dihydropyrimidine dehydrogenase polymorphisms in patients with gastrointestinal malignancies and their impact on fluoropyrimidine tolerability: Experience from a single Italian institution

BACKGROUNd Fluoropyrimidines are metabolized in the liver by the enzyme dihydropyrimidine dehydrogenase(dPd),encoded by the dPYd *** 7%of the European population is a carrier of dPYd gene polymorphisms associated with reduced dPd enzyme *** To assess the prevalence of dPYd polymorphisms and their impact on fluoropyrimidine tolerability in Italian patients with gastrointestinal *** A total of 300 consecutive patients with a diagnosis of gastrointestinal malignancy and treated with a fluoropyrimidine-based regimen were included in the analysis and divided into two cohorts:(1)149 patients who started fluoropyrimidines after dPYd testing;and(2)151 patients treated without dPYd *** the patients in cohort A,15%tested only the dPYd2A polymorphism,19%tested four polymorphisms(dPYd2A,HapB3,c.2846A>T,and dPYd13),and 66%tested five polymorphisms including *** Overall,14.8%of patients were found to be carriers of a dPYd variant,the most common being dPYd6(12.1%).Patients in cohort A reported≥G3 toxicities(P=0.00098),particularly fewer nonhematological toxicities(P=0.0028)compared with cohort B,whereas there was no statistically significant difference between the two cohorts in hematological toxicities(P=0.6944).Significantly fewer chemotherapy dose reductions(P=0.00002)were observed in cohort A compared to cohort B,whereas there was no statistically significant differences in chemotherapy *** Although this study had a limited sample size,it provides additional information on the prevalence of dPYd polymorphisms in the Italian population and highlights the role of pharmacogenetic testing to prevent severe toxicity.

Quantum cellular automata and free quantum field theory

In a series of recent papers [1-4] it has been shown how free quantum field theory can be derived without using mechanical primitives （including space-time, special relativity, quantization rules, etc.）, but only considering the easiest quantum algorithm encompassing a countable set of quantum systems whose network of interactions satisfies the simple principles of unitarity, homogeneity, locality, and isotropy. This has opened the route to extending the axiomatic information-theoretic derivation of the quantum theory of abstract systems [5, 6] to include quantum field theory. The inherent discrete nature of the informational axiomatization leads to an extension of quantum field theory to a quantum cellular automata theory, where the usual field theory is recovered in a regime where the discrete structure of the automata cannot be probed. A simple heuristic argument sets the scale of discreteness to the Planck scale, and the customary physical regime where discreteness is not visible is the relativistic one of small waveveetors. In this paper we provide a thorough derivation from principles that in the most general case the graph of the quantum cellular automaton is the Cayley graph of a finitely presented group, and showing how for the case corresponding to Euclidean emergent space （where the group resorts to an Abelian one） the automata leads to Weyl, dirac and Maxwell field dynamics in the relativistic limit. We conclude with some perspectives towards the more general scenario of non-linear automata for interacting quantum field theory.

Variable retention harvesting in Great Lakes mixed-pine forests: emulating a natural model in managed ecosystems

Variable retention harvesting(VRH)systems have gained wide use in many different forest types across the globe,but largely have been implemented in forests characterized by severe,infrequent disturbance *** has been less attention given to developing VRH approaches in forests that are characterized as having a mixed-severity disturbance regime that often results in only partial mortality of canopy trees in spatially heterogeneous *** example of such a forest type is red pine(Pinus resinosa Ait.)-dominated ecosystem of the western Great Lakes region of North *** purpose of this review is to provide a conceptual foundation for developing VRH approaches in red pine ecosystems that are based on a mixed-severity disturbance *** contention is that red pine forests managed following a natural model are more resilient to disturbances and external threats such as climate *** the red pine ecosystem,VRH application should reflect the often severe,but partial canopy removal from natural disturbance that is characteristic of this ecosystem and that results in more than trivial numbers of surviving overstory trees across a range of spatial configurations in regenerating *** live trees should span a range of diameters,but favor the larger end of the diameter distribution,as this reflects the likely pattern of survival after natural disturbance and is often a key structural element lacking from managed *** should be applied in ways that vary the spatial pattern of legacy trees in and among stands,but largely in ways that reflect the pattern of spatially patchy canopy structure,with large openings surrounded by a less disturbed matrix,as occurs with a natural disturbance *** trees and deadwood structures should reflect the composition of the pre-disturbance forest,including species in addition to dominant red ***,retained structures should be viewed as dynamic entities that grow,die,and decay and that need

