Juvenile Idiopathic Arthritis and Its HLA Association: A Study from Bangladesh

Background: HLA alleles may have association with overall JIA including specific sub-types. Determation of HLA DRB1, DPB1, DQA1, DQB1 and B27, may be helpful to diagnose JIA cases where diagnostic dilemma is present. The aim of the study was to find out the association of HLA alleles with JIA and its subtypes. Methods: This cross sectional study was conducted in the department of Pediatrics, Bangabandhu Sheikh Mujib Medical University (BSMMU), Dhaka, from April 2020 to March 2021. A total of 50 cases and 25 controls attending pediatric rheumatology clinic and pediatric OPD were enrolled in the study. Data were collected by a pre-designed questionnaire containing socio-demographic, clinical and laboratory parameters. HLA DR-B1, DQ-A1, DQ-B1 and B27 typing were done for the cases and controls from blood samples by polymerase chain reaction sequence specific primer (PCR-SSP) method. Data were analyzed by SPSS version 26. Frequency, percentage, chi square test and Fisher exact tests were done for statistical analysis. Results: HLA DR-B1_10 had significant positive association whereas HLA DR-B1_03 and B1_16 had negative association with overall JIA cases. Poly-articular JIA, oligo-articular JIA and ERA had association with HLA DRB1_01, B1_08 and B1_12 respectively. HLA DQA1-01 and A1_02 had positive association. However, DQ-A1_06 was negatively associated with JIA cases. Oligo JIA was positively associated with HLA DQ-A1_01, whereas poly JIA and ERA had association with A1_02. Though, HLA DQ_B1 had no association with overall JIA cases, analysis showed association of poly JIA with DQ-B1_04. Frequency of HLA DQ-B1_02 and 03 were higher among ERA cases, though not significant, but they had strong association with HLA B27. Conclusion: HLA associations were present with overall JIA and each sub-groups had different patterns of HLA associations including some protective roles.

Enterobacteriaceae are important human pathogens that cause many food and waterborne illness. Rapid emergence of multi-drug resistant bacteria in Bangladesh has become a serious problem. Phage-host interaction is now considered as the major driving force for the conversion of non-pathogenic bacteria to pathogenic ones. Evolution of highly pathogenic and antibiotic resistant bacteria largely depends upon the horizontal gene transfer by means of plasmid, megaplasmid and bacteriophages. Conversely, bacteria may acquire a novel defence mechanism called CRISPR (Clustered regularly interspersed short palindromic repeats) that can restrict horizontal transfer of plasmids and bacteriophages to limit the spread of antibiotic resistance genes among bacterial species. In this study, twenty bacterial strains were isolated from water of different medical waste and Buriganga river. Therefore, CRISPR locus was investigated following various biochemical and molecular analysis of those bacterial isolates. Identification of the bacterial isolates was conducted by Polymerase Chain Reaction (PCR) based assay of 16S rDNA extracted from those isolated strains. Results indicated that most strains were identified as Proteus mirabilis and Citrobacter freundii which mainly cause septicemia, and pneumonia in human. Thereafter, antibiogram of these strains was performed by using 11 different antibiotic discs where bacterial isolates from medical drainage system showed more resistant to antibiotics than the river water. In this study, CRISPR locus was also investigated within the genome of the isolated bacterial stains but unexpectedly, we did not find any CRISPR locus in their genome. In conclusion, we confirm that multi-drug resistant bacterial strains are devoid of CRISPR locus suggesting a possible negative association between CRISPR locus and antibiotic resistance. In the recent years, it has been established that b/h or h/b (b = length of the structure in the lateral direction and h = approach flow depth) is the dominant factor for the estimation of local scour depth around abutments and piers. One of the major limitations of the Lacey formula is that it predicts only one value of the scour depth for a specific river because of the absence of important parameters b and h. Therefore, its application is quite uncertain for variable b and h. Considering its limitations and experimental facts of flow concentration and shear stress amplification close to above structures, the original Lacey formula is modified introducing the parameters b and h. The applicability of the original as well as modified Lacey formula is tested to the data collected from major rivers in Bangladesh.

Reproductive stage frost poses a major constraint for wheat production in countries such as ***,little progress has been made in identifying key genes to overcome the *** the present study,a severe frost event hit two large-scale field trials consisting of six doubled haploid(DH)wheat populations at reproductive stage(young microspore stage)in Western Australia,leading to the identification of 30 robust frost QTL on 17 *** major 18 QTL with the phenotype variation over 9.5%were located on 13 chromosomes including 2 A,2 B,2 D,3 A,4 A,4 B,4 D,5 A,5 D,6 D,7 A,7 B and7 *** frost QTL were closely linked to the QTL of anthesis,maturity,Zadok stages as well as linked to anthesis related *** of those,six QTL were repetitively detected on the homologous regions on 2 B,4 B,4 D,5 A,5 D,7 A in more than two *** showed that the frost damage is associated with alleles of Vrn-A1 a,Vrn-D1 a,Rht-B1 b,Rht-D1 b,and the high copy number of ***,anthesis QTL and anthesis related genes of Vrn-B1 a and Ta FT3-1 B on chromosomes 5 B and 1 B did not lead to frost damage,indicating that these early-flowering phenotype related genes are compatible with frost tolerance and thus can be utilised in *** results also indicate that wild-type alleles Rht-B1 a and Rht-D1 a can be used when breeding for frost-tolerant varieties without delaying flowering time.

