Objective:This cross-sectional study aimed to determine the epidemiology of olfactory and gustatory dysfunctions related to COVID-19 in ***:This study was conducted by 45 tertiary Grade-A hospitals in *** and offline questionnaire data were obtained from patients infected with COVID-19 between December 28,2022,and February 21,*** collected information included basic demographics,medical history,smoking and drinking history,vaccination history,changes in olfactory and gustatory functions before and after infection,and other postinfection symptoms,as well as the duration and improvement status of olfactory and gustatory ***:Complete questionnaires were obtained from 35,566 *** overall incidence of olfactory and taste dysfunction was 67.75%.Being female or being a cigarette smoker increased the likelihood of developing olfactory and taste *** received four doses of the vaccine or having good oral health or being a alcohol drinker decreased the risk of such *** infection,the average olfactory and taste VAS scores were 8.41 and 8.51,respectively;after infection,they decreased to 3.69 and 4.29 and recovered to 5.83 and 6.55 by the time of the *** median duration of dysosmia and dysgeusia was 15 and 12 days,respectively,with 0.5%of patients having symptoms lasting for more than 28 *** overall self-reported improvement rate was 59.16%.Recovery was higher in males,never smokers,those who received two or three vaccine doses,and those that had never experienced dental health issues,or chronic accompanying ***:The incidence of dysosmia and dysgeusia following infection with the SARS-CoV-2 virus is high in *** and prognosis are influenced by several factors,including sex,SARS-CoV-2 vaccination,history of head-facial trauma,nasal and oral health status,smoking and drinking history,and the persistence of accompanying symptoms.

The Yangtze River is the mother river of *** promote the aquatic ecosystem protection of the great river,the Project of Yangtze Fisheries Resources and Environment Investigation(2017-2021)supported by the Ministry of Agriculture and Rural Affairs,*** carried out by 24 institutes and universities that located in the Yangtze River basin surveys the status of(1)fish species composition and spatial distribution,(2)current fish abundance,(3)endangered fishes,(4)Yangtze finless porpoise,(5)aquatic eco-environments,(6)water-level fluctuation areas,(7)capture fisheries and recreational fisheries of the Yangtze River mainstream and 10 of its main tributaries,including Yalong River,Heng River,Min River(including Dadu River),Chishui River,Tuo River,Jialing River,Wu River,Han River,Dongting Lake and Poyang *** results showed that there were 443 fish species(belonging to 163 genus,37 families,and 18 orders)before 2017,but only 323 fish species(including 15 new recorded exotic species)were recorded in the project of *** them,Cyprinus carpio,Carassius auratus,Hypophthalmichthys molitrix,Pelteobagrus fulvidraco,Coilia brachygnathus,Silurus asotus,Saurogobio dabryi,Ctenopharyngodon idellus,Pelteobagrus nitidus,Hemiculter leucisculus,Siniperca chuatsi,Coreius heterodon,Culter alburnus,Parabramis pekinensis,and Aristichthys nobilis were the dominant species across the whole Yangtze River *** is estimated that there were 886 million individuals weighing 124.8 million kg,merely equivalent to 27.3%of the resources in 1950s,30.9%of the resources in 1960s,or 58.7%of the resources in *** the new list of protected fishes that recorded in the Yangtze River system,only 15 of 29 were collected in this project of *** gladius has been affirmed to be Extinct by International Union for Conservation of Nature(IUCN).The wild individuals of Tenualosa reevesii and luciobrama macrocephalus have disappeared for many years and maybe have been extinct

Antifreeze proteins （AFPs） enable organisms to survive under cold conditions, and have great potential in improving cold tolerance of cold-sensitive plants, In order to determine whether expression of the carrot 36 kD antifreeze protein gene confers improved cold-resistant properties to plant tissues, we tried to obtain transgenic tobacco plants which expressed the antifreeze protein. Cold, salt, and drought induced promoter Prd29A was cloned using PCR from Arabidopsis. Two plant expression vectors based on pBI121 were constructed with CaMV35S：AFP and Prd29A：AFP. Tobacco plantlets were transformed by Agrobacterium-medicated transformation. PCR and Southern blotting demonstrated that the carrot 36 kD afp gene was successfully integrated into the genomes of transformed plantlets. The expression of the afp gene in transgenic plants led to improved tolerance to cold stress. However, the use of the strong constitutive 35S cauliflower mosaic virus （CaMV） promoter to drive expression of afp also resulted in growth retardation under normal growing conditions. In contrast, the expression of afp driven by the stress-inducible Prd29A promoter from Arabidopsis gave rise to minimal effects on plant growth while providing an increased tolerance to cold stress condition （2℃）. The results demonstrated the prospect of using Prd29A-AFP transgenic plants in cold-stressed conditions that will in turn benefit agriculture.

