Gene therapy has become the most effective treatment for monogenic *** LEPTIN deficiency is a rare autosomal recessive monogenic obesity syndrome caused by mutations in the Leptin ***/ob mouse is a monogenic obesity model,which carries a homozygous point mutation of C to T in Exon 2 of the Leptin ***,we attempted to edit the mutated Leptin gene in ob/ob mice preadipocytes and inguinal adipose tissues using CRISPR/Cas9 to correct the C to T mutation and restore the production of LEPTIN protein by *** edited preadipocytes exhibit a correction of 5.5%of Leptin alleles and produce normal LEPTIN protein when differentiated into mature *** ob/ob mice display correction of 1.67%of Leptin alleles,which is sufficient to restore the production and physiological functions of LEPTIN protein,such as suppressing appetite and alleviating insulin *** study suggests CRISPR/Cas9-mediated in situ genome editing as a feasible therapeutic strategy for human monogenic diseases,and paves the way for further research on efficient delivery system in potential future clinical application.

Strain-induced martensitic transformation in biomedical Co-Cr-W-Ni alloys

The nucleation,variant selection,and orientation dependence of the strain-induced martensitic transformation(SIMT)process in biomedical Co-Cr-W-Ni alloys were *** experimental results show that theε-hexagonal-close-packed phase was preferentially formed at theΣ3 twin boundaries and high-angle grain boundaries during the tensile *** theoretical analysis shows that the variant selection of SIMT is governed by Schmid’s ***,the SIMTedε-phase did not form equally on the two sides of the annealing twins,even though they had the same Schmid *** phenomenon is related to the mechanical work developed by the formation of theε-*** the side which has both high Schmid factor and high mechanical work can initiate the SIMT process.A strongfiber texture was formed,and theε-variants tended to appear in grains with orientations close to theanddirections during the tensile *** results can provide theoretical guidance for controlling the SIMT process of Co-Cr-W-Ni alloys to fabricate more reliable stents.

Bioinspired flexible,high-strength,and versatile hydrogel with the fiberboard-and-mortar hierarchically ordered structure

The synthetic hydrogels with high water contents are promising for various applications,however,they usually exhibit low mechanical *** this work,inspired by the natural biological soft tissues,whose hierarchically ordered fibrous structures result in high strength and good flexibility,a flexible,high-strength,and versatile hydrogel with the fiberboard-and-mortar hierarchically ordered structure(HFMOS)is developed based on ultralong hydroxyapatite(HAP)nanowires and polyacrylic acid(PAA).The as-prepared HFMOS hydrogel has a high water content(~70 wt.%),dense structure,and excellent mechanical properties,and these properties are similar to those of the human cartilage and are superior to many hydrogels reported in the *** excellent mechanical properties of the HFMOS hydrogel originate from the combination of the fiberboard-and-mortar hierarchically ordered structure,reinforcement of ultralong HAP nanowires,strong interfacial strength,and multiple energy dissipation ***,thanks to the controllable components and injection procedure,the HFMOS hydrogel with a Janus structure is prepared for particular *** HFMOS hydrogel possesses abundant ordered water channels,and can be used for loading,release,and directed delivery of various functional ***,the as-prepared flexible,high-strength,and versatile HFMOS hydrogel possesses a great potential for various applications such as water purification,pollution treatment,biomedicine,nanofluidic devices,and high-performance structural materials.

