Non-coding RNAs in exercise immunology:A systematic review

Regular physical exercise has been recognized as a potent modulator of immune function,with its effects including enhanced immune surveillance,reduced inflammation,and improved overall health.While strong evidence exists that physical exercise affects the specific expression and activity of non-coding RNAs(ncRNAs)also involved in immune system regulation,heterogeneity in individual study designs and analyzed exercise protocols exists,and a condensed list of functional,exercise-dependent ncRNAs with known targets in the immune system is missing from the literature.A systematic review and qualitative analysis was used to identify and categorize ncRNAs participating in immune modulation by physical exercise.Two combined approaches were used:(a)a systematic literature search for“ncRNA and exercise immunology”,(b)and a database search for microRNAs(miRNAs)(miRTarBase and DIANA-Tarbase v8)aligned with known target genes in the immune system based on the Reactome database,combined with a systematic literature search for“ncRNA and exercise”.Literature searches were based on PubMed,Web of Science,and SPORTDiscus;and miRNA databases were filtered for targets validated by in vitro experimental data.Studies were eligible if they reported on exercise-based interventions in healthy humans.After duplicate removal,95 studies were included reporting on 164 miRNAs,which were used for the qualitative synthesis.Six studies reporting on long-noncoding RNAs(lncRNAs)or circular RNAs were also identified.Results were analyzed using ordering tables that included exercise modality(endurance/resistance exercise),acute or chronic interventions,as well as the consistency in reported change between studies.Evaluation criteria were defined as“validated”with 100%of≥3 independent studies showing identical direction of regulation,“plausible”(≥80%),or“suggestive”(≥70%).For resistance exercise,upregulation of miR-206 was validated while downregulation of miR-133a appeared plausible.For endurance exercise,15 miRNAs were categorized as validated,wit

Ectophylla alba is a tent-making bat that roosts in mixed-sex clusters comprising adults and offspring.Our goal was to determine the genetic identity of individuals belonging to different roosting groups.We tested the hypothesis of kin selection as a major force structuring group composition.We used 9 microsatellites designed for E.alba to determine the genetic identity and probability of parentage of individuals.We analyzed parentage and kinship using the software ML-Relate,GenAIEx,and Cervus.The obtained relationship probabilities(0.5)revealed a clear maternal relationship between female adults and offspring with allele compatibility,and at least 5 relationships between male adults and pups.We found a low degree of relatedness within roosting groups.Between roosting groups at different sites,the mean probability of a half-sibling relationship ranged from 0.214 to 0.244 and,for full-sibling relationship,from 0.383 to 0.553.Genetically,adult individuals were poorly related within clusters,and kinship as an evolutionary force could not explain group membership.

Primary liver transplantation vs transplant after Kasai portoenterostomy in children with biliary atresia: A retrospective Brazilian single-center cohort

BACKGROUND Biliary atresia(BA)is the most common indication for pediatric liver transplantation,although portoenterostomy is usually performed first.However,due to the high failure rate of portoenterostomy,liver transplantation has been advocated as the primary procedure for patients with BA.It is still unclear if a previous portoenterostomy has a negative impact on liver transplantation outcomes.AIM To investigate the effect of prior portoenterostomy in infants un-dergoing liver transplantation for BA.METHODS This was a retrospective cohort study of 42 pediatric patients with BA who underwent primary liver transplantation from 2013 to 2023 at a single tertiary center in Brazil.Patients with BA were divided into two groups:Those undergoing primary liver transplantation without portoenterostomy and those undergoing liver transplantation with prior portoenterostomy.Continuous variables were compared using the Student’s t-test or the Kruskal-Wallis test,and categorical variables were compared using theχ2 or Fisher’s exact test,as appropriate.Multivariable Cox regression analysis was performed to determine risk factors for portal vein thrombosis.Patient and graft survival analyses were conducted with the Kaplan–Meier product-limit estimator,and patient subgroups were compared using the two-sided log-rank test.RESULTS Forty-two patients were included in the study(25[60%]girls),23 undergoing liver transplantation without prior portoenterostomy,and 19 undergoing liver transplantation with prior portoenterostomy.Patients with prior portoenterostomy were older(12 vs 8 months;P=0.02)at the time of liver transplantation and had lower Pediatric End-Stage Liver Disease scores(13.2 vs 21.4;P=0.01).The majority of the patients(35/42,83%)underwent livingdonor liver transplantation.The group of patients without prior portoenterostomy appeared to have a higher incidence of portal vein thrombosis(39 vs 11%),but this result did not reach statistical significance.Prior portoenterostomy was not a protective factor against portal vein

