Antiproliferative and apoptosis-inducing potential of 3β-hydroxy-△5-steroidal congeners purified from the soft coral Dendronephthya putteri

The exploration and identification of antiproliferative phytochemicals have received increased attention in medicinal chemistry. In particular, research focused on the toxicology of marine natural products has increased in recent years. Terpenoids, among many secondary metabolites, have been demonstrated to act as effective anticancer agents. Soft corals, a group of marine invertebrates, produce a variety of terpenoids with biofunctional properties. The current study presents the extraction, purification, and identification of sterol congeners from the soft coral Dendronephthya putteri. The method involves 50% chloroform-methanol extraction, polar column fractionation, and analysis through GC-MSn. Dose-dependent antiproliferative activity was observed within the sterol-rich fraction (DPCMH 2-4), which consisted of 3β-hydroxy-Δ5-steroidal congeners. This fraction inhibited the growth of HL-60 and MCF-7 cells with IC50 values of 25.27±1.43 and 22.81±0.15 μg/mL, respectively. Apoptotic body formation, DNA damage, cell cycle arrest, and apoptotic cell signaling pathway activation were also observed, reinforcing the dose-dependent antiproliferative and apoptosis-inducing activity of 3β-hydroxy-Δ5-steroidal congeners. To our knowledge, this is the first report of anticancer agent identification from the soft coral D. putteri. Based on the observations, these steroidal congeners are promising candidates for the development of anticancer drugs.

Relationship between oxidative stress and hepatic glutathione levels in ethanol-mediated apoptosis of polarized hepatic cells

AIM:To investigate the role of reactive oxygen species(ROS) in ethanol-mediated cell death of polarized hepatic(WIF-B) ***:In this work,WIF-B cultures were treated with pyrazole(inducer of cytochrome P4502E1,CYP2E1) and/or L-buthionine sulfoximine(BSO),a known inhibitor of hepatic glutathione(GSH),followed by evaluation of ROS production,antioxidant levels,and measures of cell injury(apoptosis and necrosis).RESULTS:The results revealed that ethanol treatment alone caused a significant two-fold increase in the activation of caspase-3 as well as a similar doubling in *** the activity of the CYP2E1 was increased by pyrazole pretreatment,an additional two-fold elevation in ROS was ***,the CYP2E1-related ROS elevation was not accompanied with a correlative increase in apoptotic cell injury,but rather was found to be associated with an increase in necrotic cell ***,when the thiol status of the cells was manipulated using BSO,the ethanol-induced activation of caspase-3 was ***,ethanol-treated cells displayed enhanced susceptibility to Fas-mediated apoptosis that was blocked by GSH depletion as a result of diminished caspase-8 ***:Apoptotic cell death induced as a consequence of ethanol metabolism is not completely dependent upon ROS status but is dependent on sustained GSH levels.

Velocity Profiles between Two Baffles in a Shell and Tube Heat Exchanger

Heat exchangers are extensively utilized for waste heat recovery,oil refining,chemical processing,and steam *** this study,velocity profiles are measured using a 3D particle image velocimetry(PIV)system between two baffles in a shell and tube heat exchanger for parallel and counter *** PIV and computational fluid dynamics results show the occurrence of some strong vectors near the *** vectors are assumed due to the clearance between the inner tubes and the front ***,the major parts of the vectors are moved out through the bottom opening of the rear baffle,and other vectors produce a large circle between the two *** simulations are conducted to investigate the effects of the baffle on the heat exchanger using the Fluent *** k-εturbulence model is employed to calculate the flows along the heat exchanger.

Dietary supplementation of dried plum:a novel strategy to mitigate heat stress in broiler chickens

Background:Heat stress is a significant problem in the poultry industry,causing a severe economic loss due to its detrimental effects on chickens’health and *** plum(DP)is a good source of minerals,vitamins,antioxidants,and phenolic *** have suggested that DP has several health benefits,such as maintaining the body’s redox system,immune status,and calcium *** on the health benefits of DP,we hypothesized that the dietary supplementation of DP would alleviate the detrimental effects of heat stress on broiler ***:To test the hypothesis,day-old broiler chicks(n=72)were randomly allocated to three treatment groups(n=24/group):no heat stress(NHS),heat stress(HS),and heat stress with dried plum(HS+DP),and reared under standard *** inclusion of 2.5%DP in the feed of the HS+DP group was made during the treatment period,while birds in other groups were provided with a standard finisher *** 21 days,birds in the HS and HS+DP groups were exposed to cyclic heat stress conditions(33℃ for 8 h during daytime)for 3 weeks,while those in the NHS group were reared under normal conditions(22-24℃).Weekly body weight and feed intake were recorded to calculate the average daily gain(ADG),average daily feed intake(ADFI),and feed conversion ratio(FCR).Heat stress significantly decreased the final body weight,ADG,ADFI,and increased FCR compared to the NHS group,whereas dietary supplementation of DP significantly improved these growth performance parameters compared to the HS ***,supplementation of DP significantly increased the expression of heat shock protein-related genes(HSF1,HSF3,HSP70,and HSP90),antioxidant-related genes(SOD1,SOD2,GPX1,GPX3,PRDX1,and TXN),tight junction-related genes(CLDN1,and OCLN),and immune-related genes(IL4,MUC2)in the ileum as compared to the HS *** microbiota analysis showed significant enrichment of Bacillales,Christensenellaceae,Bacillaceae,Peptostreptococcaceae,and Anaerot

