Predictors of efficacy of corticosteroid switching from abiraterone plus prednisone to dexamethasone in patients with metastatic castration-resistant prostate cancer

Corticosteroid switching can reverse abiraterone resistance in some patients with metastatic castration-resistant prostate cancer(mCRPC).Here,we investigated the potential biomarkers for predicting the efficacy of corticosteroid switching during treatment with abiraterone acetate(AA).We retrospectively analyzed 101 mCRPC patients receiving corticosteroid switching from West China Hospital and Sun Yat-Sen University Cancer Center between January 2016 and December 2018.All cases received AA plus prednisone as first-line therapy during mCRPC.Primary end points were biochemical progression-free survival(bPFS)and overall survival(OS).The risk groups were defined based on multivariate analysis.A total of 42(41.6%)and 25(24.8%)patients achieved 30%and 50%decline in prostate-specific antigen(PSA),respectively,after corticosteroid switching.The median bPFS and median OS on AA plus dexamethasone were 4.9(95%confidence interval[CI]:3.7–6.0)months and 18.8(95%CI:16.2–30.2)months,respectively.Aldo-keto reductase family 1 member C3(AKR1C3)expression(hazard ratio[HR]:2.15,95%Cl:1.22–3.80,P=0.008)and baseline serum alkaline phosphatase(ALP;HR:4.95,95%Cl:2.40–10.19,P160 IU l−1(HR:3.41,95%Cl:1.57–7.38,P=0.002)together with PSA level at switch≥50 ng ml−1(HR:2.59,95%Cl:1.22–5.47,P=0.013)independently predicted poorer OS.Based on the predictive factors in multivariate analysis,we developed two risk stratification tools to select candidates for corticosteroid switching.Detection of serum ALP level,PSA level,and tissue AKR1C3 expression in mCRPC patients could help make clinical decisions for corticosteroid switching.

Recent research efforts in the field of electromagnetic interference shielding(EMI)materials have focused on biomass as a green and sustainable resource.More specifically,wood and cellulose nano fiber(CNF)have many advantages,some of which include lightweight,porosity,widespread availability,low cost,and easy processing.These favorable properties have led researchers to consider these types of biomass as an EMI shielding material with great potential.At present,while many excellent published works in EMI shielding materials have investigated wood and CNF,this research area is still new,compared with non-biomass EMI shielding materials.More specifically,there is still a lack of in-depth research and summary on the preparation process,pore structure regulation,component optimization,and other factors affecting the EMI shielding of wood and CNF based EMI shielding materials.Thus,this review paper presents a comprehensive summary of recent research on wood and CNF based EMI shielding materials in recent three years in terms of the preparation methods,material structure design,component synergy,and EMI mechanism,and a forward future perspective for existing problems,challenges,and development trend.The ultimate goal is to provide a comprehensive and informative reference for the further development and exploration of biomass EMI shielding materials.

Updated developments on molecular imaging and therapeutic strategies directed against necrosis

Cell death plays important roles in living organisms and is a hallmark of numerous disorders such as cardiovascular diseases, sepsis and acute pancreatitis. Moreover, cell death also plays a pivotal role in the treatment of certain diseases, for example, cancer. Noninvasive visualization of cell death contributes to gained insight into diseases, development of individualized treatment plans, evaluation of treatment responses, and prediction of patient prognosis. On the other hand, cell death can also be targeted for the treatment of diseases. Although there are many ways for a cell to die, only apoptosis and necrosis have been extensively studied in terms of cell death related theranostics. This review mainly focuses on molecular imaging and therapeutic strategies directed against necrosis. Necrosis shares common morphological characteristics including the rupture of cell membrane integrity and release of cellular contents, which provide potential biomarkers for visualization of necrosis and necrosis targeted therapy. In the present review, we summarize the updated joint efforts to develop molecular imaging probes and therapeutic strategies targeting the biomarkers exposed by necrotic cells. Moreover, we also discuss the challenges in developing necrosis imaging probes and propose several biomarkers of necrosis that deserve to be explored in future imaging and therapy research.

