Prevalence of autoantibodies against cellular antigens in patients with HIV and leprosy coinfection in the Amazon region

Background:Infectious agents can activate self-reactive T *** general,infections trigger various mechanisms,including a lack of auto-tolerance,induction of costimulatory molecules on antigen presenting cells,and molecular simulation,in addition to cross-reactions between microbial antigens and *** and leprosy coinfections lead to self-immunity with the production of ***,not enough data on the immune behaviour associated with this coinfection are ***,this study focused on the detection of autoantibodies against cellular antigens(AACA)in individuals with HIV and leprosy coinfection in the Amazon ***:Patients were distributed into four groups according to their infections:(i)coinfection with HIV and leprosy(n=23),(ii)infection with leprosy(n=33),(iii)infection with HIV/AIDS(n=25),and(iv)healthy blood donor controls(n=100).AACA were identified by indirect immunofluorescence and the samples were tested using a commercial diagnosis kit containing the antinuclear antibody ***:Morphologically,all stages of cell division were assessed in addition to the morphological features associated with the nuclear matrix,nucleolus,mitotic spindle,and *** was a high prevalence of AACA in the coinfection group(47.8%,n=11)when compared with the control group of healthy blood donors(2.0%).The results showed predominantly cytoplasmic staining in all groups analysed,and no difference was observed between the presence or absence of AACA and the leprosy forms(paucibacillary and multibacillary)in the coinfection ***:The results of this study show that despite the tendency of coinfected patients to have higher levels of autoantibodies,no correlation was observed between clinical and laboratorial variables and morbidity associated with HIV and leprosy coinfections or the levels of AACA in the serum of coinfected *** data are important to elucidate this complex relationship between HIV a

Objective:To study the behavioral effects of semi purified fraction prepared from the stem bark of Erythrina velutina on ***:Three chloroformic fractions(CA) at 50 or 100 mg/kg(CA1,CA2 and CA3) were tested in two behavioral tests;plus maze and open ***:In the plus maze test,it was observed a decrease in the number of entries in the closed arms(NECA) CA2 at both doses as compared to the control *** the open field test,CA2 and CA3 fractions decreased the number of squares crossed and the time of permanence on the bar,increased the number of falls at both doses in the rota rod *** CA2 fraction decreased the NECA,but this data did not characterize an anxiolytic *** was the consequence from decrease of locomotor activity(LA) observed in the open field test,that the decrease of LA and rearing,the number of falls and the decrease in time of permanence on the bar in rota rod test were observed after treatment with CA2 and CA3 at both doses,suggesting sedative ***:The present work demonstrated that CA2 and CA3 fractions have impairing effect on LA and motor coordination.

Purpose: The aim of this study is to analyze the epidemiological profile of the oral squamous cell carcinoma cases diagnosed on the oral and facial disease public diagnose system。Methods: This is a cross-sectional study, which object of study was the histopatogical reports。The sample was constituted by 81 oral cancer cases diagnosed between January 1st 2005 and December 31st 2007 at the Mato Grosso State public laboratory (MT Laboratory, Brazil)。From those, only the oral squamous cell carcinoma cases were selected, and the following variables were analyzed: histological gradation, anatomic site, patient's age, gender and local of origin (capital or interior of State)。Results: Oral squamous cell carcinoma represented the most prevalent histological type in the universe of all diagnosed oral cancer, with 81% of the cases。The men represented 79% of the oral squamous cell carcinoma diagnosis, mainly in the ages ranging from 41 to 60 years。The majority of the patients were from inner cities of Mato Grosso State。The most attacked anatomic site was the tongue with histologically moderately differenced lesions。Conclusion: It was possible to conclude with this study that the oral squamous cell carcinoma in Mato Grosso State has an epidemiological profile close to those verified in other countries, being more incident in men at productive age that are from rural areas。

Purpose: To analyze the register of oral cancer cases diagnosed on public laboratory of Mato Grosso State。Methods: It's a longitudinal epidemiologic study。The studied object was the histopathological award, being 1.324 emitted awards between January 2005 and December 2006。The oral cancer cases were verified through the analysis of following variables: percentage of oral cancer among oral lesions, histological type, age, and the local where the patients come fi'om (interior or capital of State)。Results: After analysis of data, 44 lesions of oral cancer were found, representing 3% of all diagnosis。The histological type more incident was oral squamous cell carcinoma。The majority of diagnosis was referent to men with age between 5th and 6th decade of life that are resident in interior of State。Conclusions: We can conclude with this study that during the period of two years of historic series, the public service of pathology registered a considerable number of oral cancer cases。

It is due to the need to ensure the security and integrity of equipment, that the non-destructive tests have been increasingly used in the industrial sector. Among these, the ultrasonic pulse echo technique is the most used in industry, mainly for its simplicity and efficiency. With one transducer only, it is possible to emit the ultrasonic and receive the echo pulse. The ANNs （artificial neural networks） are artificial intelligence techniques that, when properly trained, align themselves to inspection tests becoming a powerful tool in the detection and fault identification. In this work, the echo pulse technique was used to detect discontinuities in welds, where ANNs were fed from the information obtained by digital signal processing techniques （Fourier transform）, to identify and classify three distinct classes of defects. Results showed that with the combination of feature extraction by Fourier transformation and classification with neural networks, it is possible to obtain an automatic defect detection system in welded joints with average efficiency.

The Discovery and Development of Boceprevir:A Novel, First-generation Inhibitor of the Hepatitis C Virus NS3/4A Serine Protease

An estimated 2–3%of the world's population is infected with hepatitis C virus (HCV), making it a major global health problem. Consequently, over the past 15 years, there has been a concerted effort to understand the pathophysiology of HCV infection and the molecular virology of replication, and to utilize this knowledge for the development of more effective treatments. The virally encoded non-structural serine pro-tease (NS3) is required to process the HCV polyprotein and release the individual proteins that form the viral RNA replication machinery. Given its critical role in the replication of HCV, the NS3 protease has been recognized as a potential drug target for the development of selective HCV therapies. In this review, we describe the key scientific discoveries that led to the approval of boceprevir, a first-generation, selective, small molecule inhibitor of the NS3 protease. We highlight the early studies that reported the crystal structure of the NS3 protease, its role in the processing of the HCV polyprotein, and the structural requirements critical for substrate clea-vage. We also consider the novel attributes of the NS3 protease-binding pocket that challenged development of small molecule inhibitors, and the studies that ultimately yielded milligram quantities of this enzyme in a soluble, tractable form suitable for inhibitor screening programs. Finally, we describe the discovery of boceprevir, from the early chemistry studies, through the development of high-throughput assays, to the phase Ⅲ clinical development program that ultimately provided the basis for approval of this drug. This latest phase in the development of boceprevir represents the culmination of a major global effort to under-stand the pathophysiology of HCV and develop small mole-cule inhibitors for the NS3 protease.
