From common biomass materials to high-performance tissue engineering scaffold: Biomimetic preparation, properties characterization, in vitro and in vivo evaluations

Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials,but also increase the economic value.Chitosan as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable bioactivities.However,pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio,which seriously affected their in vivo stability and integrity when applied as tissue engineering scaf-folds.Thus,simultaneously improving the mechanical strength and biological compatibility of pure chitosan(CS)scaffolds becomes very important.Here,inspired by the fiber-reinforced con-struction of natural extracellular matrix and the porous structure of cancellous bone,we built silk microfibers/chitosan composite scaffolds via ice-templating technique.This biomimetic strategy achieved 500%of mechanical improvement to pure chitosan,and meanwhile still maintaining high porosity(>87%).In addition,the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and proliferation.More importantly,after subcutaneous implantation in mice for four weeks,the composite scaffold showed greater struc-tural integrity,as well as better collagenation,angiogenesis,and osteogenesis abilities,suggesting its great potential in biomedicine.

Increased long-distance and homo-trans interactions related to H3K27me3 in Arabidopsis hybrids

In plants,the genome structure of hybrids changes compared with their parents,but the effects of these changes in hybrids remain elusive.Comparing reciprocal crosses between Col×C24 and C24×Col in Arabidopsis using high-throughput chromosome conformation capture assay(Hi-C)analysis,we found that hybrid three-dimensional(3D)chromatin organization had more long-distance interactions relative to parents,and this was mainly located in promoter regions and enriched in genes with heterosis-related pathways.The interactions between euchromatin and heterochromatin were increased,and the compartment strength decreased in hybrids.In compartment domain(CD)boundaries,the distal interactions were more in hybrids than their parents.In the hybrids of CURLY LEAF(clf)mutants clfCol×clfC24and clfC24×clfCol,the heterosis phenotype was damaged,and the long-distance interactions in hybrids were fewer than in their parents with lower H3K27me3.ChIP-seq data revealed higher levels of H3K27me3 in the region adjacent to the CD boundary and the same interactional homo-trans sites in the wild-type(WT)hybrids,which may have led to more long-distance interactions.In addition,the differentially expressed genes(DEGs)located in the boundaries of CDs and loop regions changed obviously in WT,and the functional enrichment for DEGs was different between WT and clf in the longdistance interactions and loop regions.Our findings may therefore propose a new epigenetic explanation of heterosis in the Arabidopsis hybrids and provide new insights into crop breeding and yield increase.

Noncompaction of the ventricular myocardium(NVM),the third most diagnosed cardiomyopathy,is characterized by prominent trabeculae and intratrabecular recesses.However,the genetic etiology of 40%–60%of NVM cases remains unknown.Here,we identify two infants with NVM,in a nonconsanguineous family,with a typical clinical presentation of persistent bradycardia since the prenatal period.A homozygous missense variant(R223L)of RCAN family member 3(RCAN3)is detected in both infants using whole-exome sequencing.In the zebrafish model,marked cardiac dysfunction is detected in rcan3 deficiency(MO-rcan3ATG-injected)and rcan−/−embryos.Developmental dysplasia of both endocardial and myocardial layers is also detected in rcan3-deficient embryos.RCAN3 R223L variant mRNAs can not rescue heart defects caused by rcan3 knockdown or knockout;however,hRCAN3 mRNAs rescue these phenotypes.RNA-seq experiments show that several genes involved in cardiomyopathies are significantly regulated through multiple signaling pathways in the rcan3-knockdown zebrafish model.In human cardiomyocytes,RCAN3 deficiency results in reduced proliferation and increased apoptosis,together with an abnormal mitochondrial ultrastructure.Thus,we suggest that RCAN3 is a susceptibility gene for cardiomyopathies,especially NVM and that the R223L mutation is a potential loss-of-function variant.

A new method for identifying flow pattern of spouted fluidized bed by coupling Hilbert-Huang transform characteristics of differential pressure signals in different zones

On a cold spouted fluidized bed,this study compares the characteristic differences in intrinsic mode function(IMF)energy and Hilbert–Huang spectrum between the spout zone and annulus zone under different combinations of spouted gas and fluidized gas flow rates for five typical flow patterns.The energy distribution characteristics under different flow patterns are also analyzed.The Hilbert–Huang spectrum and IMF energy of pressure difference signals exhibit distinct variations in different zones as the flow pattern changes.Moreover,there exists a correlation between the energy in the middle-frequency range and the flow pattern.Leveraging the K-means algorithm,the middle-frequency range energy of IMFs in the spout zone and annulus zone is subjected to clustering analysis,leading to the identification of partition boundaries for each flow pattern.Based on this,a flow pattern identification method of spouted fluidized bed coupled with middle-frequency range energy in spout zone and annulus zone is proposed,which has very high identification accuracy.