As the continuous development of the industrial revolution,nanomaterials with excellent characteristics have been widely applied in various fields,greatly increasing the probability of human exposure to nanomaterials and the concerns about the potential nanotoxicity.Existing studies have shown that the toxicity of nanomaterials may be closely related to oxidative stress,inflammatory response,phagocytosis dysfunction,DNA damage,etc.Based on our focus,nanomaterials may cross the human barrier through various channels and disrupt various cell-cell junctions,while the integrity of cellular barrier is a necessary for the normal physiological function of various organs.However,until now,there is still a lack of systematic discussion in this field.This review illustrates the importance of cell-cell junctions in maintaining various organ functions and highlights the mechanism of various nanomaterials disrupt cell-cell junctions,as well as the possible damage to various organs,such as brain,eye,lung,breast,intestine,placenta,testis,heart,liver,kidney,skin,etc.Awareness of the potential negative effects of nanomaterials will help scientists deeply understand the limitations of nanotechnology,inspiring them to develop safer and more efficient nanomaterials for future personalized nanomedicine.

Kinetics process for structure-engineered integrated gradient porous paper-based supercapacitors with boosted electrochemical performance

Due to their rich and adjustable porous network structure,paper-based functional materials have become a research hotspot in the field of energy storage.However,reasonably designing and making full use of the rich pore structure of paper-based materials to improve the electrochemical performance of paper-based energy storage devices still faces many challenges.Herein,we propose a structure engineering technique to develop a conductive integrated gradient porous paper-based(CIGPP)supercapacitor,and the kinetics process for the influence of gradient holes on the electrochemical performance of the CIGPP is investigated through experimental tests and COMSOL simulations.All results indicate that the gradient holes endow the CIGPP with an enhanced electrochemical performance.Specifically,the CIGPP shows a significant improvement in the specific capacitance,displays rich frequency response characteristics for electrolyte ions,and exhibits a good rate performance.Also,the CIGPP supercapacitor exhibits a low self-discharge and maintains a stable electrochemical performance in different electrolyte environments because of gradient holes.More importantly,when the CIGPP is used as a substrate to fabricate a CIGPP-PAni hybrid,it still maintains good electrochemical properties.In addition,the CIGPP supercapacitor also shows excellent stability and sensitivity for monitoring human motion and deaf-mute voicing,showing potential application prospects.This study provides a reference and feasible way for the design of structure-engineered integrated paper-based energy storage devices with outstanding comprehensive electrochemical performance.

A Nested Case-Control Study to Explore the Association between Immunoglobulin G N-glycans and Ischemic Stroke

Objective This study prospectively investigates the association between immunoglobulin G(IgG)N-glycan traits and ischemic stroke(IS) risk.Methods A nested case-control study was conducted in the China suboptimal health cohort study,which recruited 4,313 individuals in 2013–2014. Cases were identified as patients diagnosed with IS, and controls were 1:1 matched by age and sex with cases. Ig G N-glycans in baseline plasma samples were analyzed.Results A total of 99 IS cases and 99 controls were included, and 24 directly measured glycan peaks(GPs) were separated from Ig G N-glycans. In directly measured GPs, GP4, GP9, GP21, GP22, GP23, and GP24 were associated with the risk of IS in men after adjusting for age, waist and hip circumference,obesity, diabetes, hypertension, and dyslipidemia. Derived glycan traits representing decreased galactosylation and sialylation were associated with IS in men(FBG2S2/(FBG2 + FBG2S1 + FBG2S2): odds ratio(OR) = 0.92, 95% confidence interval(CI): 0.87–0.97;G1n: OR = 0.74, 95% CI: 0.63–0.87;G0n: OR =1.12, 95% CI: 1.03–1.22). However, these associations were not found among women.Conclusion This study validated that altered Ig G N-glycan traits were associated with incident IS in men, suggesting that sex discrepancies might exist in these associations.

Clinical and oncologic findings of extraprostatic extension on needle biopsy in de novo metastatic prostate cancer

