Wheat production is often affected by different types of rust diseases,in which stripe rust is triggered by a fungal pathogen,Puccinia striiformis ***(Pst),and remains the biggest challenge to plant breeders and the farming ***,pathogens exploit its effector proteins for altering the susceptibility(S)genes in the host plant,rendering host defense ***,altering the S genes by employing precise genome editing tools provides a great opportunity to develop broad-spectrum and durable,resistant ***,Wang et al.,2022 have demonstrated the TaPsIPK1 as an ideal S gene target to confer stripe rust resistance in wheat,providing an opportunity to deliver broad-spectrum,rust-resistant crop varieties.

目的:胰岛素样生长因子(IGFs)是DNA序列与胰岛素非常相似的蛋白质。IGF是细胞用来与周围环境通讯的复杂系统的一部分,它们在促进细胞增殖和抑制细胞死亡或凋亡方面发挥着作用。研究表明,胰岛素/IGF轴在衰老中起着重要作用,在诸如癌症和糖尿病等疾病中也起着重要作用,例如,IGF-1既可以刺激前列腺癌细胞也可以刺激乳腺癌细胞生长。方法:在大肠杆菌中表达与组织纤溶酶原激活剂(TPAST)融合至胰岛素样生长因子(IGF1)的重组腺皮质激素(ACTH)重组融合蛋白(His Tag-Linker-ACTH-TPAST-IGFI-PET21)。并使用Ni-NTA琼脂糖珠进行纯化。IGF1通过TPAST底物接头与ACTH融合的原因是因为在血液中存在组织纤溶酶原激活剂(TPAST),它将作用于TPAST底物接头并切割该部位以释放ACTH和IGF1。IGFI可以使ACTH的效果提高20倍以上。(1)使用Primer Express软件设计wt ACTH,TPAst,IGF1的引物F和R。(2)使用蛋白酶数据库分析了ACTH中的蛋白酶(存在于血液中)的切割位点,并通过重叠PCR方法创建了融合ACTH-IPAst-IGF1的三个基因。(3)接下来,通过使用限制酶消化物连接到p ET21a载体中,构建融合表达质粒(His Tag-Linker-ACTH-TPAST-IGFI-PET21)。(4)将重组质粒转化到大肠杆菌BL21-DE3菌株中进行蛋白表达。(5)在分析ACTH-TPAst-IGF1融合的重组蛋白的表达是否在上清液或沉淀级分中后,使用Ni-NTA琼脂糖珠进一步纯化。(6)由于蛋白质在大肠杆菌的沉淀级分中表达,因此由于形成包涵体而使其变性和微粒化。因此,需要对其进行复性和溶解以使其重折叠以增加活性。(7)下一步是在基于体外细胞的测定和体内测定中测试该蛋白的功能。结果:(1)通过重叠PCR产生融合的基因产物,然后亚克隆到p ET21a载体中。(2)构建野生型(WT)ACTH-TPAST-IGFI的质粒,然后通过DNA测序进行验证:(3)从大肠杆菌菌株BL21-DE3中表达并纯化以上所有物质。在SDS-PAGE凝胶上进行了考马斯亮蓝染色,显示了14kd融合蛋白在IPTG诱导培养物的沉淀级分和蛋白质纯化级分后的洗脱级分中的表达。(4)但是,由于蛋白质不溶,因此将其透析并重新折叠并浓缩蛋白质。(1)分子实验结果1.质粒图谱2.双重酶促裂解3.将融合基因PCR产物克隆到载体中***测序5.重组融合蛋白在大肠杆菌中的表达6.蛋白质纯化和洗脱蛋白的复性。***-PAGE。凝胶用可可蓝染色。***法测定小鼠中c AMP,CREB和Gluco皮质类固醇浓度的产生(由于Covid-19的当前情况,有待完成)结论:(1)我们成功地在大肠杆菌中以沉淀级分克隆并表达了重组融合蛋白。(2)使用Ni-NTA磁珠对该14KD蛋白进行蛋白纯化得到纯洗脱级分,纯蛋白浓度为＿(3)我们成功地通过透析使14kd融合蛋白的蛋白复性。

This communication addresses the impact of heat source/sink along with mixed convection on oblique flow of Casson fluid having variable viscosity. Similarity analysis has been utilized to model governing equations, which are simplified to set of nonlinear differential equations. Computational procedure of shooting algorithm along with 4 th order Range-Kutta-Fehlberg scheme is opted to attain the velocity and temperature distributions. Impact of imperative parameters on Casson fluid flow, temperature, significant physical quantities such as skin friction, local heat flux and streamlines are displayed via graphs.