Effect of short neuropeptide F signaling on larval feeding in Mythimna separata

Mythimna separata is a notorious phytophagous pest which poses serious threats to cereal crops owing to the gluttony of the larvae.Because short neuropeptide F(sNPF)and its receptor sNPFR are involved in a diversity of physiological functions,especially in functions related to feeding in insects,it is a molecular target for pest control.Herein,an sNPF and 2 sNPFRs were identified and cloned from M.separata.Bioinformatics analysis revealed that the sNPF and its receptors had a highly conserved RLRFamide C-terminus and 7 transmembrane domains,respectively.The sNPF and its receptor genes were distributed across larval periods and tissues,but 2 receptors had distinct expression patterns.The starvation-induced assay elucidated that sNPF and sNPFR expression levels were downregulated under food deprivation and recovered with subsequent re-feeding.RNA interference knockdown of sNPF,sNPFR1,and sNPFR2 by injection of double-stranded RNA into larvae not only suppressed food consumption and increased body size and weight,but also led to decrease of glycogen and total lipid contents,and increase of trehalose compared with double-stranded green fluorescent protein injection.Furthermore,molecular docking was performed on the interaction mode between sNPFR protein and its ligand sNPF based on the 3-dimensional models constructed by AlphaFold;the results indicated that both receptors were presumably activated by the mature peptide sNPF-2.These results revealed that sNPF signaling played a considerably vital role in the feeding regulation of M.separata and represents a potential control target for this pest.

The reaction of carbonyl-to-imine/hemiaminal conversion in the atmospheric aqueous phase is a critical pathway to produce the light-absorbing N-containing secondary organic compounds(SOC).The formation mechanism of these compounds has been wildly investigated in bulk solutions with a low ionic strength.However,the ionic strength in the aqueous phase of the polluted atmosphere may be higher.It is still unclear whether and to what extent the inorganic ions can affect the SOC formation.Here we prepared the bulk solution with certain ionic strength,in which glyoxal and ammonium were mixed to mimic the aqueous-phase reaction.Molecular characterization by High-resolution Mass Spectrometry was performed to identify the N-containing products,and the light absorption of the mixtures was measured by ultraviolet-visible spectroscopy.Thirty-nine N-containing compounds were identified and divided into four categories(N-heterocyclic chromophores,high-molecular-weight compounds with N-heterocycle,aliphatic imines/hemiaminals,and the unclassified).It was observed that the longer reaction time and higher ionic strength led to the formation of more N-heterocyclic chromophores and the increasing of the lightabsorbance of the mixture.The added inorganic ions were proposed to make the aqueous phase somewhat viscous so that the molecules were prone to undergo consecutive and intramolecular reactions to form the heterocycles.In general,this study revealed that the enhanced ionic strength and prolonged reaction time had the promotion effect on the lightabsorbing SOC formation.It implies that the aldehyde-derived aqueous-phase SOC would contribute more light-absorbing particulate matter in the industrial or populated area where inorganic ions are abundant.

Relationship between the Stratospheric Arctic Vortex and Surface Air Temperature in the Midlatitudes of the Northern Hemisphere

It is known that different relationships exist between the strength and displacement of the stratospheric polar vortex(SPV),and the surface air temperature(SAT)patterns in Eurasia and North America,but the mechanisms behind these relationships remain unclear,especially on an interannual timescale.Based on empirical orthogonal function(EOF)analysis using NCEP reanalysis data over 1958–2018,this study attempts to ascertain the relationship between the SPV intensity and displacement over the Arctic and the SATs in the midlatitudes of the Northern Hemisphere.Our results indicate that a strengthened SPV corresponds to an SAT increase in Eurasia and a decrease in eastern North America and Greenland.When the SPV is shifted towards Eurasia,however,a corresponding SAT increase occurs in both North America and Eurasia,with a larger increase in North America than in Eurasia.Specifically,a strengthened SPV tends to correspond to a positive North Atlantic Oscillation-like circulation in the troposphere with negative geopotential height(GH)anomalies in Greenland and eastern North American continent and positive GH anomalies to the north of 45°N in Eurasia,which corresponds to lower SATs in North America than in Eurasia.However,when the SPV shifted towards Eurasia,it was accompanied by a positive Pacific/North American-like pattern with a deepened Aleutian low,which corresponds to the increasing SATs in North America.These tropospheric circulation changes are related to the response of tropospheric planetary wave activity to the SPV.A strengthened SPV corresponds to the weakening of tropospheric planetary wave-1 waves,which is accompanied by a negative GH in North America but a positive GH in Eurasia.If the SPV shifted towards Eurasia,the tropospheric planetary wave-1(-2)waves strengthened(weakened),and the combined effects of the planetary wave-1 and wave-2 waves would cause positive GH anomalies in both Eurasia and North America.

