Iridoviruses are DNA virus and have caused huge economic losses in the aquaculture *** aim of this study was to establish a colorimetric loop-mediated isothermal amplification(LAMP)protocol for the on-site detection of Singapore grouper iridovirus(SGIV).The SGIV-VP61 gene was chosen as the target gene to develop a colorimetric LAMP *** optimized condition of the colorimetric LAMP assay was incubation at 63℃ for 1 *** infected with SGIV could be detected with the color change from yellow into *** sensitivity of the developed assay is 5.66 copies/μL of the viral DNA *** sensitivity was about 1000 times higher than that of conventional PCR while it was slightly lower than the one-step semi-nested PCR assay.A total of 60 DNA samples extracted from the fin tissue of the SGIV-infected Asian seabass were examined for SGIV by colorimetric LAMP,semi-nested PCR and conventional *** results of the colorimetric LAMP assay showed 94.87%agreement with the semi-nested *** addition,the DNA extraction method using NaOH showed a better performance in the colorimetric LAMP *** together,the colorimetric LAMP established was a sensitive,rapid and specific method for the detection of *** was not detected in samples randomly taken from a genetically improved line of the Asian ***,some seabass obtained from the local markets were found to contain ***,the LAMP assay has the potential application in the diagnosis of iridovirus diseases in the aquaculture industry.

Asian seabass(Lates calcarifer)is an important foodfish *** has been cultured in Singapore since the 1980s.A selective breeding program to improve growth,meat quality and disease resistance has been ongoing in Singapore since *** 2012,outbreaks of the big belly disease(BBD)occurred in two batches of offspring generated by a mass-cross among F1 brooders in a *** the outbreaks,only 437 survived in 6000 F2 *** parentage analysis with 10 microsatellites revealed that four parents(F1)produced high percentage of offspring that survived the BBD ***-two BBD survivor offspring(F2)were selected based on their growth performance at 90 days post hatch(dph),family origin and allelic *** were used as brooders to generate *** a period of five years,these F2 brooders produced six batches of offspring,where no outbreak of BBD *** the same period,outbreaks of BBD occurred in 4 out of 18 batches produced by F2 brooders selected for growth,with mortality rates ranging from 85.1%to 95.4%.In F3,the average body weight(57.1±1.09 g,n=3068)of the line for BBD survival was slightly lower than that(60.6±2.29 g,n=4280)of the line for growth at 90 *** both lines,the body weight of F3 was much higher than that(46.90±0.19 g,n=12117)of *** genetic diversity in the line for BBD survival(F3)was much lower than that in F3 line for growth due to small founder *** data indicate that the selection of BBD survival during BBD outbreaks using molecular parentage analysis is effective to reduce BBD *** further reduction of genetic diversity is essential in the line selected for increased survival against BBD.

Diesel soot aggregates emitted from a model dynamometer and 11 on-road vehicles were segregated by a micro-orifice uniform deposit impactor （MOUDI）. The elemental contents and morphological parameters of the aggregates were then examined by scanning electron microscopy coupled with an energy dispersive spectrometer （SEM-EDS）, and combined with a fractional Brownian motion （fBm） processor. Two mode-size distributions of aggregates collected from diesel vehicles were confirmed. Mean mass concentration of 339 mg/m3 （dC/dlogdp） existed in the dominant mode （180-320 nm）. A relatively high proportion of these aggregates appeared in PM 1, accentuating the relevance regarding adverse health effects. Furthermore, the fBm processor directly parameterized the SEM images of fractal like aggregates and successfully quantified surface texture to extract Hurst coefficients （H） of the aggregates. For aggregates from vehicles equipped with a universal cylinder number, the H value was independent of engine operational conditions. A small H value existed in emitted aggregates from vehicles with a large number of cylinders. This study found that aggregate fractal dimension related to H was in the range of 1.641-1.775, which is in agreement with values reported by previous TEM-based experiments. According to EDS analysis, carbon content ranged in a high level of 30%-50% by weight for diesel soot aggregates. The presence of Na and Mg elements in these sampled aggregates indicated the likelihood that some engine enhancers composed of biofuel or surfactants were commonly used in on-road vehicles in Taiwan. In particular, the morphological H combined with carbon content detection can be useful for characterizing chain-like or cluster diesel soot aggregates in the atmosphere.