BACKGROUND Prevalence of hepatocellular carcinoma(HCC)is increasing,especially in patients with metabolic dysfunctionassociated steatotic liver disease(MASLD).AIM To investigate rifaximin(RIF)effects on epigenetic/autophagy markers in animals.METHODS Adult Sprague-Dawley rats were randomly assigned(n=8,each)and treated from 5-16 wk:Control[standard diet,water plus gavage with vehicle(Veh)],HCC[high-fat choline deficient diet(HFCD),diethylnitrosamine(DEN)in drinking water and Veh gavage],and RIF[HFCD,DEN and RIF(50 mg/kg/d)gavage].Gene expression of epigenetic/autophagy markers and circulating miRNAs were obtained.RESULTS All HCC and RIF animals developed metabolic-dysfunction associated steatohepatitis fibrosis,and cirrhosis,but three RIF-group did not develop HCC.Comparing animals who developed HCC with those who did not,miR-122,miR-34a,tubulin alpha-1c(Tuba-1c),metalloproteinases-2(Mmp2),and metalloproteinases-9(Mmp9)were significantly higher in the HCC-group.The opposite occurred with Becn1,coactivator associated arginine methyltransferase-1(Carm1),enhancer of zeste homolog-2(Ezh2),autophagy-related factor LC3A/B(Map1 Lc3b),and p62/sequestosome-1(p62/SQSTM1)-protein.Comparing with controls,Map1 Lc3b,Becn1 and Ezh2 were lower in HCC and RIF-groups(P0.05).miR-122 was higher in HCC,and miR-34a in RIF compared to controls(P0.05).CONCLUSION RIF might have a possible beneficial effect on preventing/delaying liver carcinogenesis through epigenetic modulation in a rat model of MASLD

Forensic data management and database systems in forensic investigations for cases of missing and unidentified persons in Brazil

Forensic investigations,especially those related to missing persons and unidentified remains,produce different types of data that must be managed and understood.The data collected and produced are extensive and originate from various sources:the police,non-governmental organizations(NGOs),medical examiner offices,specialised forensic teams,family members,and others.Some examples of information include,but are not limited to,the investigative background information,excavation data of burial sites,antemortem data on missing persons,and postmortem data on the remains of unidentified individuals.These complex data must be stored in a secured place,analysed,compared,shared,and then reported to the investigative actors and the public,especially the families of missing persons,who should be kept informed of the investigation.Therefore,a data management system with the capability of performing the tasks relevant to the goals of the investigation and the identification of an individual,while respecting the deceased and their families,is critical for standardising investigations.Data management is crucial to assure the quality of investigative processes,and it must be recognised as a holistic integrated system.The aim of this article is to discuss some of the most important components of an effective forensic data management system.The discussion is enriched by examples,challenges,and lessons learned from the erratic development and launching of databases for missing and unidentified persons in Brazil.The main objective of this article is to bring attention to the urgent need for an effective and integrated system in Brazil.

Effects of a remote,YouTube-delivered exercise intervention on young adults’physical activity,sedentary behavior,and sleep during the COVID-19 pandemic:Randomized controlled trial

Background:Public health guidelines have called for innovative and flexible physical activity(PA)intervention strategies to promote PA and health amid the coronavirus disease 2019(COVID-19)pandemic.Therefore,this study’s purpose was to examine the effects of a home-based,YouTube-delivered PA intervention grounded in self-determination theory on young adults’free-living PA,sedentary behavior,and sleep quality(NCT04499547).Methods:Sixty-four young adults(48 females;age=22.8±3.4 years,mean±SD;body mass index=23.1±2.6 kg/m^(2))were randomized(1:1)into the intervention group,which received weekly aerobic and muscle-strengthening PA videos,or control group,which received weekly general health education videos,for 12 weeks.Our primary outcome was free-living moderate-to-vigorous PA(MVPA)and our secondary outcomes were sedentary behavior,light PA,and sleep quality(measured using ActiGraph accelerometers)along with muscle-strengthening PA frequency,self-determination theory-related motivation(non-regulation,external regulation,introjected regulation,identified regulation,integrated regulation,and intrinsic regulation),and perceived PA barriers(assessed using validated questionnaires).Repeated measures analysis of variances(ANOVAs)examined between-group differences at an adjusted significance level of 0.004 and effect sizes as partial eta-squared(η;).Results:We observed statistically significant interaction effects for MVPA,sleep efficiency,muscle-strengthening PA frequency,non-regulation,integrated regulation,intrinsic regulation,and perceived PA barriers(F(1,62)=10.75-77.67,pdelivered