Ammopiptanthus mongolicus is the only evergreen broadleaved shrub endemic species to the Alashan Desert, northwest sand area of China, and can survive at -30℃ or even lower temperature in winter. A modified solid-phase subtraction hybridization technique was developed to isolate and screen cDNAs, whose transcripts increased in cold-treated A. mongolicus seedlings. Sequence analysis of the screened clones indicated that 11 clones had coding regions, with four of them containing complete open reading frame. And nine of the 11 clones shared various degrees of homology with the genes found in the GenBank database and the other two were unidentified sequences. Sequence data further revealed that these accumulated transcripts encode three low molecular weight proteins, a late-embryogenesis protein and two cold acclimation responsive proteins; two photosynthesis-related proteins, photosystem I subunit II precursor (PsaD) and photosystem II oxygen-evolving complex 33kDa subunit (OEC33); a protease inhibitor; an adenosine triphosphatase and a 14-3-3 related protein. Analysis on the function of these proteins indicated that the low molecular weight proteins were associated with water holding ability of cytoplasm; photosynthesis-related proteins participated in the adjustments of photosynthetic apparatus to resist photoinhibition; 14-3-3 related protein could interact with adenosine triphosphatase to enhance ATPase activity and energy metabolism; and protease inhibitor involved in the prevention of unwanted cell death was caused by reactive oxygen species. We suggest that cold acclimation with low light intensity in A. mongolicus is a more complex interaction of low temperature, light, energy and signal than that assumed previously.

该文集多种分离差异表达基因方法的优点于一体,自行优化了一种基于固相扣除杂交和聚合酶链式反应(PCR)技术直接克隆全长差异表达基因的策略.该策略中的所有操作步骤,包括poly(A)+mRNA的分离,cDNA的合成和扣除杂交都是在磁珠上进行的,操作简单易行.扣除杂交是用结合在磁珠上的以oligo(dT)为引物合成的对照材料(Driver)的cDNA第一链,直接扣除加有接头的处理材料(Tester)cDNA第二链中的共有序列.多次反复使用Driver一链与Tester二链进行杂交,能有效去除Tester二链中的共有序列.磁珠的分离能最小限度地减少每次杂交造成的损失.最后一轮扣除后的Tester二链中,含有大量差异表达序列,用Tester二链特有的接头引物进行扩增,能进一步提高扣除效率.该文选用此扣除方法,成功分离到了低温驯化沙冬青幼苗和对照苗2个群体的18个差异表达克隆,其中有4个克隆是全长cDNA序列.经序列分析发现,这些基因都与低温相关,进一步说明,该方法可用于克隆未知差异表达基因.

DNA甲基化是重要的植物基因组表观遗传修饰。植物中DNA甲基化的建立与维持是由多个调控因子协同作用的结果。不同的甲基转移酶类能直接作用于不同位点胞嘧啶甲基化,其中MET1主要负责保持原初CG位点的甲基化,CMT3主要负责保持CNG位点的甲基化,并由DRM与CMT3的协同从头甲基化作用来补偿其他相关序列的甲基化。这些甲基转移酶与染色质重塑解旋酶和组蛋白修饰因子协同改变染色质的结构,行使表观遗传的功能。DNA转葡糖基酶有去甲基化活性从而减轻基因沉默。文章综述了以上植物DNA甲基化调控因子的生物学功能及其之间的相互作用和近年来的研究进展,以更好的理解DNA甲基化的建立、保持和去除的机制。

北京颐和园西堤古柳组织培养体系的建立

古树名木是大自然经过漫长的历史变迁,留给人类的宝贵遗产,具有重要的科学、文化和经济价值。为了传承历史文化,留传历史园林植物的宝贵遗传种质资源,以北京颐和园西堤古柳嫩枝为外植体进行茎段组织培养试验研究。结果表明:取古柳树外植体带有侧芽的茎条切段进行组织培养,以MS为基本培养基,附加不同浓度的6-BA和NAA进行分化诱导,其中最适启动培养基为不添加任何激素的MS培养基(2.0%蔗糖);最适不定芽诱导培养基及激素配比为MS+6-BA0.8 mg/L+NAA 0.2 mg/L+3.0%蔗糖;不定根最适诱导培养基为1/2MS+NAA0.2mg/L+1.5%蔗糖。

"第十二届全国中药与天然药物药理学术会议"于2019年10月11-13日在天津市社会山国际会议中心酒店举行。会议期间,中国药理学会中药与天然药物药理专业委员会召开了"中药复方新药研发相关重大科学和技术问题"专题研讨会。研讨会分为主题报告和综合讨论2部分。主题报告由中国药理学会中药与天然药物药理专业委员会副主任委员吴春福教授主持,刘建勋、孙晓波、段大跃和吕圭源教授作主题报告。中国药理学会理事长兼中药与天然药物药理专业委员会主任委员张永祥研究员主持综合讨论,国家卫生健康委员会科技教育司重大专项处顾金辉处长全程参加。与会专家围绕中药复方新药研发相关重大科学和技术问题进行了广泛、深入的讨论。本文整理了专家们的主要观点和建议,希望能为中药复方新药研发提供新思路和新技术,为提高我国中药复方新药的研发水平发挥积极的推动作用。
