Photodynamic therapy(PDT)is an effective treatment method for tumors.But the specifically accumulated of photosensitizer was very difficult in the tumor site,which greatly limited the efficacy of PDT.Here,mitochondria-targeted Janus mesoporous nanoplatform(JPMO-Pt-CTPP-ZnPc)for PDT was prepared,the nanoplatform has uniform size(275 nm)and good dispersion and biocompatibility.The confocal laser scanning microscopy(CLSM)revealed the signal of ZnPc of JPMO-Pt-CTPP-ZnPc were higher than JPMO-Pt-ZnPc in tumor cells,and flow cytometry results showed the cell uptake efficiency of JPMO-Pt-CTPP-ZnPc was 2.5-fold higher than that of JPMO-Pt-ZnPc.This revealed the modification of CTPP significantly improves the targeting ability of the nanoplatform.In vitro anti-tumor experiment showed the JPMO-Pt-CTPP-ZnPc significantly inhibited the growth of tumor cells upon the irradiation of low-power laser,and the survival rate of cells incubated with 60μg/mL JPMO-Pt-CTPP-ZnPc was only 3%.Simultaneously,compared with JPMO-Pt-ZnPc(not modified with mitochondria targeting molecules CTPP),the PDT efficacy of JPMO-Pt-CTPP-ZnPc was significantly better,as it has targeted mitochondria in cells.

Paragangliomas,also known as pheochromocytomas(1–9 cases per million),arise in the paraganglia.[1]Pheochromocytomas occur in the adrenal glands,while paragangliomas occur elsewhere.[2]Paragangliomas originate from paraganglion cells,which are derived from the neural ectoderm of the nerves and migrate along both sides of the median axis from the base of the skull to the pelvis during embryonic development.

Expression and clinical significance of pattern recognition receptor-associated genes in hand, foot and mouth disease

Objective:To explore which pattern recognition receptors(PRRs)play a key role in the development of hand,foot,and mouth disease(HFMD)by analyzing PRR-associated genes.Methods:We conducted a comparative analysis of PRR-associated gene expression in human peripheral blood mononuclear cells(PBMCs)infected with enterovirus 71(EV-A71)which were derived from patients with HFMD of different severities and at different stages.A total of 30 PRR-associated genes were identified as significantly upregulated both over time and across different EV-A71 isolates.Subsequently,ELISA was employed to quantify the expression of the six most prominent genes among these 30 identified genes,specifically,BST2,IRF7,IFI16,TRIM21,MX1,and DDX58.Results:Compared with those at the recovery stage,the expression levels of BST2(P=0.027),IFI16(P=0.016),MX1(P=0.046)and DDX58(P=0.008)in the acute stage of infection were significantly upregulated,while no significant difference in the expression levels of IRF7(P=0.495)and TRIM21(P=0.071)was found between different stages of the disease.The expression levels of BST2,IRF7,IFI16 and MX1 were significantly higher in children infected with single pathogen than those infected with mixed pathogens,and BST2,IRF7,IFI16 and MX1 expression levels were significantly lower in coxsackie B virus(COXB)positive patients than the negative patients.Expression levels of one or more of BST2,IRF7,IFI16,TRIM21,MX1 and DDX58 genes were correlated with PCT levels,various white blood cell counts,and serum antibody levels that reflect disease course of HFMD.Aspartate aminotransferase was correlated with BST2,MX1 and DDX58 expression levels.Conclusions:PRR-associated genes likely initiate the immune response in patients at the acute stage of HFMD.

Photodynamic therapy(PDT)has been widely used in cancer treatment.However,hypoxia in most solid tumors seriously restricts the efficacy of PDT.To improve the hypoxic microenvironment,we designed a novel mesoporous platinum(mPt)nanoplatform to catalyze hydrogen peroxide(H2 O2)within the tumor cells in situ without an extra enzyme.During the fabrication,the carboxy terminus of the photosensitizer chlorin e6(Ce6)was connected to the amino terminus of the bifunctional mercaptoaminopolyglycol(SH-PEG-NH2)by a condensation reaction,and then PEG-Ce6 was modified onto the mPt moiety via the mercapto terminal of SH-PEG-NH2.Material,cellular and animal experiments demonstrated that Pt@PEG-Ce6 catalyzed H2 O2 to produce oxygen(O2)and that Ce6 transformed O2 to generate reactive oxygen species(ROS)upon laser irradiation.The Pt@PEG-Ce6 nanoplatform with uniform diameter presented good biocompatibility and efficient tumor accumulation.Due to the high atomic number and good near-infrared absorption for Pt,this Pt@PEG-Ce6 nanoplatform showed computed tomography(CT)and photoacoustic(PA)dual-mode imaging ability,thus providing an important tool for monitoring the tumor hypoxic microenvironment.Moreover,the Pt@PEG-Ce6 nanoplatform reduced the expression of hypoxia-inducible factor-la(HIF-1α)and programmed death-1(PD-1)in tumors,discussing the relationship between hypoxia,PD-1,and PDT for the first time.