The role of soil in structure response of a building damaged by the 26 December 2018 earthquake in Italy

Local soil conditions can significantly modify the seismic motion expected on the soil surface.In most cases,the indications concerning the influence of the underlying soil provided by the in-force European and Italian Building Codes underestimate the real seismic amplification effects.For this reason,numerical analyses of the local seismic response(LSR)have been encouraged to estimate the soil filtering effects.These analyses are generally performed in free-field conditions,ignoring the presence of superstructures and,therefore,the effects of dynamic soil-structure interaction(DSSI).Moreover,many studies on DSSI are characterised by a sophisticated modelling of the structure and an approximate modelling of the soil(using springs and dashpots at the foundation level);while others are characterised by a sophisticated modelling of the soil and an approximate modelling of the structure(considered as a simple linear elastic structure or a single degree of freedom system).This paper presents a set of finite element method(FEM)analyses on a fully-coupled soil-structure system for a reinforced concrete building located in Fleri(Catania,Italy).The building,designed for gravity loads only,was severely damaged during the 26 December 2018 earthquake.The soil was modelled considering an equivalent visco-elastic behaviour,while the structure was modelled assuming both the visco-elastic and visco-inelastic behaviours.The comparison made between the results of the FEM analyses and the observed damage is valuable.

Forensic anthropologists perform specialised analysis,mainly involving skeletonised human remains,cadavers in advanced stages of decomposition,disassociated elements from the human body,and human remains in extreme carbonisation.The main objectives of the forensic anthropology expertise are human identification and assisting in determining the cause and manner of death.Estimating the time since death is also a priority for some cases,especially ones involving missing persons.This science works on individual cases,violent deaths,missing persons,mass disasters,suspected violations of human rights,and crimes against humanity.Forensic anthropological evidence is,in general,very sensitive.Thus,it is important to detail aspects relevant to the maintenance of the chain of custody at all phases of the investigation,as well as standardise the actions of the individuals involved.This aims to preserve the evidence integrity and sameness(Sameness:free translation of the Portuguese word“mesmidade”,derived from a Spanish word that does not possess a translation to Portuguese.Sameness of evidence is understood as the guarantee that the evidence under valuation(or under analysis of its probative value)is exactly and integrally the same one which was collected,corresponding,therefore,to“the same”(and not“part of”,“derived from”),safeguarding its value.Brazil Law No.13.964 of 14 December 2019 establishes and lists 10 phases related to the evidence chain of custody that must be followed.These newly introduced requirements resulted in the need for adaptation of the forensic,investigative,and legal actors involved in an investigation,and in the detailed description of the procedures for the different areas related to criminalistics,including forensic anthropology.The information provided in this article should be interpreted as recommendations,even though their non-compliance may weaken the investigative and forensic analysis processes in whole or in part.

Esophagitis Due to Actinomycosis in an Immunocompetent Patient with a History of SARS-Cov-2 Infection: Case Report

We present an unusual case of esophageal actinomycosis that developed in a patient with normal immunity and a history of acute SARS-CoV-2 infection. We report a case of a 56-year-old female patient without chronic degenerative pathologies with a history of non-severe acute SARS-CoV-2 infection that occurred two months prior to her presentation, treated with prednisone and betamethasone orally and inhaled, respectively for 21 days. The pivotal symptom is dysphagia and definitive diagnosis requires a tissue culture of the affected organ or a biopsy that shows the classic sulfur granules. Currently, antibiotic treatment with beta-lactams, such as amoxicillin/clavulanic acid, is still effective, as it was in our patient.