Soil-borne pathogens,e.g.,Plasmodiophora brassicae,causes devastating clubroot disease,and existing identified resistance(R)genes has been broken down with the co-evolution of pathogen,threatening the future of cruciferous ***,there is a dire need to identify new R genes for developing clubroot-resistant ***,Wang et al.,2023 have reported a new R gene,WeiTsing,that confers complete resistance to Plasmodiophora brassicae,providing an opportunity to deliver broad-spectrum,durable clubroot-resistant crop varieties.

The outbreak of the novel coronavirus has spread worldwide,and millions of people are being *** or detection classification is one of the first application areas of deep learning,which has a significant contribution to medical image *** classification detection,one or more images(detection)are usually used as input,and diagnostic variables(such as whether there is a disease)are used as *** novel coronavirus has spread across the world,infecting millions of ***-stage detection of critical cases of COVID-19 is essential.X-ray scans are used in clinical studies to diagnose COVID-19 and Pneumonia *** extracting the discriminative features through these modalities,deep convolutional neural networks(CNNs)are used.A siamese convolutional neural network model(COVID-3D-SCNN)is proposed in this study for the automated detection of COVID-19 by utilizing X-ray *** extract the useful features,we used three consecutive models working in parallel in the proposed *** acquired 575 COVID-19,1200 non-COVID,and 1400 pneumonia images,which are publicly *** our framework,augmentation is used to enlarge the *** findings suggest that the proposed method outperforms the results of comparative studies in terms of accuracy 96.70%,specificity 95.55%,and sensitivity 96.62%over(COVID-19 ***-COVID19 ***).

Boundary layer stagnation point flow of Casson fluid over a Riga plate of variable thickness is investigated in present article. Riga plate is an electromagnetic actuator consists of enduring magnets and gyrated aligned array of alternating electrodes mounted on a plane surface. Physical problem is modeled and simplified under appropriate transformations. Effects of thermal radiation and viscous dissipation are incorporated. These differential equations are solved by Keller Box Scheme using MATLAB. Comparison is given with shooting techniques along with RangeKutta Fehlberg method of order 5. Graphical and tabulated analysis is drawn. The results reveal that Eckert number,radiation and fluid parameters enhance temperature whereas they contribute in lowering rate of heat transfer. The numerical outcomes of present analysis depicts that Keller Box Method is capable and consistent to solve proposed nonlinear problem with high accuracy.

Cerebral Microbleeds(CMBs)are microhemorrhages caused by certain abnormalities of brain *** can be found in people with Traumatic Brain Injury(TBI),Alzheimer’s disease,and in old individuals having a brain *** research reveals that CMBs can be highly dangerous for individuals having dementia and *** CMBs seriously impact individuals’life which makes it crucial to recognize the CMBs in its initial phase to stop deterioration and to assist individuals to have a normal *** existing work report good results but often ignores false-positive’s perspective for this research *** this paper,an efficient approach is presented to detect CMBs from the Susceptibility Weighted Images(SWI).The proposed framework consists of four main phases(i)making clusters of brain Magnetic Resonance Imaging(MRI)using k-mean classifier(ii)reduce false positives for better classification results(iii)discriminative feature extraction specific to CMBs(iv)classification using a five layers convolutional neural network(CNN).The proposed method is evaluated on a public dataset available for 20 *** proposed system shows an accuracy of 98.9%and a 1.1%false-positive rate *** results show the superiority of the proposed work as compared to existing states of the art methods.

Education sector has witnessed several changes in the recent *** changes have forced private universities into fierce competition with each other to get more students *** competition has resulted in the adoption of marketing practices by private universities similar to commercial *** get competitive gain,universities must observe and examine the students’feedback on their own social media sites along with the social media sites of their *** study presents a novel framework which integrates numerous analytical approaches including statistical analysis,sentiment analysis,and text mining to accomplish a competitive analysis of social media sites of the *** techniques enable local universities to utilize social media for the identification of the most-discussed topics by students as well as based on the most unfavorable comments received,major areas for improvement.A comprehensive case study was conducted utilizing the proposed framework for competitive analysis of few top ranked international universities as well as local private universities in Lahore *** results show that diversity of shared content,frequency of posts,and schedule of updates,are the key areas for improvement for the local *** on the competitive intelligence gained several recommendations are included in this paper that would enable local universities generally and Riphah international university(RIU)Lahore specifically to promote their brand and increase their attractiveness for potential students using social media and launch successful marketing campaigns targeting a large number of audiences at significantly reduced cost resulting in an increased number of enrolments.