洗钱罪上游犯罪和洗钱罪的犯罪所得性质不同,对其没收应当区分开来。但当前部分洗钱罪的判决书对两类犯罪所得进行“一股脑”没收,混淆了两者的界限。洗钱罪上游犯罪的犯罪所得是通过实施该犯罪所获得的资产和财产性利益,与该罪的实行行为具有因果关系,在产生的时间上不能早于实行行为的实施;洗钱罪的犯罪所得是指实施洗钱行为所获得的报酬或对价。为了推动完善洗钱罪中两类犯罪所得的没收,建议删除《刑法》第191条“没收实施以上犯罪的所得及其产生的收益”的规定,厘清上游犯罪和洗钱罪两类犯罪所得的处置方向,在判决书中对两类犯罪所得的没收分别予以表述,进而将没收后的犯罪所得分别上缴国库或者返还上游犯罪被害人。

[目的]十大孔兑是黄河几字湾的重要组成部分,生态环境敏感脆弱,开展其土地利用变化与驱动力分析对黄河流域高质量发展具有重要生态意义。[方法]对十大孔兑基于1986年、2000年、2010年、2020年4期遥感影像,运用ArcMap 10.8空间分析法和主成分分析法系统分析其土地利用变化情况及驱动因素。[结果]林地、建设用地、耕地呈逐期扩张趋势,未利用地、草地、水域呈逐期缩减趋势,截至2020年林地成为占地面积最大地类,面积达6308.43 km^(2),占总面积的59.99%。2010—2020年综合动态度最高,达1.59%,表明土地利用变化在2010—2020年变化最为剧烈。平原区地势平坦,耕地和建设用地在此扩张明显;风沙区在生态治理政策与工程作用下,未利用地向林草地转变,在风沙区东部尤为明显,而在风沙区中部则小比例转变为光伏发电建设用地;丘陵区主要由未利用地转变为林地,由于区域工矿业发达,丘陵区东南部极小部分林地转变为城乡工矿建设用地;水域在孔兑内部共减少75.81 km^(2),而耕地除北部平原区扩张外,在孔兑沿岸增加明显。土地利用变化主要受社会经济、农业生产力、地势、政策驱动变化。[结论]近35年来,十大孔兑土地利用变化较大,整体生态向好的方向发展较为明显。该研究结果可为决策部门制定黄河流域生态保护政策提供科学参考。

Controlled PEGylation of periodic mesoporous organosilica nanospheres for improving their stability in physiological solutions

The stability of periodic mesoporous organosilica(PMO) nanoparticles in physiological solutions greatly affects their potential biomedical applications. Herein, thioether-bridged PMO nanospheres with a diameter of 61 nm are synthesized. Then, the thioether-bridged PMO nanospheres are modified with different molecular weighted polyethylene glycol(PEG) via click reaction for the first time. FI-IR and thermogravimetric analysis confirm the successful modification of PEG on the PMO. The influence of PEG molecular weight on the dispersity and stability of the PMO-PEG in phosphate buffer(PBS) and Dulbecco's modified Eagle's medium(DMEM) is studied. The results show that the PEG coating increases the stability and dispersity of PMO in the biological solutions. The PMO-PEG1K, PMO-PEG2K, and PMOPEG5K have better stability in PBS solution. 为明确黄蘑菇色素的理化性质和抗氧化活性,本研究采用响应面设计优化了黄蘑菇色素提取工艺,研究了色素稳定性和抗氧化活性。结果表明,黄蘑菇色素最佳提取工艺条件为去离子水作为提取剂,提取温度51℃,提取时间61 min,料液比1∶113 g·mL^(-1),提取率为90.93%。黄蘑菇色素粗提物对光比较敏感,避光条件下更稳定,耐高温和耐还原性较差,但其具有一定的耐酸碱性和耐氧化性;Zn^(2+)的添加增大了黄蘑菇色素的吸光度,Al^(3+)浓度超过0.25%时,色素吸光度下降;葡萄糖对色素稳定性影响较小;苯甲酸钠和蔗糖的浓度超过1%时,色素吸光度增加,对其稳定性影响较大。色素粗提物对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、羟自由基和2,2′-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)自由基有一定的清除能力,其半抑制率含量(IC_(50))分别为2.30、0.01和1.00 mg·mL^(-1),对Fe^(3+)有一定还原能力。DPPH IC_(50)值和ABTS IC_(50)值与粗多糖和氮含量呈正相关,羟自由基IC_(50)值与粗多糖和氮含量呈负相关。本研究结果为黄蘑菇色素的开发利用提供了理论依据。

以十大孔兑之一的西柳沟风沙区为研究区,通过野外监测与室内分析相结合,对研究区风况和输沙势特征进行分析,同时开展风沙流活动观测,测定流域两岸不同下垫面输沙通量,再通过函数拟合法科学估测风沙入黄量.结果表明:(1)研究区属于中等风能环境区,年均起沙风风速为6.41m/s,年均起沙风频率为17.85%,最大风速为11.37m/s;(2)不同时间尺度合成输沙势方向趋于一致,均以偏东南方向(SE,SSE)为主,年方向变率指数属于中等变率,年合成输沙势方向(RDD)为SSE方向,直接导致大量沙物质输入河道;(3)不同下垫面垂向输沙通量存在显著差异,流动沙地、半固定沙地和固定沙地风沙入黄量占比分别为77.09%、15.30%和7.61%.流域地表输沙率垂线变化表明不同下垫面总输沙率与风速均呈较好的指数函数关系,因此利用输沙率(q)、起沙风时长(h)与断面长度(d)之间关系,估算西柳沟流域年均风沙入黄总量约为8.09×10^(5)t/a.本文旨在探究西柳沟流域不同下垫面输沙率与风速两者之间耦合关系,为风沙入黄防治工作提供数据基础,从而降低风沙问题对黄河流域造成的安全隐患.
