought and Practice, 1927-1949

In the 1939 New County Reforms, the Nationalist government made the baojia system the lowest level of self-government in the country. This decision was the result of more than ten years of discussion among Nationalist administrators and writers who were searching for a tutelary system to train the people in their political rights in preparation for constitutional rule. In the 1920s and 1930s, Nationalist writers claimed to be following Sun Zhongshan＇s （Sun Yat-sen） philosophy by reinventing the baojia as a form of democracy. Harkening back to a reimagined national past, they ＂discovered＂ that the imperial baojia was not a system of local control, but a traditional model of bureaucratically-designed local self-government. Nationalist writers dovetailed this new baojia with Sun Zhongshan＇s philosophy in order to rationalize its position as the foundation of the Three Principles of the People State. Once philosophically legitimized, Nationalist writers endorsed the baojia as a top-down bureaucratic system that would transform the political, social, and economic life of the country; it would become the core political unit of their state-making and nation-building projects. In so doing, the baojia came to represent the Nationalists＇ deeply-held belief in the power of human agency to create state institutions capable of entirely remaking society and transforming the nation.

While robotic surgery has shown clear utility and advantages in the adult population,its role in pediatrics remains ***-sized robotic instruments and equipment are not readily available yet,so certain modifications can be made in order to make robotic surgery successful in *** the cost of robotic surgery remains high compared to open procedures,patients experience greater satisfaction and quality of life with robotic *** pyeloplasty is a standard of care in older children,and has even been performed in infants and re-do *** robotic procedures performed in children include heminephroureterectomy,ureteroureterostomy,ureteral reimplantation,urachal cyst excision,bladder diverticulectomy,and bladder reconstructive procedures such as augmentation,appendicovesicostomy,antegrade continence enema,bladder neck reconstruction and sling,as well as other *** surgery has also been used in oncologic cases such as partial nephrectomy and retroperitoneal lymph node *** improvements in technology with production of pediatric-sized robotic instruments,along with increases in robotictrained pediatric urologists and surgeon experience along each’s learning curve,will help to further advance the field of robotic surgery in pediatric urology.