Simplified approach to integrate seismic retrofitting prioritization with social cost evaluation: A case study in central Italy

In the last three decades, bridge stock seismic retrofitting prioritization has become one of the cult topics for scientific discussions in the bridge management strategies. More recent methods are focusing on the evaluation of the generalized failure cost, of a specific bridge derived from direct and indirect costs induced to the users/residents of the area exposed to the seismic hazard as a consequence of bridge collapse. However, when these approaches have to be applied to large transport networks, appear still very complex and computational demanding, and therefore simplified methods to evaluate the impact in terms of social cost related to the reduced efficiency of a transportation network due to potential bridge failure, are *** this work, a simplified method for seismic retrofitting prioritization on a bridge stock is proposed, which is based on a “blended” approach considering specific fragility curves according to several bridge features and condition state, seismic inputs and generalized failure costs related to the transportation network. The effectiveness of the method has been showed on a case study of a local bridge stock placed in central Italy and the obtained results have been compared with those provided by more refined transport simulation models, on one hand, and by more traditional prioritization approaches, on the other. It is highlighted that this method can be very useful for transportation network managers with in a limited budget scenario, in case of lack of information about possible earthquakeinduced impacts on a transportation network efficiency.

Although many causes of Alzheimer’s disease(Ad)may exist,both the original amyloid cascade and tau hypotheses posit that abnormal misfolding and accumulation of amyloid-β(Aβ)and tau protein is the central event causing the ***,that conclusion could be only partly true,and there is conflicting evidence about the role of both proteins in Ad,being able to precede and influence one *** researchers argue that these proteins are mere executors rather than primary causes of ***,there have been continuing refinements of both hypotheses,with alternative explanations proposed.Aβand tau proteins may be independently involved in specific neurotoxic pathways;yet there may be other crucial processes going on in early ***,accumulating evidence suggests that Aβand tau act synergistically,rather than additively in disease onset(Jeremic et al.,2021,2023a).

Effects of Plasmodium falciparum—infected erythrocytes on matrix metalloproteinase-9 regulation in human microvascular endothelial cells

Objective:To investigate the regulation of matrix metalloproteinases(MMPs) and tissue inhibitors of melalloproleinases(TIMPs) in human microvascular endothelium(HMEC-1) exposed to erythrocytes infected by different strains of Plasmodium falciparum(***).Methods: HMEC—1 eells were co—incubated for 72 h with erythrocytes infected by late stage trophozoite of d10(chloroquine-sensilive) or W 2(chloroquine-resistant) *** *** supernatants were then collected and the levels of pro- or active gelatinases MMP-9 and MMP-2 were evaluated by gelatin zymograpln and *** release of pro-MMP-9,***-1 and TIMP-1 proteins was analyzed by western blotting and ***:Infected erythrocytes induced de novo proMMP-9 and MMP-9 *** basal levels of proMMP-2 were altered,nor active MMP-2 was ***-3 and MMP-1 secretion was significant!) enhanced,whereas basal TIMP-1 was *** effects were similar for both strains. Precise Agriculture:Effective deep Learning Strategies to detect Pest Insects

Pest insect monitoring and control is crucial to ensure a safe and profitable crop growth in all plantation types,as well as guarantee food quality and limited use of *** aim at extending traditional monitoring by means of traps,by involving the general public in reporting the presence of insects by using *** includes the largely unexplored problem of detecting insects in images that are taken in noncontrolled ***,pest insects are,in many cases,extremely similar to other species that are ***,computer vision algorithms must not be fooled by these similar insects,not to raise unmotivated *** this work,we study the capabilities of state-of-the-art(SoA)object detection models based on convolutional neural networks(CNN)for the task of detecting beetle-like pest insects on nonhomogeneous images taken outdoors by different ***,we focus on disambiguating a pest insect from similar harmless *** consider not only detection performance of different models,but also required computational *** study aims at providing a baseline model for this kind of *** results show the suitability of current SoA models for this application,highlighting how FasterRCNN with a MobileNetV3 backbone is a particularly good starting point for accuracy and inference execution *** combination provided a mean average precision score of 92.66%that can be considered qualitatively at least as good as the score obtained by other authors that adopted more specific models.