The development of low-temperature solid oxide fuel cells(LT-SOFCs)is of significant importance for realizing the widespread application of *** has stimulated a substantial materials research effort in developing high oxide-ion conductivity in the electrolyte layer of *** this context,for the first time,a dielectric material,CaCu_(3)Ti_(4)O_(12)(CCTO)is designed for LT-SOFCs electrolyte application in this *** individual CCTO and its heterostructure materials with a p-type Ni_(0.8)Co_(0.15)Al_(0.05)LiO_(2−δ)(NCAL)semiconductor are evaluated as alternative electrolytes in LT-SOFC at 450–550℃.The single cell with the individual CCTO electrolyte exhibits a power output of approximately 263 mW cm^(-2) and an open-circuit voltage(OCV)of 0.95 V at 550℃,while the cell with the CCTO–NCAL heterostructure electrolyte capably delivers an improved power output of approximately 605 mW cm^(-2) along with a higher OCV over 1.0 V,which indicates the introduction of high hole-conducting NCAL into the CCTO could enhance the cell performance rather than inducing any potential short-circuiting *** is found that these promising outcomes are due to the interplay of the dielectric material,its structure,and overall properties that led to improve electrochemical mechanism in CCTO–***,density functional theory calculations provide the detailed information about the electronic and structural properties of the CCTO and NCAL and their heterostructure CCTO–*** study thus provides a new approach for developing new advanced electrolytes for LT-SOFCs.

Hierarchical Cluster Coordination Control Strategy for Large-scale Wind Power Based on Model Predictive Control and Improved Multi-time-scale Active Power Dispatching

The grid-connection of large-scale and high-penetration wind power poses challenges to the friendly dispatching control of the power *** coordinate the complicated optimal dispatching and rapid real-time control,this paper proposes a hierarchical cluster coordination control(HCCC)strategy based on model predictive control(MPC)*** the time-varying characteristics of wind power generation,the proposed HCCC strategy constructs an improved multitime-scale active power dispatching model,which consists of five parts:formulation of cluster dispatching plan,rolling modification of intra-cluster plan,optimization allocation of wind farm(WF),grouping coordinated control of wind turbine group(WTG),and real-time adjustment of single-machine *** time resolutions are sequentially given as 1 hour,30 min,15 min,5 min,and 1 *** addition,a combined predictive model based on complete ensemble empirical mode decomposition with adaptive noise(CEEMDAN),wavelet thresholding(WT),and least squares support vector machine(LSSVM)is *** fast predictive feature of this model cooperates with the HCCC strategy that effectively improves the predictive control *** results show that the proposed HCCC strategy enables rapid response to active power control(APC),and significantly improves dispatching control accuracy and wind power accommodation capabilities.

Spinal cord injuries lead to significant loss of motor, sensory, and autonomic functions, presenting major challenges in neural regeneration. Achieving effective therapeutic concentrations at injury sites has been a slow process, partly due to the difficulty of delivering drugs effectively. Nanoparticles, with their targeted delivery capabilities, biocompatibility, and enhanced bioavailability over conventional drugs, are garnering attention for spinal cord injury treatment. This review explores the current mechanisms and shortcomings of existing treatments, highlighting the benefits and progress of nanoparticle-based approaches. We detail nanoparticle delivery methods for spinal cord injury, including local and intravenous injections, oral delivery, and biomaterial-assisted implantation, alongside strategies such as drug loading and surface modification. The discussion extends to how nanoparticles aid in reducing oxidative stress, dampening inflammation, fostering neural regeneration, and promoting angiogenesis. We summarize the use of various types of nanoparticles for treating spinal cord injuries, including metallic, polymeric, protein-based, inorganic non-metallic, and lipid nanoparticles. We also discuss the challenges faced, such as biosafety, effectiveness in humans, precise dosage control, standardization of production and characterization, immune responses, and targeted delivery in vivo. Additionally, we explore future directions, such as improving biosafety, standardizing manufacturing and characterization processes, and advancing human trials. Nanoparticles have shown considerable progress in targeted delivery and enhancing treatment efficacy for spinal cord injuries, presenting significant potential for clinical use and drug development.