This study aimed to explore the clinical and oncologic findings in patients with de novo metastatic prostate cancer(mPCa)and extraprostatic extension(EPE)on biopsy.We retrospectively evaluated data on 630 patients with de novo mPCa between January 2009 and December 2017 in the West China Hospital(Chengdu,China),including evaluating the relationships between EPE and other variables and the association of EPE with survival outcomes by the Chi-square test,Kaplan–Meier curves,and the Cox proportional-hazards model.EPE was found in 70/630 patients,making a prevalence of 11.1%.The presence of EPE on biopsy was associated with higher Gleason scores and higher incidence of neuroendocrine differentiation(NED),intraductal carcinoma of the prostate(IDC-P),and perineural invasion(Pni).Compared with those without EPE,patients with EPE had shorter castration-resistant prostate cancer-free survival(CFS;median:14.1 vs 17.1 months,P=0.015)and overall survival(OS;median:43.7 vs 68.3 months,P=0.032).According to multivariate analysis,EPE was not an independent predictor for survival.Subgroup analyses demonstrated that patients with favorable characteristics,including negative NED or IDC-P status,Eastern Cooperative Oncology Group(ECOG)scorenicopathological features and poorer CFS and OS.These data suggest that EPE may be an indicator of poor prognosis,particularly in patients,otherwise considered likely to have favorable survival outcomes.

A comprehensive error calibration method based on dual uniform circular array

Based on the dual uniform circular array, a novel method is proposed to estimate the direction-of-arrival (DOA) and jointly calibrate gain-phase errors, position errors, and mutual coupling errors. In this paper, only one auxiliary source is required to generate three time-disjoint calibration sources with the help of the rotation platform. Subsequently, according to the principle that the signal subspace is orthogonal to the noise subspace, the cost function is constructed. The alternating iteration method is used to estimate the coefficients of the three kinds of errors. During the process, the proposed algorithm makes full use of the structural characteristics of the array when estimating mutual coupling errors, while the signal phase matrix is used to eliminate the phase influence caused by the delay in signal arrival at the antenna array when estimating gain-phase errors and position errors. Compared with the algorithm using multidimensional nonlinear search, the proposed algorithm has lower computational com-plexity. Moreover, our algorithm does not require additional auxiliary sensors. Simulation results demonstrate that the proposed algorithm is effective and can precisely and comprehensively calibrate the errors in a dual uniform circular array.

As the lightest family member of the transition metal disulfides(TMDs), TiS2has attracted more and more attention due to its large specific surface area, adjustable band gap, good visible light absorption, and good charge transport properties. In this review, the recent state-of-the-art advances in the syntheses and applications of TiS2in energy storage, electronic devices, and catalysis have been summarized. Firstly, according to the physical presentation of the TiS2synthesis reaction, it can be divided into a solid phase synthesis, a liquid phase synthesis and a gas phase synthesis. Secondly, we summarize the applications of TiS2in energy storage, electronic devices and catalytic:(1) The applications of TiS2nanostructure in energy storage direction from the aspects of Li-ion battery(LIB), Li–S battery(LSB), Na-ion battery(niB), K-ion battery(KIB), Mg-ion battery(MIB), solar cells and hydrogen storage;(2) The applications of TiS2nanostructures in various electronic devices from thermoelectric devices, high power lasers and flexible devices;(3) Applications based on energy catalysis and environmental catalysis. Based on the various synthetic technologies and wide applications of TiS2, we firmly believe that the challenges of TiS2will be solved and become a hot star material in the future. Finally, we discuss the future scope and the current challenges arising from this fascinating material.

Comparative analysis of C-type lectin domain proteins in the ghost moth, Thitarodes xiaojinensis (Lepidoptera: Hepialidae)

Insects have a large family of C-type lectins involved in cell adhesion, pathogen recognition and activation of immune responses. In this study, 32 transcripts encoding C-type lectin domain proteins (CTLDPs) were identified from the Thitarodes xiaojinensis transcriptome. According to their domain structures, six CTLDPs with one carbohydraterecognition domain (CRD) were classified into the CTL-S subfamily. The other 23 CTLDPs with two CRDs were grouped into the immulectin (IML) subfamily. The remaining three with extra regulatory domains were sorted into the CTL-X subfamily. Phylogenetic analysis showed that CTL-S and CTL-X members from different insects could form orthologous groups. In contrast, no T. xiaojinensis IML orthologues were found in other insects. Remarkable lineage-specific expansion in this subfamily was observed reflecting that these CTLDPs, as important receptors, have evolved diversified members in response to a variety of microbes. Prediction of binding ligands revealed that T. xiaojinensis, a coldadapted species, conserved the ability of CRDs to combine with Ca^2+ to keep its receptors from freezing. Comparative analysis of induction of CTLDP genes after different immune challenges indicated that IMLs might play critical roles in immune defenses. This study examined T. xiaojinensis CTLDPs and provides a basis for further studies of their characteristics.