Background: This study aims to investigate the moderating effect of family health status on the relationship between involvement in housework of two-side parents’ families and happiness of middle-aged women in Taiwan. Methods: The data used in this study were gathered from the Ministry of Science and Technology’s program—“Intergenerational expectation and collaboration: an inquiry on a new mechanism of continued tradition across generations”. The subjects (n = 512) were restricted to aged 45 - 64 years old who live with spouses and have children, and one of whose parent or parent-in-law is still alive. Multiple regression analysis was used to investigate how family health status moderates the relationship between involvement in housework of parents’ families and parents-in-law’s families and the happiness of middle-aged women. Results: 1) Family health status is positively correlated with their happiness. 2) The parents’ health status can moderate the relationship between involvement in housework of parents’ families and happiness of the middle-aged women. 3) The middle-aged women’s health status can moderate the relationship between involvement in housework of parents-in-law’s families and those women’s happiness, and fathers-in-law’s health status can moderate the relationship between husbands’ involvement in housework of the women’s parents-in-law’s families and women’s happiness. Conclusions: In future, when policies and programs related to the well-being of middle-aged women are being planned, the effect of women’s health and family health status shall be considered as a key to improve those women’s quality of life.

Background: Studies have pointed out the influence of different children’s activities and prolonged use of digital products on their social development. However, whether the parent-child activities and using digital devices were serial mediators of the relationship between children’s health and social development needs further verification. Purpose: This study explored how parent-child activities and children’s use of digital devices influence the relationship between children’s health and their social competence. Method: This study used data from Kids in Taiwan: National Longitudinal Study of Child Development and Care. A total sample of 2164 participants was used in this study. Serial mediation analyses were performed using model six of Hayes’ PROCESS (2012). Results: This study found that parent-child activities and the use of digital devices can serially mediate the relationship between children’s health and social competence. Children’s health could directly improve their social competence, but it could also serially mediate social competence by increasing parent-child activities and reducing the use of digital devices. Conclusion: Childcare policy planners and parenting educators should not only call on parents to reduce the use of electronic products for their children, but also encourage parents to spend more time interacting with their children, so that children can learn social skills by interacting with others in their daily lives.

Alternatively activated macrophages （M2 macrophages） promote central nervous system regeneration. Our previous study demonstrated that treatment with peripheral nerve grafts and fibroblast growth factor-1 recruited more M2 macrophages and improved partial functional recovery in spinal cord transected rats. The migration of macrophages is matrix metalloproteinase （MMP） dependent. We used a general inhibitor of MMPs to influence macrophage migration, and we examined the migration of macrophage populations and changes in spinal function. Rat spinal cords were completely transected at Ts, and 5 mm of spinal cord was removed （group T）. In group R, spinal cord-transected rats received treatment with fibroblast grow th factor- 1 and peripheral nerve grafts. In group RG, rats received the same treatment as group R with the addition of 200 μM GM6001 （an MMP inhibitor） to the fibrin mix. We found that MMP-9, but not MMP- 2, was upregulated in the graft area of rats in group R. Local application of the MMP inhibitor resulted in a reduction in the ratio of arginase-1 （M2 macrophage subset）/inducible nitric oxide synthase-postive cells. When the MMP inhibitor was applied at 8 weeks postoperation, the partial functional recovery observed in group R was lost. This effect was accompanied by a decrease in brain-derived neurotrophic factor levels in the nerve graft. These results suggested that the arginase-1 positive population in spinal cord transected rats is a migratory cell population rather than the phenotypic conversion of early iNOS^＋ cells and that the migration of the arginase-1^＋ population could be regulated locally. Simultaneous application of MMP in- hibitors or promotion of MMP activity for spinal cord injury needs to be considered if the coadministered treatment involves M2 recruitment.