The effective surface treatment method for steel insert composited with Al base metal by expendable pattern casting （EPC） process and the bonding interface between steel insert and Al base metal were *** was found that Zn plating on steel insert was effective on improving the bonding property between steel insert and Al base metal in EPC *** is thought to promote the formation of diffusion *** almost none content of Zn was observed in the boundary which had been plated on the steel insert.A diffusion layer consisting of Al,Si and Fe was formed at the insert/alloy interface and its hardness was higher than the steel insert as matter of course Al base *** layer turned out to be intermetallic compounds of Al-Si-Fe *** pouring temperature promoted the diffusion of Fe into Al alloy,so Fe content in intermetallic layers increased at higher pouring *** layer nearest to steel disappeared due to applied pressure.

Cu^+-incorporated TiO_2 overlayer on Cu_2O nanowire photocathodes for enhanced photoelectrochemical conversion of CO_2 to methanol

In this paper, we report photoelectrochemical（PEC） conversion of carbon dioxide（CO_2） using photocathodes based on Cu_2O nanowires（NWs） overcoated with Cu~＋-incorporated crystalline TiO_2（TiO_2–Cu~＋）shell. Cu_2O NW photocathodes show remanent photocurrent of 5.3% after 30 min of PEC reduction of CO_*** coating Cu_2O with TiO_2–Cu~＋ overlayer, the remanent photocurrent is 27.6%, which is an increase by5.2 fold. The charge transfer resistance of Cu_2O/TiO_2–Cu~＋ is 0.423 k/cm2, whereas Cu_2O photocathode shows resistivity of 0.781 k/cm2 under irradiation. Mott–Schottky analysis reveals that Cu~＋ species embedded in TiO_2 layer is responsible for enhanced adsorption of CO_2 on TiO_2 surface, as evidenced by the decrease of capacitance in the Helmholtz layer. On account of these electrochemical and electronic effects by the Cu~＋ species, the Faradaic efficiency（FE） of photocathodes reaches as high as 56.5% when TiO_2–Cu~＋ is added to Cu_2O, showing drastic increase from 23.6% by bare Cu_2O photocathodes.

Disruption of bone homeostasis caused by metastatic osteolytic breast cancer cells increases inflammatory osteolysis and decreases bone formation,thereby predisposing patients to pathological fracture and cancer *** of osteoblast function induces skeletal diseases due to the disruption of bone *** observed increased activation of pERk1/2 in osteolytic breast cancer cells and osteoblasts in human pathological specimens with aggressive osteolytic breast cancer *** confirmed that osteolytic breast cancers with high expression of pERk1/2 disrupt bone homeostasis via osteoblastic ERk1/2 activation at the bone-breast cancer *** process of inflammatory osteolysis modulates ERk1/2 activation in osteoblasts and breast cancer cells through dominant-negative MEk1 expression and constitutively active MEk1 expression to promote cancer growth within ***,an FDA-approved MEk inhibitor,not only reduced breast cancer-induced bone destruction but also dramatically reduced cancer growth in bone by inhibiting the inflammatory skeletal *** together,these findings suggest that ERk1/2 activation in both breast cancer cells and osteoblasts is required for osteolytic breast cancer-induced inflammatory osteolysis and that ERk1/2 pathway inhibitors may represent a promising adjuvant therapy for patients with aggressive osteolytic breast cancers by altering the shared cancer and bone microenvironment.

The fourth-order Zakharov integral equation for surface gravity waves derived by Stiasshie and Shemer is modified to include the effect of shear flow. Using this new equation the class instability of a Stokes wave train in shear flow is studied. The effect of the vorticity on the instability is discussed, and a kind of new unstable regions has been found.
