The most abundant immunoglobulin present in the human body is IgA. It has the highest concentrations at the mucosal lining and in biofluids such as milk and is the second most abundant class of antibodies in serum. We assessed the structural diversity and clonal repertoire of IgA1-containing molecular assemblies longitudinally in human serum and milk from three donors using a mass spectrometry-based approach. IgA-containing molecules purified from serum or milk were assessed by the release and subsequent analysis of their Fab fragments. Our data revealed that serum IgA1 consists of two distinct structural populations, namely monomeric IgA1 (∼80%) and dimeric joining (J-) chain coupled IgA1 (∼20%). Also, we confirmed that IgA1 in milk is present solely as secretory (S)IgA, consisting of two (∼50%), three (∼33%) or four (∼17%) IgA1 molecules assembled with a J-chain and secretory component (SC). Interestingly, the serum and milk IgA1-Fab repertoires were distinct between monomeric, and J-chain coupled dimeric IgA1. The serum dimeric J-chain coupled IgA1 repertoire contained several abundant clones also observed in the milk IgA1 repertoire. The latter repertoire had little to no overlap with the serum monomeric IgA1 repertoire. This suggests that human IgA1s have (at least) two distinct origins;one of these produces dimeric J-chain coupled IgA1 molecules, shared in human serum and milk, and another produces monomeric IgA1 ending up exclusively in serum.

We present how the applied ar background pressure correlates to the cation and anion composition of Cu(I) chalcogenide thin films grown by pulsed laser deposition(PLD). The as-grown films produced at ~10-2 mbar p ar show a more pronounced deficiency in lighter composition, as compared with using quasi-vacuum or ~10-1 mbar. The thermoelectric performance of the as-grown Cu2 Se varies consistently with the respective changes in Cu/Se ratio vs. p ar. The optimum thermoelectric performance of Cu2 Se is achieved by growing in vacuum or quasi-vacuum, where dense and even thin film morphology is obtained while the cation/anion composition is more congruent than at higher p ar.

XMM-Newton observation of the eclipsing binary Algol

We present an XMM-Newton observation of the eclipsing binary Algol which contains an X-ray dark B8V primary and an X-ray bright K2IV secondary. The observation covered the optical secondary eclipse and captured an X-ray flare that was eclipsed by the B star. The XMM-Newton European Photon Imaging Camera and Reflection Grating Spectrometer spectra of Algol in its quiescent state are described by a two-temperature plasma model. The cool component has a temperature around 6.4× 106 K while that of the hot component ranges from 2 to 4.0× 107 K. Coronal abundances of C, N, O, Ne, Mg, Si and Fe were obtained for each component for both the quiescent and the flare phases, generally with upper limits for S and ar, and upper limits for C, N, and O from the hot component. F-tests show that the abundances do not need to be different between the cool and the hot component and between the quiescent and the flare phase with the exception of Fe. Although the Fe abundance of the cool component remains constant at -0.14, the hot component shows an Fe abundance of -0.28, which increases to -0.44 during the flare. This increase is expected from the chromospheric evaporation model. The absorbing column density NH of the quiescent emission is 2.5 - 1020 cm-2, while that of the flare-only emission is significantly lower and consistent with the column density of the interstellar medium. This observation substantiates earlier suggestions of the presence of X-ray absorbing material in the Algol system.