Research progress on short-term mechanical properties of FRP bars and FRP-reinforced concrete beams

Fiber-reinforced polymer(FRP)bars have been increasingly recognized in the field of civil engineering due to their advantages of light weight,high strength and excellent durability.FRP bars can replace steel bars in concrete beams and effectively improve the durability of beams.In this paper,the literature relevant to the short-term mechanical properties of FRP bars and FRP-reinforced concrete beams was reviewed based on previous studies and practical engineering application.Firstly,the mechanical properties of FRP bars were reviewed.Different types of fibers or steel and fibers can be combined to obtain hybrid fiber-reinforced polymer(HFRP)or steel-fiber composite bars(SFCB)with excellent mechanical performance,respectively.The bond performance and bond-slip model between FRP bars and concrete were discussed.Several common bond-slip models were usually used to study the bond performance between carbon fiber-reinforced polymer(CFRP)bars or glass fiber-reinforced polymer(GFRP)bars and concrete,but changing the type of FRP bars will lead to larger dispersion.Then,the experimental studies,theoretical calculation methods and finite element simulation methods of flexural/shear behavior of FRP-reinforced concrete beams were presented.Finally,their applications in practical engineering were discussed and the prospects of further research were proposed.It is pointed out that FRP-reinforced ultra-high performance concrete(UHPC)beams,FRP-reinforced geopolymer concrete(GPC)beams,engineered cementitious composites(ECC)-FRP-reinforced concrete beams,prestressed FRP-reinforced concrete beams and steel/FRP hybrid-reinforced concrete beams can effectively improve the deformation resistance and poor ductility of pure FRP-reinforced concrete beams.

Rational Design of Cost-Effective Metal-Doped ZrO_(2)for Oxygen Evolution Reaction

The design of cost-effective electrocatalysts is an open challenging for oxygen evolution reaction(OER)due to the“stable-oractive”dilemma.Zirconium dioxide(ZrO_(2)),a versatile and low-cost material that can be stable under OER operating conditions,exhibits inherently poor OER activity from experimental observations.Herein,we doped a series of metal elements to regulate the ZrO_(2)catalytic activity in OER via spin-polarized density functional theory calculations with van der Waals interactions.Microkinetic modeling as a function of the OER activity descriptor(G_(O*)-G_(HO*))displays that 16 metal dopants enable to enhance OER activities over a thermodynamically stable ZrO_(2)surface,among which Fe and Rh(in the form of single-atom dopant)reach the volcano peak(i.***.the optimal activity of OER under the potential of interest),indicating excellent OER performance.Free energy diagram calculations,density of states,and ab initio molecular dynamics simulations further showed that Fe and Rh are the effective dopants for ZrO_(2),leading to low OER overpotential,high conductivity,and good stability.Considering cost-effectiveness,single-atom Fe doped ZrO_(2)emerged as the most promising catalyst for OER.This finding offers a valuable perspective and reference for experimental researchers to design cost-effective catalysts for the industrial-scale OER production.

Effect of Sc on Al_(3)Fe phase and mechanical properties of as-cast AA5052 aluminum alloy

The AA5052 aluminum alloy is widely used in automobile and aerospace manufacturing,and with the development of light-weight alloys,it is required that these materials exhibit better mechanical properties.Previous studies have demonstrated that the addition of Sc to aluminum alloys can improve both the microstructure and properties of the alloys.In this study,the effect of Sc on the Fe-rich phase and properties of the AA5052 aluminum alloy was studied by adding 0%,0.05%,0.2%,and 0.3%Sc.The results show that with the increase of Sc,the coarse needle-like Fe-rich phase gradually transforms into Chinese-script and then nearly spherical particles,reduce the size of Fe-rich phase,and refine the grain with increase of high angle grain boundaries(HAGBs).These microstructure changes enhance the strength of the AA5052 alloy through Sc addition.The ductility of the alloy is obviously improved because the addition of a lower amount of Sc changes the morphology of Fe-rich phase from needle-like into a Chinese-script,and it is subsequently reduced as a result of significant increase in HAGBs with increasing Sc content.
