External quality control and training of semen analysis in the Netherlands:starting point for further reduction of outcome variability

Semen analysis is characterized by high levels of intra-and inter-laboratory variability,due to a low level of standardization,high subjectivity of the assessments,and problems with automated *** improve consistency of laboratory results,quality control and training of technicians are important *** goals of this study are to evaluate the results of an external quality control(EQC)program and standardized training by ESHRE Basic Semen Analysis Courses(BSAC)on the variability in manual assessments of semen *** performed retrospective analyses of(1)the interlaboratory variability in the Dutch EQC program and(2)the interobserver variability in BSACs for concentration,motility,and morphology *** data showed that the interlaboratory coefficient of variation(CV)for concentration assessment decreased(range from 24.0%–97.5%to 12.7%–20.9%)but not for morphology and motility *** variability was lower if improved Neubauer hemocytometers were *** assessment showed highest CVs(up to 375.0%),with many outliers in the period of 2007–*** BSAC,a significant reduction of interobserver variability could be established for all parameters(pprogram for motility and morphology might be explained by respectively the facts that motility assessment was introduced relatively late in the EQC program(since 2013)and that criteria for morphology assessment changed in *** results might have been influenced by the pretraining level of participants and the influence of external *** EQC and training show positive effects on reducing *** willingness by laboratories to change their methods toward standards may lead to further improvements.

The accurate description of friction is critical in the finite element(FE)simulation of the sheet metal forming ***,friction is oversimplified through the use of a constant Coulomb friction *** this study,the application of an existing multiscale friction model is extended to the hot stamping *** model accounts for the effects of tool and sheet metal surface topography as well as the evolution of contact pressure,temperature,and bulk strain during hot *** load flattening and strip drawing experiments are performed to calibrate the *** results show that the model can relatively well predict friction in strip draw experiments when the tool surface evolution due to wear is ***,the application of the formulated multiscale friction model was demonstrated in the FE simulation of a hot-stamped part.

Background:An extraordinary long-term running performance may benefit from low dynamic loads and a high load-bearing *** extraordinary runner(age=55 years,height=1.81 m,mass=92 kg) scheduled a marathon a day for 100 consecutive *** running biomechanics and bone density were investigated to better understand successful long-term running in the master ***:Overground running gait analysis and bone densitometry were conducted before the marathon-a-day challenge and near its *** case’s running biomechanics were compared pre-challenge to 31 runners who were matched by a similar foot strike ***:The case’s peak vertical loading rate(Δx=-61.9 body weight(BW)/s or-57%),peak vertical ground reaction force(Δx=-0.38 BW or-15%),and peak braking force(Δx=-0.118 BW or-31%) were remarkably lower(pp at~3.3 m/*** relatively low loading-related magnitudes were attributed to a remarkably high duty factor(0.41) at the evaluated *** foot strike angle of the marathoner(29.5°) was greater than that of the control group,affecting the peak vertical loading *** powers in the lower extremity were also remarkably low in the case ***:peak power of knee absorption(Δx=-9.16 watt/kg or-48%) and ankle generation(Δx=-3.17 watt/kg or-30%).The bone mineral density increased to 1.245 g/cm;(+2.98%) near completion of the challenge,whereas the force characteristics showed no statistically significant ***:The remarkable pattern of the high-mileage runner may be useful in developing or evaluating load-shifting strategies in distance running.

Malignant bone tumors are usually treated by resection of tumor tissue followed by filling of the bone defect with bone graft ***(pMMA)cement is the most commonly used bone substitute in clinical orthopedics in view of its ***,the dense nature of pMMA renders this biomaterial unsuitable for local delivery of chemotherapeutic drugs to limit the recurrence of bone ***,we introduce porosity into pMMA cement by adding carboxymethylcellulose(CMC)to facilitate such local delivery of chemotherapeutic drugs,while retaining sufficient mechanical properties for bone reconstruction in load-bearing *** results show that the mechanical strength of pMMA-based cements gradually decreases with increasing CMC *** incorporation of≥3%CMC,the pMMA-based cements released up to 18%of the loaded cisplatin,in contrast to cements containing lower amounts of CMC which only released less than 2%of the cisplatin over 28 *** release of cisplatin efficiently killed osteosarcoma cells in vitro and the fraction of dead cells increased to 91.3%at day 7,which confirms the retained chemotherapeutic activity of released cisplatin from these pMMA-based ***,tibias filled with pMMA-based cements containing up to 3%of CMC exhibit comparable compressive strengths as compared to intact *** conclusion,we demonstrate that pMMA cements can be rendered therapeutically active by introducing porosity using CMC to allow for release of cisplatin without compromising mechanical properties beyond critical *** such,these data suggest that our dual-functional pMMA-based cements represent a viable treatment option for filling bone defects after bone tumor resection in load-bearing sites.

Delivery of a peptide(App96-110),derived from amyloid precursor protein(App),has been shown to elicit neuroprotective effects following cerebral stroke and traumatic brain *** this study,the effect of App96-110 or a mutant version of this peptide(mApp96-110)was assessed following moderate(200 kdyn,(2 N))thoracic contusive spinal cord injury(SCI)in adult Nude *** received a single tail vein injection of App96-110 or mApp96-110 at 30 minutes post-SCI and were then assessed for functional improvements over the next 8 weeks.A cohort of animals also received transplants of either viable or non-viable human mesenchymal stromal cells(hMSCs)into the SC lesion site at one week post-injury to assess the effect of combining intravenous App96-110 delivery with hMSC *** were perfused 8 weeks post-SCI and longitudinal sections of spinal cord analyzed for a number of factors including hMSC viability,cyst size,axonal regrowth,glial reactivity and macrophage *** of sensorimotor function revealed occasional significant differences between groups using Ladderwalk or Ratwalk tests,however there were no consistent improvements in functional outcome after any of the ***96-110 alone,and App96-110 in combination with both viable and non-viable hMSCs significantly reduced cyst size compared to SCI *** treatments with donor hMSCs also significantly increased βIII tubulin^(+),glial fibrillary acidic protein(GFAp^(+))and laminin+expression,and decreased ED1^(+)expression in *** preliminary study demonstrates that intravenous delivery of App96-110 peptide has selective,modest neuroprotective effects following SCI,which may be enhanced when combined with hMSC ***,the effects are less pronounced and less consistent compared to the protective morphological and cognitive impact that this same peptide has on neuronal survival and behaviour after stroke and traumatic brain *** while the efficacy of a