High-performance, low-cost joint equalizer and trellis decoder for 1000BASE-T gigabit Ethernet transceiver

This article presents an M-algorithm （MA） decoder with 4 survival paths （MA4） for Institute of Electrical and Electronics Engineers （IEEE） 802.3ab 1000BASE-T gigabit Ethernet （GbE） transceiver. To fulfill the entire requirements, various methods were introduced to accelerate the MA4 decoder while retaining the desired high performance and low complexity. Optimized look-ahead architecture was employed to solve the critical path problem with minimal gate consumption. Symbol compression methods saved registers during pipeline stages. A sorting network accelerated the kernel sorting operation at low hardware cost by utilizing the special characteristics of MA4. Simulations and synthesis results show that the proposed decoder achieves 125 MHz clock frequency and l Gb/s throughput in 1.8 V 0.18μm standard cell complementary metal-oxide-semiconductor （CMOS） process. It achieves additional 0.4 dB coding gain over 14tap parallel decision feedback decoder （PDFD） with 39% area reduction.

RESPONSE ANALYSIS OF PIEZOELECTRIC SHELLS IN PLANE STRAIN UNDER RANDOM EXCITATIONS

The random response of a piezoelectric thick shell in plane strain state under boundary random excitations is studied and illustrated with a piezoelectric cylindrical shell. The differential equation for electric potential is integrated radially to obtain the electric potential as a function of displacement. The random stress boundary conditions are converted into homogeneous ones by transformation,which yields the electrical and mechanical coupling differential equation for displacement under random excitations. Then this partial differential equation is converted into ordinary differential equations using the Galerkin method and the Legendre polynomials,which represent a random multi-degree-of-freedom system with asymmetric stiffness matrix due to the electrical and mechanical coupling and the transformed boundary conditions. The frequency-response function matrix and response power spectral density matrix of the system are derived based on the theory of random vibration. The mean-square displacement and electric potential of the piezoelectric shell are finally obtained,and the frequency-response characteristics and the electrical and mechanical coupling properties are explored.

Experimental research on the microstructure and compressive and tensile properties of nano-SiO_(2) concrete containing basalt fibers

Urban underground space resources are gaining increasing attention for the sustainable development of *** concrete cannot meet the needs of underground ***-performance concrete was prepared using varying dosages of nano-SiO_(2)and basalt fiber,and its compressive and tensile strength was *** concrete microstructure was analyzed and used to assess the mechanisms through which the nano-SiO_(2)and basalt fibers affect the strength of *** cement hydration productions in concrete produced varied with the dosage of nano-SiO_(2).When the nano-SiO_(2)dosage was between 0 and 1.8%,the mass of the C-S-H gel and AFt crystals increased gradually with the nano-SiO_(2)*** the nano-SiO_(2)dosage was 1.2%,optimum amounts of C-S-H gel and AFt crystals existed,and the compactness of concrete was well,which agreed with the results of the compressive strength *** the basalt-fiber dosage was between 3 and 4 kg/m^(3),the basalt fibers and the cement matrix were closely bonded,and the splitting tensile strength of the concrete markedly *** the basalt-fiber dosage exceeded 5 kg/m^(3),the basalt fibers clustered together,resulting in weak bonding between the basalt fibers and the cement matrix,consequently,the basalt fibers were easily pulled apart from the *** the nano-SiO_(2)and basalt fiber dosages were 1.2%and 3 kg/m^(3),respectively,the compactness of the concrete microstructure was well and the strength enhancement was the greatest;additionally,the compressive strength and splitting tensile strength were 9.04%and 17.42%,respectively,greater than those of plain *** macroscopic tests on the mechanical properties of the nano-SiO_(2)concrete containing basalt fibers agreed well with the results of microstructure analysis.

Study of neutron production and moderation for sulfur neutron capture therapy

Neutron capture therapy with Sulfur-33, similar to boron neutron capture therapy with Boron-10, is effective in treating some types of tumors including ocular melanoma. The key point in sulfur neutron capture therapy is whether the neutron beam flux and the resonance capture cross section of ^(33)S(n;α)^(30) Si reaction at 13.5 keV can achieve the requirements of radiotherapy. In this research,the authors investigated the production of 13.5 keV neutron production and moderation based on an accelerator neutron source. A lithium glass detector was used to measure the neutron flux produced via near threshold^7 Li(p,n)~7 Be reaction using the time-of-flight method. Furthermore, the moderation effects of different kinds of materials were investigated using Monte Carlo simulation.
