Computer-aided sperm analysis： a useful tool to evaluate patient＇s response to varicocelectomy

Preoperative and postoperative sperm parameter values from infertile men with varicocele were analyzed by computer-aided sperm analysis （CASA） to assess if sperm characteristics improved after varicocelectomy. Semen samples of men with proven fertility （n = 38） and men with varicocele-related infertility （n = 61） were also analyzed. Conventional semen analysis was performed according to WHO （2010） criteria and a CASA system was employed to assess kinetic parameters and sperm concentration. Seminal parameters values in the fertile group were very far above from those of the patients, either before or after surgery. no significant improvement in the percentage normal sperm morphology （P = 0. 10）, sperm concentration （P = 0.52）, total sperm count （P = 0.76）, subjective motility （%）（P = 0.97） nor kinematics （P = 0.30） was observed after varicocelectomy when all groups were compared. neither was significant improvement found in percentage normal sperm morphology （P = 0.91）, sperm concentration （P =0. 10）, total sperm count （P = 0.89） or percentage motility （P = 0.77） after varicocelectomy in paired comparisons of preoperative and postoperative data. Analysis of paired samples revealed that the total sperm count （P = 0.01） and most sperm kinetic parameters： curvilinear velocity （P = 0.002）, straight-line velocity （P = 0.0004）, average path velocity （P = 0.0005）, linearity （P = 0.02）, and wobble （P = 0.006） improved after surgery. CASA offers the potential for accurate quantitative assessment of each patient＇s response to varicocelectomy.

Oligodendrocyte precursor cell maturation: role of adenosine receptors

Oligodendrocyte-formed myelin sheaths allow fast synaptic transmission in the brain and their degeneration leads to demyelinating diseases such as multiple sclerosis. Remyelination requires the differentiation of oligodendrocyte progenitor cells into mature oligodendrocytes but, in chronic neurodegenerative disorders, remyelination fails due to adverse environment. Therefore, a strategy to prompt oligodendrocyte progenitor cell differentiation towards myelinating oligodendrocytes is required. The neuromodulator adenosine, and its receptors(A1, A(2A), A(2B) and A3 receptors: A1R, A(2A)R, A(2B)R and A3R), are crucial mediators in remyelination processes. It is known that A1Rs facilitate oligodendrocyte progenitor cell maturation and migration whereas the A3Rs initiates apoptosis in oligodendrocyte progenitor cells. Our group of research contributed to the field by demonstrating that A(2A)R and A(2B)R inhibit oligodendrocyte progenitor cell maturation by reducing voltage-dependent K^+ currents necessary for cell differentiation. The present review summarizes the possible role of adenosine receptor ligands as potential therapeutic targets in demyelinating pathologies such as multiple sclerosis.

Self-assembling peptides （SAPs） are synthetic bioinspired biomaterials that can be feasibly multi-functionalized for cell transplantation and/or drug delivery therapies. Despite their superior biocompatibility and ease of scaling-up for production, they are unfortunately hampered by weak mechanical properties due to transient non-covalent interactions among and within the self-assembled peptide chains, thus limiting their potential applications as fillers, hemostat solutions, and fragile scaffolds for soft tissues. Here, we have developed and characterized a cross-linking strategy that increases both the stiffness and the tailorability of SAP hydrogels, enabling the preparation of transparent flexible threads, discs, channels, and hemispherical constructs. Empirical and computational results, in close agreement with each other, confirmed that the cross-linking reaction does not affect the previously self-assembled secondary structures. In vitro tests also provided a first hint of satisfactory biocompatibility by favoring viability and differentiation of human neural stem cells. This work could bring self-assembling peptide technology to many applications that have been precluded so far, especially in regenerative medicine.

Anti-TnF alpha in the treatment of ulcerative colitis:A valid approach for organ-sparing or an expensive option to delay surgery?

Ulcerative colitis (UC) is an inflammatory bowel disease affecting large bowel with variable clinical course. The history of disease has been modified by the introduction of biologic therapy, in particular Infliximab (IFX), that has demonstrated efficacy in inducing fast symptoms remission, promoting mucosal healing and maintaining long-term remission. However, surgery is still needed for UC patients: in case of failure of medical therapy and if acute complications or a malignancy occurred. Surgical treatment is associated with a short-term post-operative mortality and morbidity respectively of 0%-4% and 30%. In this study we systematically analyzed: the role of IFX in reducing the colectomy rate, the risk of post-operative morbidity in pre-operatively IFX-treated patients and the cost-effectiveness of IFX therapy. Four of 5 analyzed randomized controlled trials demonstrated that therapy with IFX significantly reduces the colectomy rate. Moreover, pre-operative treatment with IFX doesn’t seem to increase post-operative infectious complications. By an economic point of view, the cost-effectiveness of IFX-therapy was demonstrated for UC patients suffering from moderate to severe UC in a study based on a cost estimation of the national Health Service of England and Wales. However, the argument is debated.

The efficiency of direct electron flow from electron donors to electron acceptors in redox reactions is significantly influenced by the spatial separation of these ***,a class of redox-active substances naturally present in soil-water systems,act as electron reservoirs,reversibly donating,storing,and accepting *** capability allows the temporal and spatial decoupling of redox half-reactions,providing a flexible electron transfer *** this review,we systematically examine the critical role of geobatteries in influencing electron transfer and utilization in environmental biogeochemical *** redox-active centers within geobatteries,such as quinone-like moieties,nitrogen-and sulfur-containing groups,and variable-valent metals,possess the potential to repeatedly charge and *** characterization techniques,ranging from qualitative methods like elemental analysis,imaging,and spectroscopy,to quantitative techniques such as chemical,spectroscopic,and electrochemical methods,have been developed to evaluate this reversible electron transfer ***,current research on the ecological and environmental significance of geobatteries extends beyond natural soil-water systems(e.g.,soil carbon cycle)to engineered systems such as water treatment(e.g.,nitrogen removal)and waste management(e.g.,anaerobic digestion).Despite these advancements,challenges such as the complexity of environmental systems,difficulties in accurately quantifying electron exchange capacity,and scaling-up issues must be addressed to fully unlock their *** review underscores both the promise and challenges associated with geobatteries in responding to environmental issues,such as climate change and pollutant transformation.