Dynamic polymers with both physical interactions and dynamic covalent bonds exhibit superior performance,but achieving such dry polymers in an effi-cient manner remains a ***,we report a novel organic solvent quenched polymer synthesis using the natural molecule thioctic acid(TA),which has both a dynamic disulfide bond and carboxylic *** effects of the solvent type and concentration along with reaction times on the proposed reaction were thoroughly explored for polymer ***-state proton nuclear magnetic resonance(1 H NMR)and first-principles simulations were carried out to investigate the reaction *** show that the chlorinated solvent can efficiently stabilize and mediate the depolymerization of poly(TA),which is more kinetically favorable upon lowering the *** to the numerous dynamic covalent disulfide bonds and noncovalent hydrogen bonds,the obtained poly(TA)shows high extensibility,self-healing,and reprocessable *** can also be employed as an efficient adhesive even on a Teflon surface and 3D printed using the fused deposition modeling *** new polymer synthesis approach of using organic solvents as catalysts along with the unique reaction mechanism provides a new pathway for efficient polymer synthesis,especially for multifunctional dynamic polymers.

Objective:To investigate the clot lysis,antimicrobic and membrane stabilizing potentials of ethanolic extractives of the leaves of Heliotropium indicum(***).Methods:Crude ethanolic extracts of *** leaves were partitioned successively using solvents of different polarity and subjected to determine qualitatively and quantitatively for the presence of different bioactive constitutes and fractions which were assessed for their possible clot lysis,antimicrobic,and membrane stabilizing activities as compared with the known ***:For the thrombolytic activity,the petroleum ether soluble fraction showed the highest percent of clot lysis(35.4%)among all fractions,while streptokinase and water resulted in 65.15%and 3.77%clot lysis,*** respect to the membrane stabilizing activity,carbon tetrachloride soluble fraction of *** profoundly interdicted the hemolysis of erythrocytes brought about by osmotic induction(39.24%)or by heat(40.20%).The other fractions exhibited less significant membrane stabilizing *** contrast,acetylsalicylic acid resulted in 72.25%±0.30%inhibition of osmotically induced hemolysis and showed a lower level of protection of heat induced hemolysis(42.56%).Crude ethanolic extracts were moderately sensitive against known pathogenic *** pharmacological activities of *** are due to the presence of bioactive compounds,we detected and quantified the presence of significant levels of flavonoid and tannin ***:The outcomes of this research show that the leaves of *** have the potential to be used as a remedy for thrombosis,inflammatory diseases and against few important bacterial pathogens.

Background: Patients with chronic kidney disease (CKD) are at increased risk of morbidity & mortality. Educational interventions aimed at empowering patients are successful in chronic disease management including CKD. Objective: To explore the awareness regarding CKD among patients attending in a tertiary care hospital in Bangladesh. Methodology: This was a descriptive observational study, which includes 100 adult patients attending the department of Medicine in Bangabandhu Sheikh Mujib Medical University (BSMMU), Dhaka, Bangladesh from January 2013 to June 2013. Data were collected on a pre-tested questionnaire by face-to-face interview to investigate awareness toward: 1) basic knowledge of personal health;2) perceptions of factors increasing the risk of CKD;3) knowledge of therapies to slow CKD progression;4) perceptions of CKD increasing the risk of other medical conditions and 5) demographic information. Main outcome variables were demographic parameters, socio-economic status and awareness about chronic kidney disease. Data were analyzed and compared by statistical tests. Results: Almost one third (32.0%) respondents were in 3rd decade and male to female ratio was 1:1.5. Majority (43.0%) patients came from lower-middle income family. In all age groups, the majority (60%) respondents had low knowledge of CKD. Among the study population, 44.0% believe that smoking increases risk of CKD;42.0% believe that restricting salt intake reduces the progression of CKD;34.0% mention that CKD increases the risk for hypertension and 82.0% mention that renal transplantation is the treatment of choice in CKD. Among the participants, 32.8% had knowledge of increasing risk factor of CKD, 30.8% had knowledge of the method of slow progression of CKD, 30.3% had knowledge of conditions for increase risk of CKD and 41.7% respondent had knowledge of treatment of CKD.
